This paper investigates the impact of increased salinity on touch-induced receptor and action potentials of Chara internodal cells. We resolved underlying changes in ion transport by current/voltage analysis. In a saline medium with a low Ca(2+) ion concentration [(Ca(2+))(ext)], the cell background conductance significantly increased and proton pump currents declined to negligible levels, depolarizing the membrane potential difference (PD) to the excitation threshold [action potential (AP)(threshold)]. The onset of spontaneous repetitive action potentials further depolarized the PD, activating K(+) outward rectifying (KOR) channels. K(+) efflux was then sustained and irrevocable, and cells were desensitized to touch. However, when [Ca(2+)](ext) was high, the background conductance increased to a lesser extent and proton pump currents were stimulated, establishing a PD narrowly negative to AP(threshold). Cells did not spontaneously fire, but became hypersensitive to touch. Even slight touch stimulus induced an action potential and further repetitive firing. The duration of each excitation was extended when [Ca(2+)](ext) was low. Cell viability was prolonged in the absence of touch stimulus. Chara cells eventually depolarize and die in the saline media, but touch-stimulated and spontaneous excitation accelerates the process in a Ca(2+)-dependent manner. Our results have broad implications for understanding the interactions between mechano-perception and salinity stress in plants.  相似文献

Acoustic communication among birds plays an important role in attracting mates and defending territories. For the successful transmission of songs, individuals of different species often avoid singing at the same time to reduce acoustic interference from background noise and overlapping signals from heterospecifics. Such behavioural acoustic niche partitioning may occur especially among closely related species due to their ecological similarities. In this study, we recorded bird sounds in a subtropical forest in China in May–June 2019 and detected seven cuckoo species. Extracting characteristics of the cuckoo calls, we found that only four of the 21 pairs of species overlapped in frequency range, and 19 pairs were classified accurately using a linear discriminant analysis classifier based on their features. The remaining two species pairs could be separated based on temporal or spatial distribution patterns. We also analysed the temporal distribution patterns and overlap time of the calls, finding that the seven species exhibit partitioning in at least one of three acoustic dimensions (site, frequency, activity time). We conclude that the seven sympatric cuckoo species were strongly partitioned in acoustic signal space and minimally masked each other's signals.  相似文献

Mariner-like elements (MLEs) are widespread transposable elements in animal genomes. They have been divided into at least five sub-families with differing host ranges. We investigated whether the ability of transposases encoded by Mos1, Himar1 and Mcmar1 to be actively imported into nuclei varies between host belonging to different eukaryotic taxa. Our findings demonstrate that nuclear importation could restrict the host range of some MLEs in certain eukaryotic lineages, depending on their expression level. We then focused on the nuclear localization signal (NLS) in these proteins, and showed that the first 175 N-terminal residues in the three transposases were required for nuclear importation. We found that two components are involved in the nuclear importation of the Mos1 transposase: an SV40 NLS-like motif (position: aa 168 to 174), and a dimerization sub-domain located within the first 80 residues. Sequence analyses revealed that the dimerization moiety is conserved among MLE transposases, but the Himar1 and Mcmar1 transposases do not contain any conserved NLS motif. This suggests that other NLS-like motifs must intervene in these proteins. Finally, we showed that the over-expression of the Mos1 transposase prevents its nuclear importation in HeLa cells, due to the assembly of transposase aggregates in the cytoplasm.  相似文献

The effect of varying membrane capacitance, conductance, and rate constants on the properties of the nerve impulse is considered in terms of the degree of regeneration in the Hodgkin-Huxley model for the squid giant axon. It is shown through computer simulation that reducing regeneration generally increases the duration of the action potential and decreases its amplitude, rate of rise, and conduction velocity. The threshold becomes much less sharp and the amplitude of the response of a patch of membrane grades with stimulus strength. A second stimulus, applied shortly after a first stimulus, considerably perturbs the membrane potential from its original time-course. Under certain conditions, the nerve signal can propagate with a small decrement.  相似文献

We demonstrate a novel synthesis strategy for the preparation of Pr-doped SrTiO3 ceramics via a combination of solid state reaction and spark plasma sintering techniques. Polycrystalline ceramics possessing a unique morphology can be achieved by optimizing the process parameters, particularly spark plasma sintering heating rate. The phase and morphology of the synthesized ceramics were investigated in detail using X-ray diffraction, scanning electron microcopy and energy-dispersive X-ray spectroscopy. It was observed that the grains of these bulk Pr-doped SrTiO3 ceramics were enhanced with Pr-rich grain boundaries. Electronic and thermal transport properties were also investigated as a function of temperature and doping concentration. Such a microstructure was found to give rise to improved thermoelectric properties. Specifically, it resulted in a significant improvement in carrier mobility and the thermoelectric power factor. Simultaneously, it also led to a marked reduction in the thermal conductivity. As a result, a significant improvement (> 30%) in the thermoelectric figure of merit was achieved for the whole temperature range over all previously reported maximum values for SrTiO3-based ceramics. This synthesis demonstrates the steps for the preparation of bulk polycrystalline ceramics of non-uniformly Pr-doped SrTiO3.  相似文献

