A high throughput screen showed the ability of a 1-amino-2,4-dicyanopyrido[1,2-a]benzimidazole analogue to directly inhibit the lytic activity of the pore-forming protein perforin. A series of analogues were prepared to study structure-activity relationships (SAR) for the this activity, either directly added to cells or released in situ by KHYG-1 NK cells, at non-toxic concentrations. These studies showed that the pyridobenzimidazole moiety was required for effective activity, with strongly basic centres disfavoured. This class of compounds was relatively unaffected by the addition of serum, which was not the case for a previous class of direct inhibitors.  相似文献

The structure of an extracellular polysaccharide (EPS) from Streptococcus thermophilus THS has been determined. A combination of component analysis, methylation analysis and NMR spectroscopy shows that the polysaccharide is composed of pentasaccharide repeating units. Sequential information was obtained by two-dimensional (1)H,(1)H-NOESY and (1)H,(13)C-HMBC NMR experiments. NMR data indicate different mobility within the EPS with a stiffer backbone and a more flexible side-chain.  相似文献

Brachytheciaceae is often considered a taxonomically difficult group of mosses. For example, morphological variation has led to difficulty in generic delimitation. We used DNA sequence data (chloroplast psbT‐H and trnL‐F and nuclear ITS2) together with morphology (63 characters) to examine the relationships within this family. The combined unaligned length of the DNA sequences used in the phylogenetic analyses varied between 1277 and 1343 bp. For phylogeny reconstruction we performed direct optimization, as implemented in POY. Analyses were performed with three different gap costs and the morphological data partition was weighted both: (1) equal to gap cost, and (2) with a weight of one. The utility of sensitivity analysis has recently been cast into doubt; hence in this study it was performed only to explore the effects of weighting on homology statements and topologies and to enable more detailed comparisons between earlier studies utilizing the direct optimization method. The wide sequence length variation of non‐coding ITS2 sequences resulted in character optimizations (i.e., “alignments”) of very different lengths when various gap costs were applied. Despite this variation, the topologies of equally parsimonious trees remained fairly stable. The inclusion of several outgroups, instead of only one, was observed to increase the congruence between data sets and to slightly increase the resolution. An inversion event in the 9 bp loop region in the chloroplast psbT‐N spacer in mosses has been postulated to include only uninformative variation, thus possibly negatively impacting the phylogeny reconstruction. Despite this inversion, its variation within Brachytheciaceae was clearly congruent with information from other sources, but inclusion of these 9 bp in the analysis had only a minor effect on the phylogenetic results. In the most parsimonious topology, which was obtained with equal weighting of all data, Meteoriaceae and Brachytheciaceae were resolved as monophyletic sister groups, which had recently been suggested based on a few shared morphological characters. Our study revealed some new generic relationships within the Brachytheciaceae, which are discussed in light of the morphological characters traditionally used for generic delimitation.  相似文献

Competing hypotheses that rely either on a stepping-stone dispersal via the North Atlantic or the Bering land bridges, or more recent transoceanic dispersal, have been proposed to explain the disjunct distribution of Mediterranean flora in southern Europe and western North America. These hypotheses were tested with molecular dating using a phylogeny of the moss genus Homalothecium based on ITS, atpB-rbcL, and rpl16 sequence data. The monophyly of two main lineages in Western Palearctic (Europe, central Asia and north Africa) and North America is consistent with the ancient vicariance hypothesis. The monophyly of Madeiran H. sericeum accessions supports the recognition of the Macaronesian endemic H. mandonii. A range of absolute rates of molecular evolution documented in land plants was used as probabilistic calibration prior by a Bayesian inference implementing a relaxed-clock model to derive ages for the nodes of interest. Our age estimates for the divergence of the American and Western Palearctic Homalothecium clade (5.7 Ma, IC 3.52-8.26) and the origin of H. mandonii (2.52 Myr IC 0.86-8.25) are not compatible with the ancient vicariance hypothesis. Age estimates suggests that species distributions result from rare instances of dispersal and subsequent sympatric diversification. The calibrated phylogeny indicates that Homalothecium has undergone a fast radiation during the last 4 Myr, which is consistent with the low levels of morphological divergence among sibling species.  相似文献

We report the isolation and cross species amplification of 42 Drosophila virilis microsatellite loci. Nine loci were isolated from mapped P1 bacteriophage clones and 33 were obtained from genomic DNA or GenBank searches. Cross species amplification was tested for all members of the D. virilis group. The amplification success was high (varying from 45% to 100%) and most of the loci were polymorphic. This set of loci can be applied for several genetic studies such as mapping behavioural quantitative trait loci (QTL) and for studying population structure in a phylogeographical framework in D. virilis group species.  相似文献

Methane (CH4) and nitrous oxide (N2O) dynamics were studied in a boreal Sphagnum fuscum pine bog receiving annually (from 1991 to 1996) 30 or 100 kg NH4NO3-N ha–1. The gas emissions were measured during the last three growing seasons of the experiment. Nitrogen treatment did not affect the CH4 fluxes in the microsites where S. fuscum and S. angustifolium dominated. However, addition of 100 kg NH4NO3-N ha–1 yr–1 increased the CH4 emission from those microsites dominated by S. fuscum. This increase was associated with the increase in coverage of cotton grass (Eriophorum vaginatum) induced by the nitrogen treatment. The differences in the CH4 emissions were not related to the CH4 oxidation and production potentials in the peat profiles. The N2O fluxes were negligible from all microsites. Only minor short-term increases occurred after the nitrogen addition.  相似文献

The metabolite profiles of 20 Stachybotrys spp.isolates from Finnish water-damaged buildings were compared with their biological activities. Effects of purified compounds on cytotoxicity and production of inflammatory mediators such as nitric oxide, IL-6 and TNFα in murine RAW264.7 macrophage cells were studied. The 11isolates belonging to the satratoxin-producing chemotype were highly cytotoxic to the macrophages. The isolates inducing inflammatory mediators all belonged to the atranone-producing chemotype, but pure atranones B, and D did not elicit a response in the bioassay. Altogether, cytotoxicity ofStachybotrys sp. isolates appear to be related to satratoxin production whereas the specific component inducing inflammatory responses in atranone-producing isolates remains obscure. The effects of 4-weeks ethanol application (20% ethanol, w/w, 2 g X kg-1 on the alcohol oxidizing systems and gluconeogenic enzyme activities of the liver in guinea pigs kept in the cold (+4 degrees C) and at room temperature (+20 degrees C) were studied. The controls were guinea pigs reared at room temperature or in a cold environment without ethanol. The study showed a significant increase (1.5-fold) in liver microsomal cytochrome P-450 after chronic ethanol treatment at room temperature, but not in a cold environment. Microsomal NADPH oxidase activity did not significantly change in any group. Ethanol treatment in a cold environment resulted in a significant increase in liver mitochondrial cytochromes, aa3 and c+c1, and at room temperature in cyt aa3. The activities of total liver homogenate alcohol dehydrogenase or catalase did not change after chronic ethanol treatment. The activity of liver fructose-1.6-diphosphatase showed a significant ethanol induced decrease at room temperature, an effect not observed in the cold environment. Ethanol increased glucose-6-phosphatase activity in the cold, but not at room temperature. In conclusion, the stimulation of liver mitochondrial cytochromes and microsomal cyt P-450 as a consequence of chronic ethanol treatment indicated an increased oxidation capacity for ethanol. The stimulation of glucose-6-phosphatase in a cold environment might be responsible for increasing glucose for heat production after chronic ethanol treatment in cold adapted animals.  相似文献
