The HIV-1 epidemic in sub-Saharan Africa is driven largely by heterosexual transmission of non-subtype B viruses, of which subtypes C and A are predominant. Previous studies of subtype B and subtype C transmission pairs have suggested that a single variant from the chronically infected partner can establish infection in their newly infected partner. However, in subtype A infected individuals from a sex worker cohort and subtype B individuals from STD clinics, infection was frequently established by multiple variants. This study examined over 1750 single-genome amplified viral sequences derived from epidemiologically linked subtype C and subtype A transmission pairs very early after infection. In 90% (18/20) of the pairs, HIV-1 infection is initiated by a single viral variant that is derived from the quasispecies of the transmitting partner. In addition, the virus initiating infection in individuals who were infected by someone other than their spouse was characterized to determine if genital infections mitigated the severe genetic bottleneck observed in a majority of epidemiologically linked heterosexual HIV-1 transmission events. In nearly 50% (3/7) of individuals infected by someone other than their spouse, multiple genetic variants from a single individual established infection. A statistically significant association was observed between infection by multiple genetic variants and an inflammatory genital infection in the newly infected individual. Thus, in the vast majority of HIV-1 transmission events in cohabiting heterosexual couples, a single genetic variant establishes infection. Nevertheless, this severe genetic bottleneck can be mitigated by the presence of inflammatory genital infections in the at risk partner, suggesting that this restriction on genetic diversity is imposed in large part by the mucosal barrier.  相似文献

The biochemical and molecular mechanisms used by alkaliphilic bacteria to acquire iron are unknown. We demonstrate that alkaliphilic (pH > 9) Bacillus species are sensitive to artificial iron (Fe3+) chelators and produce iron-chelating molecules. These alkaliphilic siderophores contain catechol and hydroxamate moieties, and their synthesis is stimulated by manganese(II) salts and suppressed by FeCl3 addition. Purification and mass spectrometric characterization of the siderophore produced by Caldalkalibacillus thermarum failed to identify any matches to previously observed fragmentation spectra of known siderophores, suggesting a novel structure.Iron is an abundant element in nature; however, in most aqueous aerobic environments iron forms insoluble ferric hydroxide, Fe(OH)3. This poses a major problem for most aerobic bacteria, as ferric hydroxide has a solubility constant of 10−39 M, therefore limiting the concentration of ferric ions to 10−18 M at pH 7.0. For example, bacteria living in seawater (approximate pH 8.0) require iron, yet dissolved iron is only present at 0.02 to 2.0 nM (5). Despite this apparent lack of bioavailability, iron has been repeatedly demonstrated to be an essential element for aerobic bacterial growth (1).With the lack of readily accessible iron at physiological pH, most bacteria have evolved systems to deal with the incumbent problem of iron acquisition. Under iron-rich conditions, Fe2+ uptake receptors, such as FeoAB, are synthesized in bacteria, which passively import iron in the immediate vicinity of the cell (1, 23). No equivalent system has been identified for Fe3+ transport. To acquire Fe3+ under aqueous aerobic conditions, bacteria commonly have import systems involving the synthesis, secretion, and regathering of a group of secondary metabolites known as siderophores (1, 11). Siderophores are low-molecular-weight chemical moieties that chelate Fe3+ and typically have complex formation (Kf) constants in the range of 1023 to 1052 (11). Siderophores, like other chelators, are known to increase the solubility of iron by hindering the formation of Fe-oxyhydroxides at high pH, at which the Fe-oxyhydroxides are the dominating inorganic species (27). Siderophores are also known to facilitate the dissolution of Fe from minerals (3). Siderophore-iron complexes can either be transported through cellular membranes using dedicated transport systems or if the Fe(III) central atom is reduced, making the iron bioavailable for cellular processes (10, 14). Three major groups of siderophores have been described in bacteria: hydroxamates, catecholates, and carboxylates. Hydroxamates and catechols are commonly produced by aerobic bacteria living at neutral to alkaline pH, whereas carboxylates are significantly more common in bacteria living in mildly acidic pH (11-13). In the genus Bacillus, Bacillus megaterium and Bacillus subtilis are producers of schizokinen and bacillibactin, respectively (6, 20). Bacillus anthracis produces both a catechol and a hydroxamate siderophore (7, 34), and B. licheniformis strain VK21 is the only known example of a thermoresistant catecholate-producing Gram-positive bacterium (32).Although there is extensive literature on iron capture mechanisms in bacteria that thrive at neutral pH, there is little information at a biochemical or molecular level on how aerobic bacteria growing at extreme alkaline pHs (i.e., pH 9 to 11) acquire iron. At alkaline pH, the solubility constant for iron decreases far below the requirement for living cells, and the concentration of bioavailable iron is estimated to be approximately 10−23 M at pH 10 (11). Taking this extreme lack of iron into account, the sequestering mechanisms of alkaliphilic bacteria must be powerful, yet there has been little analysis of the types of iron-chelating molecules these bacteria produce.  相似文献

