Mitochondrial DNA variation and genetic structure in populations of Drosophila melanogaster 总被引:5,自引:0,他引:5
The understanding of the genetic structure of a species can be improved by
considering together data from different types of genetic markers. In the
past, a number of worldwide populations of Drosophila melanogaster have
been extensively studied for several such markers, including allozymes,
chromosomal inversions, and quantitative characters. Here we present
results from a study of restriction- fragment-length polymorphisms of
mitochondrial DNA (mtDNA) in 92 isofemale lines from many of the same
geographic populations of D. melanogaster. Eleven restriction enzymes were
used, of which four revealed restriction-site polymorphism. A total of 24
different haplotypes were observed, of which 18 were unique to single
populations. In many populations, the unique haplotypes have reached high
frequency without being observed in neighboring populations. A Wagner
parsimony tree reveals that mutationally close variants show geographical
clumping, suggesting local differentiation of mtDNA in populations. The
Old-World and the New-World populations are differentiated, with the
predominant Old-World haplotype being virtually absent from the New World.
These results contrast with those for the nuclear genes, in which many loci
show parallel clines in different continents, and suggest a common origin
of D. melanogaster populations in North America.

The human T cell receptor for antigen (Ti) has recently been identified on IL-2 dependent T cell clones as a 90 kd disulfide-linked heterodimer comprised of one 49-51 kd alpha (alpha) and one 43 kd beta (beta) chain. These subunits are noncovalently associated with a monomorphic 20-25 kd T3 molecule. Here, we produce monoclonal antibodies to a human tumor (REX) derived from an earlier stage of thymic differentiation in order to determine whether clonotypic structures are expressed and to define the ontogeny of Ti. The results of SDS-PAGE and peptide map analyses indicate that an homologous T3-associated heterodimer is synthesized and expressed by REX. This glycoprotein shares several peptides in common with clonotypic structures on an IL-2 dependent T cell clone. In addition, similar Ti related molecules appear during intrathymic ontogeny in parallel with surface T3 expression. The latter findings provide the structural basis for the immunological competence observed exclusively within the T3+ thymocyte compartment. 相似文献

This publication looks specifically at soil and plant nematology. First, the societal impact of nematodes and benefits of nematology research are briefly presented. Next, the opportunities facing nematology in the next decade are outlined, as well as the resources needed to address these priorities. A quantitative method is described which permits a determination of the relative amount of trypsin or pepsin present in a gelatin-enzyme digestion mixture, provided the gelatin and trypsin solutions are purified. This method is dependent upon the change in viscosity of such solutions. It is found that the time required to cause a given percentage change in the viscosity is nearly inversely proportional to the amount of enzyme present. It is pointed out that the particular value of the method lies in the fact that enzyme reactions which take place in the presence of "buffer" salts may be studied. 相似文献
