土壤重金属与农产品质量息息相关.本研究通过设置砷胁迫条件下水稻种植盆栽试验,分析络合型含铁材料不同施用时期和不同施用量对水稻SPAD值、抗氧化酶(SOD、POD、CAT)活性以及水稻砷吸收、累积的影响.结果表明,砷胁迫对水稻SOD、POD、CAT活性有着不同程度的抑制作用,添加络合型含铁材料可以降低水稻MDA含量,缓解砷对水稻光合作用和SOD、POD、CAT活性的抑制;同时能减少水稻对土壤砷的吸收和累积.与移栽前添加络合型含铁材料相比,选择水稻孕穗期前添加络合型含铁材料,水稻砷吸收的抑制效果较好;水稻植株地上部砷含量与络合型含铁材料的添加浓度呈显著负相关,孕穗期前添加0.2 g·kg-1络合型含铁材料对水稻植株砷的吸收抑制效果最好,水稻植株地上部砷含量降低59.22%.  相似文献

Though initially identified as necessary for neural migration, Disconnected and its partially redundant paralog, Disco-related, are required for proper head segment identity during Drosophila embryogenesis. Here, we present evidence that these genes are also required for proper ventral appendage development during development of the adult fly, where they specify medial to distal appendage development. Cells lacking the disco genes cannot contribute to the medial and distal portions of ventral appendages. Further, ectopic disco transforms dorsal appendages toward ventral fates; in wing discs, the medial and distal leg development pathways are activated. Interestingly, this appendage role is conserved in the red flour beetle, Tribolium (where legs develop during embryogenesis), yet in the beetle we found no evidence for a head segmentation role. The lack of an embryonic head specification role in Tribolium could be interpreted as a loss of the head segmentation function in Tribolium or gain of this function during evolution of flies. However, we suggest an alternative explanation. We propose that the disco genes always function as appendage factors, but their appendage nature is masked during Drosophila embryogenesis due to the reduction of limb fields in the maggot style Drosophila larva.  相似文献

以抗病和感病泡核桃无性系为实验材料,人工接种褐斑病病原菌后测定不同时期叶片中保护酶活性、总酚、类黄酮、叶绿素含量等相关生理生化指标,探讨不同抗性泡核桃响应褐斑病病原菌侵染的生理生化差异。结果表明:(1)接种病原菌后,感病无性系64叶片带菌率随着侵染时间的增加而升高,且显著高于抗病无性系199(P<0.05)。(2)抗病无性系199和感病无性系64叶片的SOD、POD、CAT、APX和PPO活性随着侵染时间均呈现先升高后降低的变化趋势,其中SOD、POD和APX活性均在16 d时达到最大值;与较感病无性系相比,接种后抗病无性系的POD和APX活性较强;在接种前期(1~16 d),感病无性系PPO活性高于抗病无性系,后期(16~34 d)CAT活性也较抗病无性系高。(3)抗病无性系叶片叶绿素含量始终高于感病无性系;抗病无性系MDA含量在接种后无明显变化,而感病无性系先增加后降低,其细胞膜脂过氧化较重。(4)两个无性系叶片可溶性蛋白和可溶性糖含量变化较平缓,且差异不显著,在接种后期(34 d)有升高的趋势;接种5 d以后,感病无性系叶片类黄酮和总酚含量始终显著高于抗病无性系。研究发现,泡核桃抗病无性系叶片带菌率较低,较难受到侵染,并且通过提高POD和APX活性以及积累较多叶绿素、可溶性蛋白和可溶性糖来应对病原菌侵染引起的氧化胁迫,抑制病原菌的繁殖,从而提高其抗病能力。  相似文献

A defining character of adaptive radiations is the evolution of a diversity of morphological forms that are associated with the use of different habitats, following the invasion of vacant niches. Island adaptive radiations have been thoroughly investigated but continental scale radiations are more poorly understood. Here, we use 52 species of Australian agamid lizards and their Asian relatives as a model group, and employ three‐dimensional geometric morphometrics to characterize cranial morphology and investigate whether variation in cranial shape reflects patterns expected from the ecological process of adaptive radiation. Phylogenetic affinity, evolutionary allometry, and ecological life habit all play major roles in the evolution of cranial shape in the sampled lizards. We find a significant association between cranial shapes and life habit. Our results are in line with the expectations of an adaptive radiation, and this is the first time detailed geometric morphometric analyses have been used to understand the selective forces that drove an adaptive radiation at a continental scale.  相似文献

