The possibility that the GST1 phenotype of human liver cytosol is a determinant of bile salt binding has been investigated by using equilibrium dialysis and gel-exclusion chromatography. Binding of bile salts was non-saturable and whereas the glutathione S-transferases did not appear to be major bile salt binders, other binding components with molecular weights of 35 000 and 11 000 were identified in both fetal and adult cytosols. 相似文献

A method is described for the analysis of [3H]-thymidine incorporation in microtitre cultures of bone marrow-derived mouse macrophage responding to macrophage colony-stimulating factor (CSF-1). [3H]-thymidine incorporation depends on cell density, culture medium, and the concentration of CSF-1 and serum, but is independent of Ca2+. Bone marrow-derived macrophages are strongly adherent, but adherence can be dissociated from [3H]-thymidine incorporation. 相似文献

Sheet preparations of the stratum granulosum from the epithelium of the ventral surface of mouse tongue permit examination of cell replacement of this maturation compartment of the tissue. The cell transit rate/day is related to the cell desquamation rate and the cell production rate. The latter is approximately 6500-8000 cells/mm2/day, suggesting a 4-5-fold greater turnover compared with mouse dorsal skin epithelium. The use of [3H]IUdR and [3H]TdR at different times of day provides evidence for a reutilization of label from [3H]TdR released during nuclear degradation in the stratum granulosum. In the course of a hemorrhagic shock, pathological changes occur, which result in intensifying the insufficiency of various vital organs. It can also lead to the development of the multiorgan dysfunction syndrome (MODS) that is the cause of high posthemorrhagic mortality. As a result of the ischemia in the lung there appear proinflammatory factors that mobilize and activate mast cells, inducing degranulation in them. The aim of the study was the analysis of cellular composition and cytomorphometric evaluation of mast cells present in the lavage fluid from the pleural cavity of rats in a sham operated group and in the group presenting hemorrhagic shock. The results revealed an increase of the total cell count in the lavage fluid from the pleural cavity. In the cytological smears a statistically significant accumulation of inflammatory cells was present, especially neutrophils. The increase in mast cells and eosinophils number was not statistically significant. There was not a change in the morphometric parameters of mast cells except the circularity index. A decline of the circularity index indirectly may suggest the degranulation of mast cells, which reflects an inflammatory process in the lungs. 相似文献
