The histochemical and structural properties of red and white muscle from the American eel, Anguillu rostrata , are examined. The results suggest that in addition to the classic red and white fibre types, with the generalized pattern of SDH, LDH and myosin ATPase activities, the eel also has two further fibre types. Both are localized directly adjacent to the perimysium separating the red and white muscle masses: one is found only in white myotomes at the lateral line, while the other is unique to red myotomes. During the transition from immature yellow to mature bronze eels, the red muscle myofibrils become partitioned as a result of increases in mitochondria1 numbers and lipid content. It is suggested (1) the myofibrillar partitioning may reflect an altered functional role of red muscle and (2) eel white muscle may provide a portion of the muscular power required to sustain cruising speeds necessary for the long seaward migration of this animal. 相似文献

Both basal metabolic rate (BMR) and maximum lifespan potential (MLSP) vary with body size in mammals and birds and it has
been suggested that these are mediated through size-related variation in membrane fatty acid composition. Whereas the physical
properties of membrane fatty acids affect the activity of membrane proteins and, indirectly, an animal’s BMR, it is the susceptibility
of those fatty acids to peroxidation which influence MLSP. Although there is a correlation between body size and MLSP, there
is considerable MLSP variation independent of body size. For example, among bird families, Galliformes (fowl) are relatively
short-living and Psittaciformes (parrots) are unusually long-living, with some parrot species reaching maximum lifespans of
more than 100 years. We determined BMR and tissue phospholipid fatty acid composition in seven tissues from three species
of parrots with an average MLSP of 27 years and from two species of quails with an average MLSP of 5.5 years. We also characterised
mitochondrial phospholipids in two of these tissues. Neither BMR nor membrane susceptibility to peroxidation corresponded
with differences in MLSP among the birds we measured. We did find that (1) all birds had lower n-3 polyunsaturated fatty acid
content in mitochondrial membranes compared to those of the corresponding tissue, and that (2) irrespective of reliance on
flight for locomotion, both pectoral and leg muscle had an almost identical membrane fatty acid composition in all birds. 相似文献

The gene Lr46 has provided slow-rusting resistance to leaf rust caused by Puccinia triticina in wheat (Triticum aestivum), which has remained durable for almost 30 years. Using linked markers and wheat deletion stocks, we located Lr46 in the deletion bin 1BL (0.84–0.89) comprising 5% of the 1BL arm. The distal part of chromosome 1BL of wheat is syntenic
to chromosome 5L of rice. Wheat expressed sequence tags (ESTs) mapping in the terminal 15% of chromosome 1BL with significant
homology to sequences from the terminal region of chromosome 5L of rice were chosen for sequence-tagged site (STS) primer
design and were mapped physically and genetically. In addition, sequences from two rice bacterial artificial chromosome clones
covering the targeted syntenic region were used to identify additional linked wheat ESTs. Fourteen new markers potentially
linked to Lr46 were developed; eight were mapped in a segregating population. Markers flanking (2.2 cM proximal and 2.2 cM distal) and cosegregating
with Lr46 were identified. The physical location of Lr46 was narrowed to a submicroscopic region between the breakpoints of deletion lines 1BL-13 [fraction length (FL)=0.89–1] and

Data concerning the acyl composition of tissue phospholipids from mammal species, ranging in size from the shrew (7 g) to cattle (370 kg), has been collated from the literature and analysed allometrically. Phospholipids from heart, skeletal muscle, liver and kidney exhibited similar allometric trends whereby phospholipids had a significant decrease in unsaturation index (number of double bonds per 100 acyl chains) as species body size increased whilst there was no change in the percent of unsaturated acyl chains. Whilst total polyunsaturate content did not change with body mass, both heart and skeletal muscle phospholipids showed a significant allometric decrease in the omega-3 polyunsaturate content. The content of the highly polyunsaturated docosahexaenoic acid (22:6 n-3) in phospholipids showed significant and substantial allometric decline with increasing body mass in all four tissues (exponents ranged from -0.19 in liver to -0.40 in skeletal muscle). Brain phospholipids showed no allometric trends in acyl composition and were highly polyunsaturated in all species. These trends are discussed in light of the hypothesis that the relative content of polyunsaturated acyl chains in membranes, and especially docosahexaenoate (22:6 n-3), can act as a membrane pacemaker for metabolic activity. 相似文献

The chromosome 1R of rye, or the midget chromosome, is necessary for plump, viable seed development and fertility restoration in the alloplasmic line with rye cytoplasm and a hexaploid wheat nucleus. The midget chromosome of rye represents 1/15th of the physical length of the chromosome 1R of rye. C-banding analysis indicated that the centromeric and pericentric region (approximately 30% physical length) of the midget chromosome is heterochromatic and the distant 70% physical length is euchromatic. These data suggest that the midget chromosome may represent the pericentric region of the long arm of chromosome 1R. In contrast with earlier reports, our results indicate that an array of rye-specific repeated sequences (both dispersed and tandem) are present on the midget chromosome. Various rye-specific repeated DNA sequences that are present on the midget chromosome will be useful in constructing a long-range map and studying the genomic organization of the midget chromosome. It is unclear if any of these repeated DNA sequences are involved in the origin of the midget chromosome. 相似文献

The concept of basal metabolic rate (BMR) was developed to compare the metabolic rate of animals and initially was important in a clinical context as a means of determining thyroid status of humans. It was also important in defining the allometric relationship between body mass and metabolic rate of mammals. The BMR of mammals varies with body mass, with the same allometric exponent as field metabolic rate and with many physiological and biochemical rates. The membrane pacemaker theory proposes that the fatty acid composition of membrane bilayers is an important determinant of a species BMR. In both mammals and birds, membrane polyunsaturation decreases and monounsaturation increases with increasing body mass and a decrease in mass-specific BMR. The secretion and production of thyroid hormones in mammals are related to body mass, with the allometric exponent similar to BMR; yet there is no body size-related variation in either total or free concentrations of thyroid hormones in plasma of mammals. It is suggested that in different-sized mammals, the secretion/production of thyroid hormones is a result of BMR differences rather than their cause. Plant-feeding insects have been recently found to use microbes to manipulate host plant physiology and morphology. Gall midges are one of the largest groups of insects that manipulate host plants extensively. Hessian fly (HF, Mayetiola destructor) is an important pest of wheat and a model system for studying gall midges. To examine the role of bacteria in parasitism, a systematic analysis of bacteria associated with HF was performed for the first time. Diverse bacteria were found in different developmental HF stages. Fluorescent in situ hybridization detected a bacteriocyte-like structure in developing eggs. Bacterial DNA was also detected in eggs by PCR using primers targeted to different bacterial groups. These results indicated that HF hosted different types of bacteria that were maternally transmitted to the next generation. Eliminating bacteria from the insect with antibiotics resulted in high mortality of HF larvae, indicating that symbiotic bacteria are essential for the insect to survive on wheat seedlings. A preliminary survey identified various types of bacteria associated with different HF stages, including the genera Enterobacter, Pantoea, Stenotrophomonas, Pseudomonas, Bacillus, Ochrobactrum, Acinetobacter, Alcaligenes, Nitrosomonas, Arcanobacterium, Microbacterium, Paenibacillus, and Klebsiella. Similar bacteria were also found specifically in HF-infested susceptible wheat, suggesting that HF larvae had either transmitted bacteria into plant tissue or brought secondary infection of bacteria to the wheat host.
