Illus. 1961.—A study of the distribution of sclereids in cleared leaves taken from 1-, 2-, and 4-year-old shoots of an adult tree of Pseudotsuga menziesii (Mirb.) Franco showed a repeated pattern of sclereid distribution along the shoot axis with many sclereids in the basal leaves grading into few or no sclereids in the terminal leaves of each year's growth. Attempts were made to influence sclereid distribution by bud defoliation of attached branches with and without auxin treatment and by testing the effects of growth-regulating substances on sclereid formation in leaves of excised buds of Pseudotsuga cultured in vitro. Whereas removal of the basal ¾ of the leaves at the time of bud unfolding had no effect on bud, leaf or sclereid development, removal of the leaves of the upper half or complete defoliation led to premature expansion of next year's terminal bud with leaves developing in part from presumptive bud-scale primordia. Indoleacetic acid at 0.5% in lanolin paste applied to the defoliated region prevented this premature bud expansion. Defoliation of the basal half did not affect sclereid formation in the terminal leaves. Sclereid development in leaves of prematurely expanded buds on defoliated branches was normal except in the few cases where bud expansion occurred in the presence of low-auxin concentrations. Then, sclereid development was inhibited. Sclereid formation in leaves of excised buds grown in nutrient culture was generally much less frequent than in intact branches, and auxin treatment still further reduced the frequency of sclereids. It was concluded that sclereid initiation and differentiation in the intact plant may well be under the control of hormonal factors in the plant, one of which may be auxin. 相似文献
S100A13 is a 98-amino acid, calcium binding protein. It is known to participate in the non-classical secretion of signal peptide-less proteins, such as the acidic fibroblast growth factor. In this study, we investigate the lipid binding properties of S10013 using a number of biophysical techniques, including multidimensional NMR spectroscopy. Isothermal titration calorimetry and steady state fluorescence experiments show that apoS100A13 exhibits preferential binding to small unilamelar vesicles of l-phosphatidyl serine (pS). In comparison, Ca2+-bound S100A13 is observed to bind weakly to unilamelar vesicles (SUVs) of pS. Equilibrium thermal unfolding and limited trypsin digestion analysis reveal that apoS100A13 is significantly destabilized upon binding to SUVs of pS. Results of the far UV circular dichroism and ANS (8-anilino-1-napthalene sufonate) binding experiments indicate a subtle conformational change resulting in the increase in the solvent-accessible hydrophobic surface in the protein. Availability of the solvent-exposed hydrophobic surface(s) in apoS10013 facilitates its interaction with the lipid vesicles. Our data suggest that Ca2+ binding dictates the membrane binding affinity of S100A13. Based on the results of this study, a model describing the sequence of molecular events that possibly can occur during the non-classical secretion of FGF-1 is presented. 相似文献
Hydatidiform mole is a benign trophoblastic neoplasia characterized by an abnormal development of the embryo and proliferation
of placental villi. Using microsatellite markers amplified by the polymerase chain reaction, we have performed a genetic study
on eight independent molar tissues occurring in two sisters. Karyotype and genotype data demonstrate a diploid and biparental
constitution in seven of the analyzed moles suggesting a common mechanism underlying the etiology of the various molar pregnancies
in this family. The data reported here suggest that complete and partial hydatidiform moles are not always separate entities
and that women with familial recurrent hydatidiform moles are homozygous for an autosomal recessive mutation.

