The in vitro and in vivo inhibitory effects of 5-(3α, 12α-dihydroxy-5-β-cholanamido)-1,3,4-thiadiazole-2-sulfonamide (1), 5-(3α, 7α, 12α-trihydroxy-5-β-cholanamido)-1,3,4-thiadiazole-2-sulfonamide (2), 5-(3α, 7α, 12α-triacetoxy-5-β-cholanamido)-1,3,4-thiadiazole-2-sulfonamide (3) and acetazolamide on rainbow trout (Oncorhynchus mykiss) (RT) erythrocyte carbonic anhydrase (CA) were investigated. The RT erythrocyte CA was obtained by affinity chromatography with a yield of 20.9%, a specific activity of 422.5?EU/mg protein and a purification of 222.4-fold. The purity of the enzyme was confirmed by SDS-PAGE. Inhibitory effects of the sulfonamides and acetazolamide on the RT erythrocyte CA were determined using the CO2-Hydratase method in vitro and in vivo studies. From in vitro studies, it was found that all the compounds inhibited CA. The obtained I50 value for the sulfonamides (1), (2) and (3) and acetazolamide were 0.83, 0.049, 0.82 and 0.052?μM, respectively. From in vivo studies, it was observed that CA was inhibited by the sulfonamides (1), (2) and (3) and acetazolamide. 相似文献

Soil chemical extractions are widely used to predict the nutritional status of soils. However, the correlation between extracted elements and plant uptake is often poor, especially if compared over a range of soil types. The aim of this study was to examine a new method called Diffusive Gradients in Thin films (DGT), which measures the diffusive supply of elements, thereby mimicking a plant root. The ability of DGT to assess plant-available P, Zn and Cu was tested in a wide range of typical Scandinavian agricultural soils along with conventional methods (EDTA and DTPA for Cu and Zn; NaHCO3 for P and soil solution concentrations). Extracted soil concentrations were compared to that of the element in the youngest fully developed leaf of barley (Hordeum vulgare L.) grown in pots. For Zn and P, only DGT could predict plant uptake while conventional extraction methods and soil solution analyses performed poorly. All soil tests could predict Cu concentration in leaves, but the DGT technique proved to be most accurate followed by the soil solution concentration of Cu. We conclude that DGT is much more accurate at predicting plant-available P, Zn and Cu than commonly used methods for analysing plant-available nutrients in soil. 相似文献

Effects of moderate maternal undernourishment during late gestation on the intermediary metabolism and maturational changes in young lambs were investigated. 20 twin-bearing sheep, bred to two different rams, were randomly allocated the last 6 wk of gestation to either a NORM diet [barley, protein supplement, and silage ad libitum approximately 15 MJ metabolizable energy (ME)/day] or a LOW diet (50% of ME intake in NORM, offered exclusively as silage approximately 7 MJ ME/day). Post partum, ewes were fed to requirement. After weaning, lambs were fed concentrate and hay ad libitum. At 10 and 19 wk of age, lambs were subjected to an intravenous glucose tolerance test (IGTT) followed by 24 h of fasting. Heat energy (HE) was determined in a respiration chamber at 9 or 20 wk of age. LOW lambs had a lower birth weight and continued to be lighter throughout the experiment. Glucose tolerance did not differ between groups. However, 19-wk-old LOW lambs secreted less insulin during IGTT, released more NEFA, and tended to have lower leptin during fasting than NORM. Surprisingly, several metabolite and hormone responses during IGTT and fasting were greatly influenced by the paternal heritage. In conclusion, when lambs entered adolescence (19 wk) programming effects of late prenatal malnutrition on the glucose-insulin homeostasis and metabolism were manifested: LOW lambs had less insulin-secretory capacity, but this was apparently compensated for by increased target tissue sensitivity for insulin, and adipose lipolytic capacity increased during fasting. Thereby, glucose may be spared through increased lipid oxidation, but overall energetic efficiency is apparently deteriorated rather than improved. 相似文献

The third component of complement (C3) is a 190 kDa glycoprotein essential for eliciting the complement response. The protein consists of two polypeptide chains (alpha and beta) held together with a single disulfide bridge. The beta-chain is composed of six MG domains, one of which is shared with the alpha-chain. The disulfide bridge connecting the chains is positioned in the shared MG domain. The alpha-chain consists of the anaphylatoxin domain, three MG domains, a CUB domain, an alpha(6)/alpha(6)-barrel domain and the C-terminal C345c domain. An internal thioester in the alpha-chain of C3 (present in C4 but not in C5) is cleaved during complement activation. This mediates covalent attachment of the activated C3b to immune complexes and invading microorganisms, thereby opsonizing the target. We present the structure of bovine C3 determined at 3 Angstroms resolution. The structure shows that the ester is buried deeply between the thioester domain and the properdin binding domain, in agreement with the human structure. This domain interface is broken upon activation, allowing nucleophile access. The structure of bovine C3 clearly demonstrates that the main chain around the thioester undergoes a helical transition upon activation. This rearrangement is proposed to be the basis for the high level of reactivity of the thioester group. A strictly conserved glutamate residue is suggested to function catalytically in thioester proteins. Structure-based design of inhibitors of C3 activation may target a conserved pocket between the alpha-chain and the beta-chain of C3, which appears essential for conformational changes in C3. 相似文献

