Rectangular game theory and metabolic response to random changes in environmental states

Rectangular game theory is applied to the response of organisms to random environmental changes: A hypothetical example of the response of a facultative algae to random changes in illumination is analyzed. Two strategies are discussed: Bayes' risk and maximin. It is shown how to detect such strategies in populations. The analysis does not require assumptions about the form of the relationship between metabolic activities and selective advantage, but assumptions about evolutionary optimization are required. The maximin strategy is shown to be related to metabolic homeostasis.     

Demographic genetic approach in anthropologic research. II. A method for calculating the coefficient of consanguinity in a complex nongraphic pedigree

A simplified method of manual calculation for inbreeding coefficient (and/or relationship) from complicated pedigree tables presented in a nongraphic mode is offered. The method is based on the use of positional dual codes of ancestors in which all genealogical links of the proband are reflected.     

Merkel cell tumor: diagnosis, prognosis, and management

The histologic features of the Merkel cell and the pathologic characteristics of the Merkel cell tumor are described. From a literature review, 68 cases are examined; 23 cases that have been followed up accurately are presented from the Mayo Clinic. A representative case is used to demonstrate the problems of diagnosis and suggested treatment. The latter should be wide local resection with removal of nodal metastases if present. When indicated, this should be followed up with radiation therapy. Chemotherapy is of little or no value. Survival rates are presented.     

Isolation, macromolecular properties, and combining site of a chito-oligosaccharide-specific lectin from the exudate of ridge gourd (Luffa acutangula)

A lectin specific for chito-oligosaccharides from the exudate of ridge gourd (Luffa acutangula) fruits has been purified to homogeneity by affinity chromatography. The lectin has a molecular weight of 48,000, an S(0)20,w of 4.06 S and a Stokes radius of 2.9 nm. Upon sodium dodecyl sulfate-polyacrylamide gel electrophoresis, a single band corresponding to Mr of 24,000 was observed both in the presence and absence of beta-mercaptoethanol. The subunits in this dimeric lectin are, therefore, held together solely by noncovalent interactions. The lectin is not a glycoprotein, and secondary structure analysis by CD measurements showed 31% alpha-helix. The hemagglutinating activity of L. acutangula agglutinin was not inhibited by any of the monosaccharides tested. Among the disaccharides only di-N-acetylchitobiose was inhibitory. The inhibitory potency of chito-oligosaccharides increased dramatically with their size up to penta-N-acetylchitopentaose. The lectin has two binding sites for saccharides. The affinity of chito-oligosaccharides for L. acutangula lectin, as monitored by titrating the changes in the near UV-CD spectra and intrinsic fluorescence, increased strikingly with the number of GlcNAc units in them. The values of delta G, delta H, and delta S for the binding process showed a pronounced dependence on the size of the chito-oligosaccharides, indicating that the binding of higher oligomers is progressively more favored thermodynamically than di-N-acetylchitobiose. The thermodynamic data are consistent with an extended binding site in this lectin, which accommodates a tetrasaccharide.     

Immunohistochemical and electron microscopic identification of serotonin, melatonin and beta-endorphin in the granules of natural killers

The ability of serotonin, melatonin and beta-endorphin synthesis by secreting granules of natural killer cells (NK cells) was established by immunohistochemical method. It was shown that cytotoxic effect of NK cells can be to a certain extent related to the presence of secreting granules. A possible direct contribution of NK cell-synthesized hormones to the natural killer antitumour effect is discussed.     

An inquiry into the source of stereospecificity of lactate dehydrogenase using substrate analogues and molecular modeling.

Lactate dehydrogenase catalyzes the stereospecific hydride transfer to and from the re face of the nicotinamide coenzyme. The demonstrated probability of transfer to the si face of less than 2 x 10(-8) indicates that the free energy of any diastereotopic transition state leading to a si transfer must be over 10 kcal/mol greater than the free energy for transfer to or from the re face. The general notion of closed, desolvated active sites suggests the a priori hypothesis that steric hindrance prevents the nicotinamide ring from assuming a conformation that would lead to transfer of the pro-S hydrogen. In this paper we report that the probability of transfer of the pro-S proton is less than 9 x 10(-7) with 3-pyridinealdehyde adenine dinucleotide as coenzyme and less than 4 x 10(-7) during the lactate dehydrogenase catalyzed disproportionation of glyoxylate. Examination of the crystal structure of lactate dehydrogenase further suggests that steric exclusion does not enforce the extreme stereospecificity of the reaction. An electrostatic interaction with the macrodipole associated with the alpha 2F helix is suggested as a potential molecular source of the stereospecificity.     

Purification and immunological studies of glutathione reductase from rat liver. Evidence for an antigenic determinant at the nucleotide-binding domain of the enzyme

Glutathione reductase has been purified to homogeneity by a method which is an improvement of an earlier procedure (Carlberg, I. and Mannervik, B. (1975) J. Biol. Chem. 250, 5475-5480). The new steps in the purification scheme include affinity chromatography on 2',5' ADP-Sepharose 4B. Antibodies to glutathione reductase from rat liver were raised in rabbits and used for analysis of the enzyme by quantitative 'rocket' immunoelectrophoresis. Glutathione reductase from human erythrocytes, porcine erythrocytes, and calf-liver gave precipitin lines showing partial identity with the rat liver enzyme in Ouchterlony double diffusion experiments. Enzyme from spinach, yeast (Saccharomyces cerevisiae), and the photosynthetic bacterium Rhodospirillum rubrum did not give precipitates with the antibodies to the enzyme from rat liver. Titration of glutathione reductase from the different sources with antibodies confirmed the cross-reactivity of the mammalian enzymes; the human enzyme giving the strongest heterologous reaction. No reaction was observed with the enzyme from spinach, yeast, and Rhodospirillum rubrum. NADPH, NADP+, and 2',5' ADP were found to inhibit the interaction between antibodies and glutathione reductase from rat liver and human erythrocytes. NADH, glutathione, or glutathione disulfide did not protect the enzyme from reacting with the antibodies. It is concluded that glutathione reductase has an antigenic binding site for the antibodies at the pyridine nucleotide-binding site of the enzyme molecule.     

The portal vein and its main affluents. An anatomic and morphometric study

There are reported the results of certain measurements made on 158 human cadavers of both sexes. A series of main parameters are given, such as the distance between the superior mesenteric vein and on the one hand its confluence with other vessels and on the other hand the pancreas and the horizontal part of the duodenum: the distance between the superior mesenteric vein together with the portal vein, and the inferior caval vein; the diameters of the portal trunk and the confluence angle between the roots of the portal vein. The results reflect the possibility to perform a troncular portocaval anastomosis in at least 90 per cent of the cases. The authors' attention was, however, directed towards the anatomy of the superior mesenteric vein, as this vessel is preferred by some surgeons in the achievement of the portocaval anastomosis.