In order to understand wabusk (polar bear, Ursus maritimus) behaviours and interactions with people in the Hudson Bay lowlands of northern Ontario we conducted this collaborative study of Cree kiskayndamowin/knowledge. Our findings reveal that Cree knowledge supports previously published information on polar bears, while adding further contextual findings: that male polar bears travel greater distances into the muskeg than previously recorded; that wabusk prey on amisk (beaver, Castor canadensis); that wabusk interact with muskwa (black bears, Ursus americanus); and that human-polar bear interactions occur in this region of northern Canada. Bearing in mind that Cree knowledge has been recognized in wildlife management strategies (i.e., for beaver, caribou and moose) elsewhere in Canada, this particular body of information is timely, especially since polar bears are considered threatened under the Ontario Endangered Species Act, and the province is developing a recovery strategy for the Southern Hudson Bay polar bear population. The federal government is also contemplating listing polar bears in Canada as a “species of special concern” under the Species at Risk Act (SARA). While it is unclear as to how these decisions will impact the Cree-polar interactions, the listing of polar bears by both governments, but especially the provincial government of Ontario, must recognize treaty and Aboriginal rights, acknowledge its duties to consult and properly accommodate Aboriginal people’s views, incorporate Cree kiskayndamowin/knowledge of wabusk, and re-examine the proposed Wabusk Co-Management Agreement draft developed by the Ontario Ministry of Natural Resources (OMNR) and Coastal Cree First Nations of Northern Ontario in 1994. The article provides recommendations that highlight how the Northern Cree First Nations, through the development of the Recovery Strategy for Polar Bear in Ontario, can become engaged in the management of wabusk in Ontario and throughout Canada.  相似文献

Morphological and chemical differences between plant genera influence phyllosphere microbial populations, but the factors driving within-species variation in phyllosphere populations are poorly understood. Twenty-six lettuce accessions were used to investigate factors controlling within-species variation in phyllosphere bacterial populations. Morphological and physiochemical characteristics of the plants were compared, and bacterial community structure and diversity were investigated using terminal restriction fragment length polymorphism (T-RFLP) profiling and 16S rRNA gene clone libraries. Plant morphology and levels of soluble carbohydrates, calcium, and phenolic compounds (which have long been associated with plant responses to biotic stress) were found to significantly influence bacterial community structure. Clone libraries from three representative accessions were found to be significantly different in terms of both sequence differences and the bacterial genera represented. All three libraries were dominated by Pseudomonas species and the Enterobacteriaceae family. Significant differences in the relative proportions of genera in the Enterobacteriaceae were detected between lettuce accessions. Two such genera (Erwinia and Enterobacter) showed significant variation between the accessions and revealed microbe-microbe interactions. We conclude that both leaf surface properties and microbial interactions are important in determining the structure and diversity of the phyllosphere bacterial community.  相似文献

Glial‐derived neurotrophic factor (GDNF) and nerve growth factor (NGF) have both been shown to enhance peripheral nerve regeneration following injury and target different neuronal populations. The delivery of either growth factor at the site of injury may, therefore, result in quantitative differences in motor nerve regeneration and functional recovery. In this study we evaluated the effect of affinity‐based delivery of GDNF or NGF from fibrin‐filled nerve guidance conduits (NGCs) on motor nerve regeneration and functional recovery in a 13 mm rat sciatic nerve defect. Seven experimental groups were evaluated consisting of GDNF or NGF and the affinity‐based delivery system (DS) within NGCs, control groups excluding the DS and/or growth factor, and nerve isografts. Groups with growth factor in the conduit demonstrated equivalent or superior performance in behavioral tests and relative muscle mass measurements compared to isografts at 12 weeks. Additionally, groups with GDNF demonstrated greater specific twitch and tetanic force production in extensor digitorum longus (EDL) muscle than the isograft control, while groups with NGF produced demonstrated similar force production compared to the isograft control. Assessment of motor axon regeneration by retrograde labeling further revealed that the number of ventral horn neurons regenerating across NGCs containing GDNF and NGF DS was similar to the isograft group and these counts were greater than the groups without growth factor. Overall, the GDNF DS group demonstrated superior functional recovery and equivalent motor nerve regeneration compared to the isograft control, suggesting it has potential as a treatment for motor nerve injury. There are over 10,000 species of parasitic protozoa, a subset of which can cause considerable disease in humans. Here we examine in detail the complex immune response generated during infection with a subset of these parasites: Trypanosoma cruzi, Leishmania sp., Toxoplasma gondii, and Plasmodium sp. While these particular species perhaps represent the most studied parasites in terms of understanding how T cells function during infection, it is clear that the lessons learned from this body of work are also relevant to the other protozoa known to induce a CD8+ T cell response. This review will highlight some of the key studies that established that CD8+ T cells play a major role in protective immunity to protozoa, the factors that promote the generation as well as maintenance of the CD8+ T cell response during these infections, and draw attention to some of the gaps in our knowledge. Moreover, the development of new tools, including MHC-Class I tetramer reagents and the use of TCR transgenic mice or genetically modified parasites, has provided a better appreciation of how parasite specific CD8+ T cell responses are initiated and new insights into their phenotypic plasticity  相似文献