Ontogenic development is divided into infant, juvenile, adolescent and adult life‐stages. Although the developmental trajectory of an individual is a flexible entity, which differs within species, environment and sex, life‐stage classifications are generally structured, age‐based systems. This invariably leads to rigidity within a dynamic system and consequently hampers our understanding of primate life history strategies. We propose that life‐stage classifications should be quantitative, flexible entities, which use a reliable measurement of development. Here, we provide a methodological example where placement into a life‐stage is based upon behavioral variance between other similar‐aged individuals. Behavioral data were collected from 12 male (3–11 years old) and 9 female (3–8 years old) captive immature western gorillas (Gorilla gorilla gorilla) housed in five family groups, using continuous focal sampling; 900 hr of data were collected over 131 days. Data were applied to four published life‐stage classifications for mountain gorillas (Gorilla beringei beringei), which showed variable ability to determine life‐stage in western gorillas. A new life‐stage classification (Hutchinson & Fletcher) was proposed specifically for western gorillas, whereby multiple co‐varying behavior provided a robust measure of linear development across immaturity. Each life‐stage was found to be a distinct ontogenic phase and the classification discriminated life‐stage with a high level of accuracy. Using the Hutchinson & Fletcher classification we provide evidence for disparity in developmental trajectories between the sexes from the juvenile period onwards. To expand the understanding of primate life histories, we propose that flexible classifications should be used to enable comparison of allometric life history traits within and between species, from birth onwards. Chronic (5 weeks) exposure of freshwater-adapted European flounder, Platichthys flesus (L.), to environmental concentrations of sediment-associated tri-n-butyltin chloride (TBTCl) and triphenyltin chloride (TPhTCl) caused significant changes to hydromineral fluxes and membrane permeability, mechanisms that maintain osmotic homeostasis. The half-time of exchange of tritiated water (THO) in TBTCl- and TPhTCl-exposed fish was significantly increased during the first 2 weeks of the experiment and then decreased steadily, eventually reaching the level that the control group had constantly maintained throughout the experiment. This change in apparent water permeability was accompanied by a significant decrease in diffusional water flux across the membranes. Passive Na(+)-efflux across the gills was increased significantly but effluxes in the control group were near constant over the same time span. Drinking rates in the organotin groups increased significantly while the rate of urine production did not change. This lead to an increased net water balance in the organotin groups and consequently to a significant reduction of the blood osmolality of both organotin groups when compared to a control. There would appear to be a metabolic cost attached to the changes produced by exposure to environmental levels of organotin compounds which are manifested as a minimal increase in body length compared to the controls.  相似文献

It is well known that some protein folds (superfolds) occur very frequently. We show that compared to other folds, most superfold structures have a higher proportion of their alpha-helical or beta-strand residues in one of three basic units of supersecondary structure (alpha-hairpin, beta-hairpin or betaalphabeta-unit). Furthermore, by taking into consideration two more complex motifs, the four-stranded Greek-key (beta4) and the betaalpha-Greek key (betaalphabetabeta), we demonstrate that the remaining superfold structures contain many of these higher order units of three-dimensional packing. The implications of these results for folding are discussed.  相似文献

Mutations in the Streptomyces peucetius dnrD gene block the ring cyclization leading from aklanonic acid methyl ester (AAME) to aklaviketone (AK), an intermediate in the biosynthetic pathway to daunorubicin (DNR) and doxorubicin. To investigate the role of DnrD in this transformation, its gene was overexpressed in Escherichia coli and the DnrD protein was purified to homogeneity and characterized. The enzyme was shown to catalyze the conversion of AAME to AK presumably via an intramolecular aldol condensation mechanism. In contrast to the analogous intramolecular aldol cyclization catalyzed by the TcmI protein from the tetracenomycin (TCM) C pathway in Streptomyces glaucescens, where a tricyclic anthraquinol carboxylic acid is converted to its fully aromatic tetracyclic form, the conversion catalyzed by DnrD occurs after anthraquinone formation and requires activation of a carboxylic acid group by esterification of aklanonic acid, the AAME precursor. Also, the cyclization is not coupled with a subsequent dehydration step that would result in an aromatic ring. As the substrates for the DnrD and TcmI enzymes are among the earliest isolable intermediates of aromatic polyketide biosynthesis, an understanding of the mechanism and active site topology of these proteins will allow one to determine the substrate and mechanistic parameters that are important for aromatic ring formation. In the future, these parameters may be able to be applied to some of the earlier polyketide cyclization processes that currently are difficult to study in vitro.  相似文献