The liver is an important organ, and hepatic ischemia–reperfusion (IR) injury is a frequent pathophysiological process that can cause significant morbidity and mortality. Thus, our study aimed to investigate the effect of targeting PI3K/p-Akt/eNOS (phosphoinositide 3-kinase/phospho-protein kinase B/endothelial nitric oxide synthase), Nrf2/HO-1 (nuclear factor-erythroid 2-related factor-2/heme oxygenase-1), and NF-κB/p53 (nuclear factor-κB/tumor protein 53) signaling pathways by using angiotensin (1–7) [ang-(1–7)] against hepatic injury induced by IR. Thirty-two male rats were included in sham group, ang-(1–7)-treated group, hepatic IR group, and hepatic IR group treated with ang-(1–7). The levels of hepatic ang-(1–7), angiotensin II (Ang II), angiotensin-converting enzyme 2 (ACE2), HO-1, malondialdehyde (MDA), PI3K, and p-Akt were assessed. The expressions of eNOS and B-cell leukemia/lymphoma-2 (BCL-2) in the liver were determined. Histological assessment and immunohistochemical expression of NF-κB, p53, and Nrf2 were carried out. The levels of reduced glutathione (GSH), aspartate aminotransferase (AST), alanine aminotransferase (ALT), tumor necrosis factor-α (TNF-α), and interleukin-6 (IL-6) in serum were estimated. Results showed that administration of ang-(1–7) to hepatic IR rats led to significant amelioration of hepatic damage through a histological evaluation that was associated with significant upregulation of the expressions of PI3K/p-Akt/eNOS and Nrf2/HO-1 with downregulation of NF-κB/p53 signaling pathways. In conclusion, PI3K/p-Akt/eNOS and Nrf2/HO-1 signaling pathways are involved in the protective effects of ang-(1–7) against hepatic damage induced by IR. Therefore, ang-(1–7) can be used to prevent hepatic IR, which occurs in certain conditions such as liver transplantation, hemorrhagic shock, and severe infection. 相似文献
The current investigation aims to develop and evaluate novel ocular proniosomal gels of lomefloxacin HCl (LXN); in order to improve its ocular bioavailability for the management of bacterial conjunctivitis. Proniosomes were prepared using different types of nonionic surfactants solely and as mixtures with Span 60. The formed gels were characterized for entrapment efficiency, vesicle size, and in vitro drug release. Only Span 60 was able to form stable LXN-proniosomal gel when used individually while the other surfactants formed gels only in combination with Span 60 at different ratios. The optimum proniosomal gel; P-LXN 7 (Span 60:Tween 60, 9:1) appeared as spherical shaped vesicles having high entrapment efficiency (>80%), appropriate vesicle size (187?nm) as well as controlled drug release over 12?h. Differential scanning calorimetry confirmed the amorphous nature of LXN within the vesicles. Stability study did not show any significant changes in entrapment efficiency or vesicle size after storage for 3 months at 4?°C. P-LXN 7 was found to be safe and suitable for ocular delivery as proven by the irritancy test. The antibacterial activity of P-LXN 7 evaluated using the susceptibility test and topical therapy of induced ocular conjunctivitis confirmed the enhanced antibacterial therapeutic efficacy of the LXN-proniosomal gel compared to the commercially available LXN eye drops. 相似文献
Context: Topical treatment of skin disease needs to be strategic to ensure high drug concentration in the skin with minimum systemic absorption.
Objective: The aim of this study was to produce semisolid nanostructured lipid carrier (NLC) formulations, for topical delivery of the corticosteroid drug, diflucortolone valerate (DFV), with minimum systemic absorption.
Method: NLC formulations were developed using a high shear homogenization combined with sonication, using Precirol® ATO5 or Tristearin® as the solid lipid, Capryol? or isopropyl myristate as the liquid lipid and Poloxamer® 407 as surfactant. The present study addresses the influence of different formulations composition as solid lipid, liquid lipid types and concentrations on the physicochemical properties and drug release profile from NLCs.
Results and discussion: DFV-loaded NLC formulations possessed average particle size ranging from 160.40?nm to 743.7?nm with narrow polydispersity index. The encapsulation efficiency was improved by adding the lipid-based surfactants (Labrasol® and Labrafil® M1944CS) to reach 68%. The drug release from the investigated NLC formulations showed a prolonged release up to 12?h. The dermatopharmacokinetic study revealed an improvement in drug deposition in the skin with the optimized DFV-loaded NLC formulation, in contrast to a commercial formulation.
Conclusion: NLC provides a promising nanocarrier system that work as reservoir for targeting topical delivery of DFV. 相似文献
