Presence of heavy metals including lead (Pb) in the textile effluents is a crucial factor affecting the growth and potential of the dye decolorizing bacterial strains. This work was planned to isolate and characterize a bacterial strain exhibiting the potential to decolorize a range of azo dyes as well as the resistance to Pb. In this study, several Pb tolerant bacteria were isolated from effluents of textile industry. These bacterial isolates were screened for their potential of decolorizing the reactive red-120 (RR120) azo dye with presence of Pb (50 mg L?1). The most efficient isolate was further characterized for its potential to resist Pb and decolorize different azo dyes under varying cultural and incubation conditions. Out of the total 82 tested bacterial isolates, 30 bacteria were found to have varying potentials to resist the presence of lead (Pb) and carry out decolorization of an azo dye reactive red-120 (RR120) in the medium amended with Pb (50 mg L?1). The most efficient selected bacterium, Pseudomonas aeruginosa strain HF5, was found to show a good potential not only to grow in the presence of considerable concentration of Pb but also to decolorize RR120 and other azo dyes in the media amended with Pb. The strain HF5 completely (>?90%) decolorized RR120 in mineral salt medium amended with 100 mg L?1 of Pb and 20 g L?1 NaCl. This strain also considerably (>?50%) decolorized RR120 up to the presence of 2000 mg L?1 of Pb and 50 g L?1 of NaCl but with reduced rate. The optimal decolorization of RR120 by HF5 was achieved when the pH of the Pb amended (100 mg L?1) mineral salt media was adjusted at 7.5 and 8.5. Interestingly, this strain also showed the tolerance to a range of metal ions with varying MIC values. The Pseudomonas aeruginosa strain HF5 harboring the unique potentials to grow and decolorize the azo dyes in the presence of Pb is envisaged as a potential bioresource for devising the remediation strategies for treatment of colored textile wastewaters loaded with Pb and other heavy metal ions. 相似文献

Phenol is a commonly found organic pollutant in industrial wastewaters. Its ecotoxicological significance is well known and, therefore, the compound is often required to be removed prior to discharge. In this study, plant-bacterial synergism was established in floating treatment wetlands (FTWs) in an attempt to maximize the removal of phenol from contaminated water. A common wetland plant, Typha domingensis, was vegetated on a floating mat and augmented with three phenol-degrading bacterial strains, Acinetobacter lwofii ACRH76, Bacillus cereus LORH97, and Pseudomonas sp. LCRH90, to develop FTWs for the remediation of water contaminated with phenol. All of the strains are known to have phenol-reducing properties, and grow well in FTWs. Results showed that T. domingensis was able to remove a small amount of phenol from the contaminated water; however, bacterial augmentation enhanced the removal potential significantly, i.e., 0.146 g/m2/day vs. 0.166 g/m2/day, respectively. Plant biomass also increased in the presence of bacterial consortia; and inoculated bacteria displayed successful colonization/survival in the rhizosphere, root interior and shoot interior of the plant. Similarly, highest reduction in chemical oxygen demand (COD), biochemical oxygen demand (BOD5), and total organic carbon (TOC) was achieved by the combined application of plants and bacteria. The study demonstrates that the plant-bacterial synergism in a FTW may be a more effective approach for the remediation of phenol-contaminated water. 相似文献

Human recombinant vascular endothelial growth factor-A121 (hrVEGF-A121) has applications in pharmaceutical industry especially in regenerative medicine. Here, we report the expression, purification, and characterization of hrVEGF-A121 in Escherichia coli expression system using human small ubiquitin-related modifier-3 (hSUMO3) fusion partner. Total RNA was isolated from healthy human gingival tissue, VEGF-A121 gene was RT-PCR amplified, and hSUMO3 gene was tagged at N-terminus. The fusion gene (SUMO3-VEGF-A121) was cloned in pET-22b(+) expression vector and transferred into E. coli strains; BL21 codon?+?and Rosetta-gami B(DE3). The hrVEGF-A121 expression was optimized for temperature, IPTG concentration, and time in Terrific Broth (TB). The positive transformants were sequenced and hrVEGF-A121 nucleotide sequence was submitted to Genbank (Accession No. KT581010). Approximately 40% of total cell protein expression was observed in soluble form on 15% SDS-PAGE. The hSUMO3 was cleaved from hrVEGF-A121 with SUMO protease and purified by Fast Protein Liquid Chromatography using anionic Hi-trap Resource Q column. From 100 ml TB, ~?25.5% and ~?6.8 mg of hrVEGF-A121 protein was recovered. The dimerized hrVEGF-A121 was characterized by Native PAGE and Western blot, using human anti-VEGF-A antibody and ESI-MS showed dimeric hrVEGF-A121 at 31,015 Da. In mature flowers of the southern Andean parasitic herb, Quinchamalium chilense (Santalaceae), the stigmas and anthers are closer together in the pin morph than in the thrum morph. While the stigmas and anthers of the two morphs are in reciprocal positions as the flowers open, such reciprocity is lost as the result of post-anthesis allometric growth of the styles and stamens. Experimental pollinations reveal that both morphs of Q. chilense are self-compatible. Natural fruit set is higher on the pin morph. The latter is also statistically under-represented in natural populations with respect to a 1:1 ratio for pins and thrums. Pin flowers produce larger numbers of pollen grains than thrum flowers and pollen of pin flowers is smaller in size than that of thrum flowers. Higher fruit set on the pin morph is consistent with some tendency towards subdioecious breeding behavior, although an expected excess of geitonogamous pollinations on the pin morph might also be contributing to the difference in fruit set on the two morphs. The unusual floral morphology of Q. chilense could have evolved as a result of selection for larger flowers by certain pollinator groups less likely to effect geitonogamous pollination. Alternatively, extended development of post-anthesis flowers might be a reflection of selection for an array of flower sizes on individual inflorescences, producing a target effect for pollinator attraction. The evolution of such inflorescence morphology would have been facilitated by the fact that a trend in the direction of subdioecious breeding behavior accrues the same outcrossing advantages as strict distyly. Precise information on periods of stigma receptivity is required to further our understanding of the floral morphology of Q. chilense. 相似文献

