Background

Isolated growth hormone deficiency (IGHD) and multiple pituitary hormone deficiency (MPHD) are heterogeneous disorders with several different etiologies and they are responsible for most cases of short stature. Mutations in different genes have been identified but still many patients did not present mutations in any of the known genes. Chromosomal rearrangements may also be involved in short stature and, among others, deletions of 18q23 defined a critical region for the disorder. No gene was yet identified.

Methodology/Principal Findings

We now report a balanced translocation X;18 in a patient presenting a breakpoint in 18q23 that was surprisingly mapped about 500 Kb distal from the short stature critical region. It separated from the flanking SALL3 gene a region enriched in highly conserved non-coding elements (HCNE) that appeared to be regulatory sequences, active as enhancers or silencers during embryonic development.

Conclusion

We propose that, during pituitary development, the 18q rearrangement may alter expression of 18q genes or of X chromosome genes that are translocated next to the HCNEs. Alteration of expression of developmentally regulated genes by translocation of HCNEs may represent a common mechanism for disorders associated to isolated chromosomal rearrangements.  相似文献

Biodynamics and injury potential of operators in stand-up rider lift truck accidents have been investigated with a special focus on head injury. An anthropomorphic test device (ATD) model was used as an operator surrogate in computer simulations of off-the-dock (OTD) and tip-over (TO) accidents. The biomechanical model representing the ATD was developed based on rigid body segments, and then combined with a rigid body truck model in the accident simulations. The operator compartment of the truck model was enclosed with a rear door. The computed kinematics are in agreement with the results of previous experimental testing. A 2D finite element model of the head was created to compute head impact decelerations in the sagittal plane. Values of the head injury criterion for the TO cases were computed from the model and shown to compare favourably with experimental values. The results advance the state of knowledge concerning injury potential in TO and OTD accidents and simulation models for such accidents.  相似文献

Lupoid leishmaniasis is a unique form of cutaneous leishmaniasis characterized by unusual clinical features and a chronic relapsing course, mostly caused by infection with Leishmania tropica. In this clinical form, 1-2 yr after healing of the acute lesion, new papules and nodules appear at the margin of the remaining scar. Herein, we describe a case of this clinical form that was resistant to 2 courses of treatments: systemic glucantime and then a combination therapy with allopurinol and systemic glucantime. However, marked improvement was seen after a combination therapy with topical trichloroacetic acid solution (50%) and systemic glucantime, and there were no signs of recurrence after 1 yr of follow-up.  相似文献

Among the 49 strains of moderately halophilic bacteria isolated from the salty environments of Iran, a Gram-positive coccus designated as strain QW6 showed high capacity in the removal of toxic oxyanions of tellurium in a wide range of culture medium factors including pH (5.5-10.5), temperature (25-45 degrees C), various salts including NaCl, KCl, and Na(2)SO(4) (0.5-4M), selenooxyanions (2-10mM), and at different concentrations of potassium tellurite (0.5-1mM) under aerobic condition. Phenotypic characterization and phylogenetic analyses based on 16S rDNA sequence comparisons indicated that this strain was a member of the genus Salinicoccus. The maximum tellurite removal was exhibited in 1.5M NaCl at 35 degrees C, while the activity reduced by 53% and 47% at 25 and 45 degrees C, respectively. The optimum pH for removal activity was shown to be 7.5, with 90% and 83% reduced removal capacities at the two extreme values of 5.5 and 10, respectively. The impact of different concentrations of selenooxyanions (2-10mM) on tellurite removal by strain QW6 was evaluated. The ability of strain QW6 in the removal of tellurite in the presence of 6mM selenite increased by 25%. The concentration of toxic potassium tellurite in the supernatant of the bacterial culture medium decreased by 99% (from 0.5 to 0.005mM) after 6 days and the color of the medium changed to black due to the formation of less toxic elemental tellurium.  相似文献

The nuclear receptor Ad4BP/SF-1 is essential for development of the adrenal cortex and the gonads, which derive from a common adrenogonadal primordium. The adrenal cortex subsequently forms morphologically distinct compartments: the inner (fetal) and outer (definitive or adult) zones. Despite considerable effort, the mechanisms that mediate the differential development of the adrenal and gonadal primordia and the fetal and adult adrenal cortices remain incompletely understood. We previously identified a fetal adrenal-specific enhancer (FAdE) in the Ad4BP/SF-1 locus that directs transgene expression to the fetal adrenal cortex and demonstrated that this enhancer is autoregulated by Ad4BP/SF-1. We now combine the FAdE with the Cre/loxP system to trace cell lineages in which the FAdE was active at some stage in development. These lineage-tracing studies establish definitively that the adult cortex derives from precursor cells in the fetal cortex in which the FAdE was activated before the organization into two distinct zones. The potential of these fetal adrenocortical cells to enter the pathway that eventuates in cells of the adult cortex disappeared by embryonic day 14.5. Thus, these studies demonstrate a direct link between the fetal and adult cortices involving a transition that must occur before a specific stage of development.  相似文献