Enterocytes maintain fluid-electrolyte homeostasis by keeping a tight barrier and regulating ion channels. Carbon monoxide (CO), a product of heme degradation, modulates electrolyte transport in kidney and lung epithelium, but its role in regulating intestinal fluid-electrolyte homeostasis has not been studied. The major source of endogenous CO formation comes from the degradation of heme via heme oxygenase. We hypothesized that heme activates electrolyte transport in intestinal epithelial cells. Basolateral hemin treatment increased baseline Caco-2 cell short-circuit currents (I(sc)) twofold (control = 1.96 +/- 0.14 microA/cm(2) vs. hemin = 4.07 +/- 0.16 microA/cm(2), P < 0.01); apical hemin had no effect. Hemin-induced I(sc) was caused by Cl- secretion because it was inhibited in Cl- -free medium, with ouabain, 5-nitro-2-(3-phenylpropylamino) benzoic acid (NPPB), or DIDS. Apical electrogenic Na+ channel inhibitor benzamil had no effect on hemin-induced I(sc). Hemin did not alter the ability of Caco-2 cells to respond maximally to forskolin, but a soluble guanylate cyclase inhibitor, [1,2,4]oxadiazolo-[4,3-a]quinoxalin-1-one (ODQ) inhibited the effects of hemin. A CO-releasing molecule, tricarbonyldichlororuthenium II, induced active Cl- secretion that was also inhibited with ODQ. We conclude that hemin induces active Cl- secretion in Caco-2 cells via a cGMP-dependent pathway. These effects are probably the consequence of CO formation. A morphological transformation involving loss of adhesion between myelin lamellae and formation of myelin vesicles has been described as a mechanism for demyelination in multiple sclerosis and marmoset experimental allergic encephalomyelitis (EAE). Although protein interactions are involved in maintaining normal myelin structure, we describe here how lipids contribute to myelin stability and how lipid changes in EAE, including increases in lipid polyunsaturation and negatively charged phosphatidylserine (PS), promote demyelination. Three physico-chemical techniques were used to identify these changes: (1) Langmuir monolayer isotherms indicated that EAE white matter lipids were significantly more "expanded" (fluid) than controls. (2) NMR spectroscopy indicated that EAE myelin lipids were more polyunsaturated than controls. (3) High-performance liquid chromatography (HPLC) with an evaporative light scattering detector indicated increased PS in EAE compared to controls, while sphingomyelin (SM), sulfatides and phosphatidylcholine (PC) were decreased. We present a physical model considering electrostatic, van der Waals and undulation forces to quantify the effect of these changes on myelin adhesion at the extracellular interface. Taken together, the isotherm, NMR, HPLC and modeling results support a mechanism for autoimmune demyelination whereby the composition of myelin lipids is altered in a manner that increases myelin fluidity, decreases myelin adhesion, increases membrane curvature, and promotes vesiculation. 相似文献

Eight different winter barley genotypes were grown in a plot experiment in northern Denmark, on soil where Mn deficiency had been previously demonstrated, to assess field performance and grain yield under two treatments: with foliar Mn fertilisation and without foliar Mn fertilisation. A high Mn efficiency was demonstrated for two of the genotypes and a low Mn efficiency was observed for one of the genotypes in three consecutive years; highly variable results were obtained for other genotypes. Mn efficiency based on grain yields could not be related to whole shoot Mn concentration, biomass or Mn uptake at various vegetative growth stages. Chlorophyll a fluorescence analysis was able to differentiate between the two Mn treatments up to 7weeks after the last Mn application, whereas Mn concentrations, shoot biomass and Mn accumulation could not. It was not possible to fully alleviate Mn deficiency by repeated foliar spraying on the control plots. Given the growing conditions at the trial site and the low Mn in plant tissue, it is possible that Mn deficiency was extreme enough that Mn efficiency mechanisms broke down. 相似文献

Lack of geographic variation in anonymous nuclear polymorphisms in the American oyster, Crassostrea virginica 总被引:1,自引:0,他引:1
Comparing geographic variation of noncoding nuclear DNA polymorphisms,
which presumably are neutral to natural selection, with geographic
variation of allozymes is potentially a good way to detect the effects of
selection on allozyme polymorphisms. A previous study of four anonymous
nuclear markers in the American oyster, Crassostrea virginica, found
dramatic differences in allele frequency between the Gulf of Mexico and the
Atlantic Ocean. In contrast, 14 allozyme polymorphisms were fairly uniform
in frequency between the two areas. This led to the conclusion that all of
the allozyme polymorphisms were kept uniform in frequency by balancing
selection. To test the robustness of this pattern, six additional anonymous
nuclear DNA polymorphisms were surveyed in oysters from Panacea, Fla, and
Charleston, S.C. on the Gulf and Atlantic coasts, respectively. Unlike the
previously studied DNA markers, the six DNA polymorphisms examined here
show geographic variation that is not significantly greater than that of
allozymes. The reason for the discrepancy between the two sets of DNA
polymorphisms is unclear.
