The study examines correlation between the degree of stenosis of the internal carotid artery (ICA) and the night sleep parameters. The possible neurophysiological mechanisms of sleep disorder in blood flow disturbances in the carotid system are also discussed. Twenty-four patients (19 men and 5 women) were examined, including 6 cases of 50% ICA stenosis, 7 cases of 50–70% ICA stenosis; and 11 cases of ICA occlusion. A polygraphic study of night sleep was conducted and the sleep stages were analyzed. The patients filled in the quality of sleep questionnaire with the presence of the somnolence scoring system. In order to assess brain perfusion, single-photon emission CT imaging of the brain was performed. The results of the study showed that the night sleep structure in the cases of 50% ICA stenosis was unchanged: all the sleep phases and stages whose quantitative parameters corresponded to the reference normal data were recorded, or only stage II sleep representation declined; in the cases of 50–70% ICA stenosis, predominantly stage II sleep and slow-wave sleep were compromised; in patients with ICA occlusion, slow-wave sleep and REM-sleep (45% of cases) were disturbed.  相似文献

Holcoperla magna n.sp. is described and figured from material collected on Headlong Peak, Mt Aspiring National Park. New information is given on H. angularis (Wisely) and H. jacksoni McLellan, and the 3 species are compared. It is hypothesized that the elevation of the temperature of the blood during heat stress may cause an increase of the shedding of erythrocyte membrane vesicles. Therefore, the increase of vesicle numbers following heat stress may be indicative of and proportional to the level heat stress. In order to test this hypothesis, erythrocytes and the vesicles shed by erythrocytes were collected from rat blood and analyzed after the elevation of body temperature by exposure to external heat. The images of erythrocytes and vesicles were analyzed by a custom light microscopy system with spatial resolution of better than 90 nm. The samples were observed in an aqueous environment and required no freezing, dehydration, staining, shadowing, marking or any other manipulation. The elevation of temperature from 36.7±0.3 to 40.3±0.4 °C resulted in significant increase of the concentration of vesicles in blood. At a temperature of 37 °C, mean vesicle concentrations and diameters found in rat blood were (1.4±0.2)×106 vesicles/μL and 0.436±0.03 μm, respectively. The concentration of free vesicles increased after exposure to heat to (3.8±0.3)×106 vesicles/μL. It was estimated that 80% of all vesicles found in rat blood are smaller than 0.45 μm. The increase in the number of vesicle associated with elevated temperatures may be indicative of the heat stress level and serve as diagnostic test of erythrocyte stability and heat resistance.  相似文献

Xanthomonas citri subsp. malvacearum (Xcm) causes severe qualitative and quantitative losses to farmers in cotton-growing areas of the world. Isolates of Xcm were extracted from cotton seeds obtained from five ginneries located in Funtua, Malumfashi, Gusau and Zaria and standardised to 10?5?cfu/ml. One isolate per location was used to inoculate three sets of 10 cotton differential lines known to differentiate races of Xcm through possession of B-genes for resistance to bacterial blight. Each cotton differential line was inoculated with the isolates at the six-leaf stage and SDW was inoculated as control. One hundred and sixty pots used were arranged in completely randomised design on the screen house bench. Four different pathogenic races were identified namely race 1, race 12, race 13 and race 16. This confirms the existence of an evolution of the species across northern Nigeria.  相似文献

Photoreceptor composition and retinal visual pigments in three newt (Caudata, Salamandridae, Pleurodelinae) species (Pleurodeles waltl, Lissotriton (Triturus) vulgaris, and Cynops orientalis) were studied by light microscopy and single-cell microspectrophotometry. Retinas of all three species contain “red” (rhodopsin/porphyropsin) rods, large and small single cones, and double cones. Large single cones and both components of double cones contain red-sensitive (presumably LWS) visual pigment whose absorption spectrum peaks between 593 and 611 nm. Small single cones are either blue- (SWS2, maximum absorption between 470 and 489 nm) or UV-sensitive (SWS1, maximum absorption between 340 and 359 nm). Chromophore composition of visual pigments (A1 vs. A2) was assessed both from template fitting of absorption spectra and by the method of selective bleaching. All pigments contained a mixture of A1 (11-cis retinal) and A2 (11-cis-3,4-dehydroretinal) chromophore in the proportion depending on the species and cell type. In all cases, A2 was dominant. However, in C. orientalis rods the fraction of A1 could reach 45%, while in P. waltl and L. vulgaris cones it did not exceed 5%. Remarkably, the absorption of the newt blue-sensitive visual pigment was shifted by up to 45 nm toward the longer wavelength, as compared with all other amphibian SWS2-pigments. We found no “green” rods typical of retinas of Anura and some Caudata (ambystomas) in the three newt species studied.  相似文献

Using high performance liquid chromatography, UV-VIS spectra and mass-spectra (FAB MS), 7 carotenoid species were identified in tissues of the bivalve mollusc Anadara inaequivalvis (Bruguiere, 1789): trans- and cis-pectenolon, alloxanthin, pectenol A, β-carotene, zeaxanthin, and diatoxanthin. Their quantitative ratio in hepatopancreas, gills and foot of animals were determined. A negative correlation (R 2 is about 0.9) was revealed between tissues content of a series of carotenoids (trans- and cis-pectenolon, zeaxanthin, alloxanthin, and diatoxanthin) and activity of antioxidant enzymes (catalase and superoxide dismutase). The existence of competitive interrelations between these molecular systems is proposed and underlying causes are discussed.  相似文献

Foot-and-mouth disease virus (FMDV), the causative agent of foot-and-mouth disease, is an Aphthovirus within the Picornaviridae family. During infection with FMDV, several host cell membrane rearrangements occur to form sites of viral replication. FMDV protein 2C is part of the replication complex and thought to have multiple roles during virus replication. To better understand the role of 2C in the process of virus replication, we have been using a yeast two-hybrid approach to identify host proteins that interact with 2C. We recently reported that cellular Beclin1 is a natural ligand of 2C and that it is involved in the autophagy pathway, which was shown to be important for FMDV replication. Here, we report that cellular vimentin is also a specific host binding partner for 2C. The 2C-vimentin interaction was further confirmed by coimmunoprecipitation and immunofluorescence staining to occur in FMDV-infected cells. It was shown that upon infection a vimentin structure forms around 2C and that this structure is later resolved or disappears. Interestingly, overexpression of vimentin had no effect on virus replication; however, overexpression of a truncated dominant-negative form of vimentin resulted in a significant decrease in viral yield. Acrylamide, which causes disruption of vimentin filaments, also inhibited viral yield. Alanine scanning mutagenesis was used to map the specific amino acid residues in 2C critical for vimentin binding. Using reverse genetics, we identified 2C residues that are necessary for virus growth, suggesting that the interaction between FMDV 2C and cellular vimentin is essential for virus replication.  相似文献

This study aimed to evaluate the participation of actin and tubulin in the process of internalisation, the interaction of bacterial phagosomes with lysosomes, the morphometric changes and the expression of inflammatory cytokines in Caco-2 cells infected with Campylobacter jejuni. Both actin and tubulin participated in the process of internalisation. Inside the cells, lysosomes fuse with phagosomes, which may lead to bacterial death because after 2 h, the bacteria were not detected by Transmission electron microscopy (TEM). There is increased expression of TGF-β3 during the early stages, and IL-8 was expressed after 60 min p.i. This work showed that C. jejuni invades and causes major morphometric changes in epithelial cells. In response, the cells increase their expression of cytokines that can lead to inflammation. The mechanisms of invasion are dependent on actin and tubulin, and once internalised, lysosomes fuse with phagosomes.  相似文献