Predation pressure and resource availability often interact in structuring herbivore communities, with their relative influence varying in space and time. The operation of multiple ecological pressures and guild-specific herbivore responses may combine to override simple predictions of how the roles of plant quality and predation pressure vary in space. For 2 years at the Coweeta LTER in the Southern Appalachian Mountains, we conducted a bird exclosure experiment on red oak (Quercus rubra) saplings to investigate the effects of bird predation on red oak arthropod communities. We established bird exclosures at six sites along an elevational gradient and estimated variation in foliar nitrogen and bird predation pressure along this gradient. Foliar nitrogen concentrations increased with elevation while our index of bird predation pressure was variable across sites. Greater arthropod densities were detected inside exclosures; however, this result was mainly driven by the response of phloem feeders which were much more prevalent inside exclosures than on control trees. There was little evidence for an effect of bird predation on the other arthropod guilds. Consequently, there was no evidence of a trophic cascade either in terms of leaf damage or tree growth. Finally, we found more variation in arthropod density among trees within sites than variation in arthropod density among sites, indicating the importance of micro-site variation in structuring arthropod communities.  相似文献

BACKGROUND:The COVID-19 pandemic has exacerbated disparities in poverty and illness for people in vulnerable circumstances in ethnocultural communities. We sought to understand the evolving impacts of COVID-19 on ethnocultural communities to inform intersectoral advocacy and community action.METHODS:The Illuminate Project used participatory action research, with cultural health brokers as peer researchers, from Sept. 21 to Dec. 31, 2020, in Edmonton, Alberta. Twenty-one peer researchers collected narratives from members of ethnocultural communities and self-interpreted them as they entered the narratives into the SenseMaker platform, a mixed-method data collection tool. The entire research team analyzed real-time, aggregate, quantitative and qualitative data to identify emerging thematic domains, then visualized these domains with social network analysis.RESULTS:Brokers serving diverse communities collected 773 narratives. Identified domains illuminate the evolving and entangled impacts of COVID-19 including the following: COVID-19 prevention and management; care of acute, chronic and serious illnesses other than COVID-19; maternal care; mental health and triggers of past trauma; financial insecurity; impact on children and youth and seniors; and legal concerns. We identified that community social capital and cultural brokering are key assets that facilitate access to formal health and social system supports.INTERPRETATION:The Illuminate Project has illustrated the entangled, systemic issues that result in poor health among vulnerable members of ethnocultural communities, and the exacerbating effects of COVID-19, which also increased barriers to mitigation. Cultural brokering and community social capital are key supports for people during the COVID-19 pandemic. These findings can inform policy to reduce harm and support community resiliency.

Mahatma Gandhi observed that “the true measure of any society can be found in how it treats its most vulnerable members.” Ethnocultural communities, defined by their unique shared characteristics (e.g., cultural traditions, language, country of origin),1 face greater challenges and have higher rates of poverty and illness than the general Canadian population. Migration results in conditions that affect all social determinants of health and disproportionally affect health outcomes, herein referred to as vulnerable circumstances.2,3 The emergence of major outbreaks of SARS-CoV-2 infections in ethnocultural communities highlights both the vulnerable circumstances of these communities and the disparities they face in accessing high-quality, culturally appropriate information and support.47 Studies have shown substantial variation in deaths attributed to COVID-19 based on factors such as age, sex, ethnicity, length of time in Canada, income and education.811 However, given the well-known gap in reporting comprehensive COVID-19 data in relation to race and ethnicity, efforts to measure its impact are hampered.812 There is an urgent need to understand the evolving challenges of COVID-19 to inform action and public policy that can mitigate these challenges.To understand evolving situations of complexity and crisis, sensemaking, defined as “a continuous process to establish situational awareness,”13 is a crucial undertaking.14 Using participatory action research,1518 we sought to understand the evolving impacts of COVID-19 on ethnocultural communities to inform broader national efforts to migitate the impacts of COVID-19. Particularly, we sought to understand how the challenges of COVID-19 are entangled with contextual factors at multiple levels, how families and communities are leveraging strengths and social capital to adapt, and the role of cultural brokers in managing the crisis.  相似文献