Alpha-1-syntrophin (SNTA1) and Rac1 are part of a signaling pathway via the dystrophin glycoprotein complex (DGC). Both SNTA1 and Rac1 proteins are over-expressed in various carcinomas. It is through the DGC signaling pathway that SNTA1 has been shown to act as a link between the extra cellular matrix, the internal cell signaling apparatus and the actin cytoskeleton. SNTA1 is involved in the modulation of the actin cytoskeleton and actin reorganization. Rac1 also controls actin cytoskeletal organization in the cell. In this study, we present the interplay between f-actin, SNTA1 and Rac1. We analyzed the effect of actin depolymerization on SNTA1 tyrosine phosphorylation and Rac1 activity using actin depolymerizing drugs, cytochalasin D and latrunculin A. Our results indicate a marked decrease in the tyrosine phosphorylation of SNTA1 upon actin depolymerization. Results suggest that actin depolymerization mediated loss of SNTA1 phosphorylation leads to loss of interaction between SNTA1 and Rac1, with a concomitant loss of Rac1 activation. The loss of SNTA1tyrosine phosphorylation and Rac1 activity by actin depolymerization results in increased apoptosis, decreased cell migration and decreased reactive oxygen species (ROS) levels in breast carcinoma cells. Collectively, our results present a possible role of f-actin in the SNTA1-Rac1 signaling pathway and implications of actin depolymerization on cell migration, ROS production and apoptosis. 相似文献

In the yttrium aluminium system, the YAlO3 phosphor is a prominent host because of the yttrium aluminium ratio (1:1). Phosphor was synthesized by the solid‐state reaction method at variable concentrations of erbium (0.1–2.5 mol%). This method is suitable for large‐scale production and is a less time‐consuming method when compared with the soft synthesis method. The prepared sample was characterized by X‐ray diffraction technique and the crystallite size was calculated by Scherer's formula. Vibrational and bending analysis of prepared phosphor for optimized concentration of erbium ion is described based on the Fourier transform infrared spectroscopic technique. The photoluminescence (PL) emission spectra of prepared phosphor for variable concentrations of erbium ion were recorded and the excitation spectrum was found to be at 291 nm with three shoulder peaks at 305, 270 and 242 nm. For 291 nm excitation, the emission spectrum was found at 546 nm and 552 nm. PL intensity increased with increasing concentrations of erbium and after 2 mol% emission intensity decreased due to concentration quenching. Spectrophotometric determination of YAlO3:Er3+ is described by CIE co‐ordinates and shows an intense emission in the green region such that the prepared phosphor can act as a single host for green light emission. Thermoluminescence glow curve analysis of the YAlO3:Er3+ phosphor was recorded for different ultraviolet (UV) light exposures and gamma exposure. Genetic variability studies for needle and wood traits were carried out for the different half sib progenies of Chir pine, raised in 1985 at the main campus of University. There existed a significant variation for these traits among the different half sib progenies, viz., needle length (18.1–24.6 cm), needle thickness (0.53–0.71 mm), number of stomata per mm of a row (7.3–12.0), specific gravity of wood (0.36–0.46), tracheid length (1.51–1.85) and moisture content of wood (47.76–58.81). This variability was found under genetic control, as all these progenies are growing under same environment, and are of same age. Traits having high heritability and genetic gain like, needle thickness, wood specific gravity, tracheid length and others, indicate high genetic control. This variability can be exploited in tree improvement programs through selection and breeding approaches for development of advanced generations. Correlation studies for different traits at genotypic and phenotypic levels provided the basic knowledge of association to chalk out efficient breeding strategy for higher productivity through indirect selection. 相似文献

pT181 is a naturally occurring Staphylococcus aureus plasmid, encoding inducible resistance to tetracycline. The plasmid has a copy number of about 20 per cell, and belongs to the incompatibility group inc3. The complete nucleotide sequence of pT181 has been determined and consists of 4437 bp. The nucleotide sequence contains 69.8% A-T and 30.2% G-C pairs. pT181 was found to contain four open reading frames capable of coding for polypeptides containing more than 50 amino acids. All the putative polypeptides are coded by one strand. The molecular weights of the four putative polypeptides are (in daltons): A, 37,500; B, 35,000; C, 23,000, and D, 18,000. Polypeptide A corresponds to the repC protein, earlier shown to be specifically required for pT181 replication. Polypeptide B (and possibly polypeptide D) are involved in tetracycline resistance. No role has yet been established for polypeptide C; deletion of the coding sequence for the C polypeptide has no detectable effect on any property of the pT181 plasmid. A region consisting of about 1200 bp contains information for the replication and copy number control of this plasmid. The sequencing results are discussed in relation to the replication properties and tetracycline resistance associated with the pT181 plasmid. 相似文献