Nitric oxide(NO) is synthesized in normal muscle fibers by the neuronal (nNOS) andthe endothelial (ecNOS) isoforms of nitric oxide synthase (NOS). NOcontributes to the regulation of several processes such asexcitation-contraction coupling and mitochondrial respiration. Weassessed in this study whether NO production is regulated in responseto an acute increase in muscle activation. Three groups ofanesthetized, tracheostomized, spontaneously breathing rats wereexamined after an experimental period of 3 h. Group 1 served as a control (no loading), whereasgroups 2 and3 were exposed to moderate and severeinspiratory resistive loads, respectively, which elicited trachealpressures of 30 and 70% of maximum, respectively. Ventilatory(diaphragm, intercostal, and transverse abdominis) and limb(gastrocnemius) muscles were excised at the end of the experimentalperiod and examined for NOS activity and NOS protein expression.Neither submaximal nor maximum tracheal pressures were altered after 3 h of resistive loading. Diaphragmatic and intercostal muscle NOSactivities declined significantly in response to moderate and severeloading, whereas those of transverse abdominis and gastrocnemiusmuscles remained unchanged. On the other hand, resistive loading had nosignificant effect on ventilatory and limb muscle NOS isoformexpression. We propose that a contraction-induced decline in muscle NOSactivity represents a compensatory mechanism through which musclecontractility and mitochondrial function are protected from theinhibitory influence of NO.

The aim of this investigation was to evaluate the effects of 2 different cold atmospheric plasma (CAP) sources, photodynamic therapy and sodium hypochlorite (NaOCl), on infected root canals. Therefore, 50 standardized curved human root canals were infected with Enterococcus faecalis and assigned to 5 groups—negative control (NC), plasma jet (CAP I), dielectric barrier discharge (CAP II), photodynamic therapy (PDT) and NaOCl + passive ultrasonic irrigation—for 30 s. Colony forming units (CFUs) were determined. NaOCl was significantly more effective at reducing CFUs than all test groups (P < .0001 [Mann‐Whitney U test]) in both parts of the root canal. CFUs in PDT were significantly lower than those in CAP II (P = .015), and those in CAP I were lower than those in CAP II (P = .05). Among all other groups and in the apical parts, no significant differences were found (P > .05).   相似文献

DNA double‐strand breaks (DSBs) can be repaired by two major pathways: non‐homologous end‐joining (NHEJ) and homologous recombination (HR). DNA repair pathway choice is governed by the opposing activities of 53BP1, in complex with its effectors RIF1 and REV7, and BRCA1. However, it remains unknown how the 53BP1/RIF1/REV7 complex stimulates NHEJ and restricts HR to the S/G2 phases of the cell cycle. Using a mass spectrometry (MS)‐based approach, we identify 11 high‐confidence REV7 interactors and elucidate the role of SHLD2 (previously annotated as FAM35A and RINN2) as an effector of REV7 in the NHEJ pathway. FAM35A depletion impairs NHEJ‐mediated DNA repair and compromises antibody diversification by class switch recombination (CSR) in B cells. FAM35A accumulates at DSBs in a 53BP1‐, RIF1‐, and REV7‐dependent manner and antagonizes HR by limiting DNA end resection. In fact, FAM35A is part of a larger complex composed of REV7 and SHLD1 (previously annotated as C20orf196 and RINN3), which promotes NHEJ and limits HR. Together, these results establish SHLD2 as a novel effector of REV7 in controlling the decision‐making process during DSB repair.  相似文献

A simple and scalable direct laser machining process to fabricate MXene‐on‐paper coplanar microsupercapacitors is reported. Commercially available printing paper is employed as a platform in order to coat either hydrofluoric acid‐etched or clay‐like 2D Ti3C2 MXene sheets, followed by laser machining to fabricate thick‐film MXene coplanar electrodes over a large area. The size, morphology, and conductivity of the 2D MXene sheets are found to strongly affect the electrochemical performance due to the efficiency of the ion‐electron kinetics within the layered MXene sheets. The areal performance metrics of Ti3C2 MXene‐on‐paper microsupercapacitors show very competitive power‐energy densities, comparable to the reported state‐of‐the‐art paper‐based microsupercapacitors. Various device architectures are fabricated using the MXene‐on‐paper electrodes and successfully demonstrated as a micropower source for light emitting diodes. The MXene‐on‐paper electrodes show promise for flexible on‐paper energy storage devices.  相似文献
