Rapid and reproducible method is described here for the purification of the 43 kDa fimbrial protein from P. gingivalis by preferential fractionation in the presence of 1% SDS and 0.2M of a bivalent cation at pH 6.5. Homogeneity of the purified 43 kDa was confirmed by SDS-PAGE and Western blot analysis using monoclonal and polyclonal antibodies raised against this protein. Amino acid composition and the amino acid sequence of the first 30 amino acid residues of the purified fimbriae are consistent with the composition and sequence predicted from the cloned gene of the fimbrial subunit. Circular dichroism spectra shows high levels of beta-sheet structure. The purified 43 kDa polymer shows fimbriae-like morphology under the electron microscope. Ultrastructural localization of the 43 kDa protein by the immunogold technique revealed specific labeling of the fimbriae with a diameter of approximately 3.5 to 5.0 nm. Localization of this protein suggest that the 43 kDa component is a fimbrial subunit.  相似文献

Avian oviductal fluids contain phosphorylating-dephosphorylating enzymes that might function in sperm-oocyte interactions. Phosphorprotein phosphatase and protein kinase have been purified 20-fold and 40-fold, respectively. The latter easily aggregates and is highly labile. A cDNA clone encoding the N-terminal sequence of the murine integrin beta 7 subunit, a novel member of the leukocyte cell adhesion molecule subset (Leu-CAM), has been isolated. An N-terminal region of 13 contiguous amino acids deduced from the cDNA shows complete identity with the N-terminus of the 120 kDa subunit of the M290 antigen, a surface molecule found highly expressed on mouse intestinal intraepithelial lymphocytes (IEL). This unexpected result focuses two previously unconnected areas of research and suggests that integrins may have a special role to play in the defence of the gut mucosa.  相似文献

Abstract— When day-old chicks were injected intraperitoneally with 1.62mmoles of l -phenylalanine, they developed a condition resembling narcosis. Simultaneously, whole brain levels of phenylalanine were 2–4 μmol/g, whereas those in control brain were 0.06 μmol/g. Examination of some glycolytic intermediates in the brain revealed significant decreases in fructose-1,6-diphosphate, l -α-glycerol phosphate and lactate, in comparison to the levels of these compounds in the saline-injected control animals. Levels of glucose and glucose-6-phosphate either increased or did not change, whereas levels of glycogen did not differ significantly. Phosphocreatine increased reciprocally with the decrease in inorganic phosphate. The levels of adenine nucleotide (energy charge) were not affected. Utilization of cerebral high-energy phosphates was depressed by 50–70 per cent when determined as a function of metabolic rate in the brain at 15- and 30-s periods of ischaemia according to the ‘closed-system’ technique. Explanations for these data have been examined, such as toxicity of phenylacetate and inhibition of glycolytic enzymes by phenylpyruvate and l -phenylalanine and their relevance to this study is discussed.  相似文献

Abstract— The folate coenzymes of mouse brain underwent quantitative and qualitative changes during the first few weeks after birth. The total folate coenzymes per unit wet weight declined by approximately 60 per cent. In the newborn brains a relatively small proportion of the total folates were poly-γ-glutamyl derivatives, but the percentage of the total folates in these forms increased as the brain matured.  相似文献