Atherosclerotic plaques develop in regions of the vasculature associated with chronic inflammation due to disturbed flow patterns. Endothelial phenotype modulation by flow requires the integration of numerous mechanotransduction pathways, but how this is achieved is not well understood. We show here that, in response to flow, the adaptor protein Shc is activated and associates with cell-cell and cell-matrix adhesions. Shc activation requires the tyrosine kinases vascular endothelial growth factor receptor 2 and Src. Shc activation and its vascular endothelial cadherin (VE-cadherin) association are matrix independent. In contrast, Shc binding to integrins requires VE-cadherin but occurs only on specific matrices. Silencing Shc results in reduction in both matrix-independent and matrix-dependent signals. Furthermore, Shc regulates flow-induced inflammatory signaling by activating nuclear factor kappaB-dependent signals that lead to atherogenesis. In vivo, Shc is activated in atherosclerosis-prone regions of arteries, and its activation correlates with areas of atherosclerosis. Our results support a model in which Shc orchestrates signals from cell-cell and cell-matrix adhesions to elicit flow-induced inflammatory signaling.  相似文献

In Southeast Asian tropical rainforests, community-level masting (CM) occurs at irregular intervals of 2–10 years. During CM periods, many plant species from various families synchronously flower and subsequently undergo community-level fruiting. Seed predation is a key factor in understanding the ecological and evolutionary factors affecting CM. Masting is proposed to decrease seed mortality due to predation in two ways: by depressing predator abundance through extended and unpredictable absences of seeds; and by satiating predators via mass seed production (predator satiation hypothesis). If the hypothesis is valid in these rainforests, the incidence of seed predation will be higher in a fruiting event that occurs soon after a previous fruiting event, because the intervening period of seed absence would be inadequate to starve the predators. In this study, we examined seed predation by insects, focusing on five dipterocarp species that exceptionally reproduced twice during an extended CM period. All of the five species suffered more intense seed predation in the second fruiting event, consistent with the prediction expected from the predator satiation hypothesis. Weevils, bark beetles and mammals were the main cause of increased seed predation in three, one and one plant species, respectively. However, seed predation intensity did not increase during the second fruiting event in a few combinations of predator and plant species. We discuss the possibility that competition for seeds among predators and/or the interspecific differences in life history traits among predators might affect the varying intensities of seed predation among dipterocarp species by different seed predators.  相似文献

Crested wheatgrass (Agropyron cristatum L.) is a cool-season perennial grass, which has demonstrated its potential for use as turfgrass. However, limited information is available on its drought and salinity tolerance. The main purpose of this study was to investigate the changes in the antioxidant defence system and physiological traits of six Iranian crested wheatgrass genotypes under drought and salinity stresses. The experimental design comprised a split plot with water treatments (control well-watered, salinity stress and water stress) as main plots and genotypes as subplots. This study demonstrated the variations in drought and salinity tolerance among crested wheatgrass genotypes. ‘ACSKI’, ‘ACAMI’ and ‘ACDTI’ generally performed better than other genotypes under drought and salinity conditions, mainly by maintaining higher activities of antioxidant enzymes like superoxide dismutase, peroxidase, catalase, ascorbate peroxidase and non-enzyme antioxidants like glutathione, higher proline and total non-structural carbohydrates content. The increased, decreased, and unchanged activities of antioxidant enzymes in the crested wheatgrass genotypes indicates a different forms of metabolism of antioxidant enzymes in response to drought and salinity stress. In general, drought and salinity stress increases the malondialdehyde (MDA) content and hydrogen peroxide (H2O2) content; however, ‘ACSKI’, ‘ACAMI’ and ‘ACDTI’ genotypes could tolerate an increase in MDA content and H2O2 content; therefore, lower levels of MDA content and H2O2 content were observed. The results showed that increasing levels of diamine oxidase and polyamine oxidase have been associated with increasing drought and salinity tolerance. Based on the biochemical and physiological parameters that were evaluated, we concluded that the genotype ‘ACSKI’ was superior in terms of drought and salinity tolerance. This superiority was mainly a result of better enzymatic and non-enzymatic antioxidant defence system and better osmotic adjustment under stress conditions.  相似文献

Objective

To develop a deliberately engineered expression and purification system for an active chimeric-recombinant tissue plasminogen activator (crtPA) using co-expression with polyhydroxybutyrate (PHB) operon genes.

Results

Fusion of crtPA with PhaC-synthase simplified the purification steps through crtPA sedimentation with PHB particles. Moreover, the covalently immobilized crtPA was biologically active as shown in a chromogenic assay. Upon WELQut-protease activity, the released single-chain crtPA converted to the two-chain form which produced a pattern of bands with approx. MW of 32 and 11 kDa in addition to the full length crtPA.

Conclusion

Fusion of crtPA with PhaC-synthase not only simplifies purification from the bacterial host lysate, but also co-expression of PHB operon genes creates an oxidative environment, thereby reducing the inclusion body formation possibility. The isolated crtPA-PHB granules exhibited crtPA serine protease activity. Thus, fusion with the PhaC protein could be used as a scaffold for covalent displaying of functional disulfide-rich proteins.
