To study the effect of acetate inhibition on the parameters of yield and maintenance for bacterial growth, Pseudomonas putida ATCC 23467 was grown in a minimal salts medium with acetate as the sole carbon source with limiting and with excess quantities of urea in the feed medium. The behavior of the chemostat cultures under sole acetate limitation results in low residual acetate present in the fermentation broth. These cultures can be described satisfactorily using the equation q(s) = D/Y(g) + m, i.e., the acetate is consumed only for growth and maintenance,. Those cultures in which urea was limiting or where urea was present in large excess contained significant amounts of residual acetate in the broth. For these cultures it was necessary to add a third term for acetate inhibition to the above expression.  相似文献

This study deals with characterizing the extracellular xylanases produced by a strain of the thermophilic bacterial genus Thermomonospora. Supernatant from centrifuged fermentation broth was used as a crude enzyme preparation. From pH 5.5 to pH 7.7 the temperature optimum based on a 10-min assay of activity was 80 degrees C. The crude enzyme had a half-life of approximately 1 month when stored at 55 degrees C at pH 6.5. The enzyme produced a mixture of xylose oligomers from xylan, with xylobiose occuring in greatest quantity on a molar basis. Only trace quantities of xylose were produced by this hydrolysis.  相似文献

Forty-two cycling, multiparous beef cows (percentage-Brahman) were given two injections of 500 ug Cloprostenol (CLP) 11 days apart. Cows were randomly allocated to be ovariectomized at 0, 12, 24, 36, 48, 60 or 72 hr after the second CLP injection. Mean CL weight declined within 36 hr after CLP. Mean concentration of P4 in luteal tissue increased between o and 60 hr, while mean P4 content per CL declined by 12 hr after CLP. There was a precipitous decrease in mean serum P4 by 12 hr following CLP injection. Serum E2 was elevated until 24 hr and then declined through 72 hr after CLP. Follicular T concentration increased from 0 to 48 hr and then decreased by 60 hr. We conclude that CLP caused rapid diminution of luteal function which was accompanied by a reduction in P4 content but not in P4 concentration. Futhermore, the concentration of E2 in large follicles decreased by 72 hr post-CLP which is consistent with an alteration of the steroidogenic pathway in the periovulatory follicle.  相似文献

Summary The fluorescence of a batch culture ofPseudomonas putida grown on phenol was investigated. A linear relationship was found between cell concentration and culture fluorescence during in the early exponential growth phase. Accumulation of a metabolite in the culture broth affects fluorescence by NADH consumption and inner filter effects. The fluorescence is also affected by changes in the metabolic activity of the cells. These effects during the course of the fermentation are discussed.  相似文献

Rhodococcus erythropolis N1-36, a desulfurization strain, was grown in continuous culture at 10 different dilution rates with 50 (mu)M dibenzothiophene sulfone (DBTO(inf2)) as the growth-limiting nutrient. The steady-state biomass, concentrations of substrate (DBTO(inf2)) and product (monohydroxybiphenyl), saturation constant (0.39 (mu)M DBTO(inf2)), and cell yield coefficient (9 mg of biomass(middot)(mu)M(sup-1) DBTO(inf2)) were measured. Continuous cultures at five temperatures allowed calculation of activation energy (0.84 kcal(middot)mol(sup-1) [ca. 3.5 kJ(middot)mol(sup-1)]) near the optimal temperature (30(deg)C) for growth. A washout technique was used to calculate the maximum specific growth rate (0.235 h(sup-1)), a value equivalent to a minimum generation time of 2.95 h.  相似文献

Aerobic and anaerobic plate counts were compared for routine monitoring of the microflora, dominated by lactic acid bacteria, developing on vacuum- and carbon dioxide-packaged raw meat during chilled storage. No statistical differences were observed between aerobic and anaerobic enumerations, made on plate count and blood agar plates, of the microflora developing on beef striploins packaged under vacuum or carbon dioxide during 14 weeks' storage at 0°C. With both techniques the spoilage microflora development differed between the two packaging regimes. The results indicate that there is no necessity for aerobic plate counts to be replaced by anaerobic plate counts in the routine microbiological examination of the spoilage microflora developing on chilled meats packaged under anoxic modified atmospheres.  相似文献

The adoptive secondary anti-DNP antibody response of primed spleen cells was specifically and lastingly inhibited by exposure in vitro to highly radioactive DNP conjugates. When the cells were treated with multivalent DNP-TGL125I, their responsiveness was almost completely inhibited, and took about 2 months to recover. The recovery was attributable to the transferred cells, since the contribution of the recipients was estimated to be small. Similarities to the recovery after tolerance are discussed.Incomplete suicide, after exposure to radioactive multivalent or monovalent DNP conjugates, affected the high- and low-affinity secondary antibody responses impartially. In a primary system, however, there was some indication that the antibody was of lower affinity after partial suicide than in control groups. The results are discussed in terms of the very high avidity of multipoint binding, and of the lower density of receptors on nonimmune B cells already postulated by Klinman.  相似文献