The effects of frequency of follicular aspiration and treatment of donor cattle with FSH on in vivo oocyte recovery and in vitro embryo production were studied. Simmental heifers (n = 24) formed 8 replicates of 3 treatments in which oocyte donors were aspirated 1) once a week, 2) twice a week, or 3) once a week following treatment with FSH for 3 d prior to aspiration. Oocytes were graded, washed, matured for 20 to 24 h and then inseminated with frozen/thawed semen from a single sire, followed by co-culture on granulosa cell layers. Embryo development was observed until Day 7 after insemination. Significantly fewer follicles per heifer per week were counted (14.7+/-2.3 vs. 27.4+/-3.1 vs. 23.1+/-2.8) and aspirated (12.0+/-2.0 vs. 21.8+/-2.7 vs. 20.1+/-2.6) in heifers on the once-weekly than twice-weekly aspiration treatment (P<0.01) or on the once-weekly aspiration after FSH treatment (P<0.05). There were no significant differences between treatments in the total number of oocytes recovered per week (5.6+/-1.2 vs. 8.9+/-1.5 vs. 6.1+/-1.2), but significantly more oocytes per heifer per week recovered from animals treated with FSH were graded Category 1 (2.8+/-0.4), i.e., >4 layers good cumulus with a clear, even cytoplasm, than from animals aspirated once (0.9+/-0.2; P<0.01) or twice a week (1.5+/-0.3; P<0.05). The number of transferable morulae plus blastocysts produced per heifer per week was higher from animals aspirated twice a week (2.4+/-0.4; P<0.05) or once a week following FSH treatment (2.1+/-0.4; P<0.05) than from animals aspirated once a week without FSH treatment (1.0+/-0.3). In conclusion, FSH treatment of bovine oocyte donors aspirated once a week enabled a similar number of transferable embryos to be produced per donor week as aspiration twice a week without FSH treatment. These 2 treatments produced twice as many transferable embryos per donor week as aspiration once a week without FSH treatment.  相似文献

I used a simple mathematical model of the inverse dynamics of locomotion to estimate the minimum muscle masses required to maintain quasi-static equilibrium about the four main limb joints at mid-stance of fast running. Models of 10 extant taxa (a human, a kangaroo, two lizards, an alligator, and five birds) were analyzed in various bipedal poses to examine how anatomy, size, limb orientation, and other model parameters influence running ability. I examined how the muscle masses required for fast running compare to the muscle masses that are actually able to exert moments about the hip, knee, ankle, and toe joints, to see how support ability varies across the limb. I discuss the assumptions and limitations of the models, using sensitivity analysis to see how widely the results differed with feasible parameter input values. Even with a wide range of input values, the models validated the analysis procedure. Animals that are known to run bipedally were calculated as able to preserve quasi-static equilibrium about their hindlimb joints at mid-stance, whereas non-bipedal runners (iguanas and alligators) were recognized as having too little muscle mass to run quickly in bipedal poses. Thus, this modeling approach should be reliable for reconstructing running ability in extinct bipeds such as nonavian dinosaurs. The models also elucidated how key features are important for bipedal running capacity, such as limb orientation, muscle moment arms, muscle fascicle lengths, and body size. None of the animals modeled had extensor muscle masses acting about any one joint that were 7% or more of their body mass, which provides a reasonable limit for how much muscle mass is normally apportioned within a limb to act about a particular joint. The models consistently showed that a key biomechanical limit on running ability is the capacity of ankle extensors to generate sufficiently large joint moments. Additionally, the analysis reveals how large ratite birds remain excellent runners despite their larger size; they have apomorphically large extensor muscles with relatively high effective mechanical advantage. Finally, I reconstructed the evolution of running ability in the clade Reptilia, showing that the ancestors of extant birds likely were quite capable runners, even though they had already reduced key hip extensors such as M. caudofemoralis longus.  相似文献
