A xylanase gene, xynE2, was cloned from thermoalkaline Anoxybacillus sp. E2 and was expressed in Escherichia coli BL21 (DE3). The gene consisted of 987 bp and encoded a 328-residue xylanase with a calculated molecular weight of 38.8 kDa.
On the basis of amino acid sequence similarities, this enzyme was assigned as a member of glycoside hydrolase family 10. Purified
recombinant XynE2 showed maximal activity at pH 7.8 and 65°C, and was thermostable at 60°C. The enzyme was highly active and
stable over a broad pH range, showing more than 90% of maximal activity at pH 6.6–pH 8.6 and retaining more than 80% of activity
at pH 4.6–pH 12.0, 37°C for 1 h, respectively. These favorable properties make XynE2 a good candidate in the pulp and paper

Common bean (Phaseolus vulgaris) symbiotically associates with its partner Rhizobium etli, resulting in the formation of root nitrogen-fixing nodules. Compatible bacteria can reach cortical cells in a tightly regulated infection process, in which the specific recognition of signal molecules is a key step to select the symbiotic partner. In this work, we show that RabA2, a monomeric GTPase from common bean, is required for the progression of the infection canal, referred to as the infection thread (IT), toward the cortical cells. Expression of miss-regulated mutant variants of RabA2 resulted in an increased number of abortive infection events, including bursting of ITs and a reduction in the number of nodules. Nodules formed in these plants were small and contained infected cells with disrupted symbiosome membranes, indicating either early senescence of these cells or defects in the formation of the symbiosome membrane during bacterial release. RabA2 localized to mobile vesicles around the IT, but mutations that affect GTP hydrolysis or GTP/GDP exchange modified this localization. Colocalization of RabA2 with ArfA1 and a Golgi marker indicates that RabA2 localizes in Golgi stacks and the trans-Golgi network. Our results suggest that RabA2 is part of the vesicle transport events required to maintain the integrity of the membrane during IT progression.465.
达乌尔黄鼠(Spermophilusdauricus)是国内研究冬眠生理学的主要模式动物,且日益为医学研究所关注。实验室条件下繁殖种群具有明确生活史特征,是相关研究的理想对象,但以往研究中,达乌尔黄鼠实验室条件下繁殖成功率及后代成活率均很低。本研究通过采取丰富饲料种类、雄性出眠后的暖温暂养、“双笼配对”等措施,对实验室条件下越冬达乌尔黄鼠进行配对,从21对动物中成功繁殖出18窝子代,繁殖成功率85.7 %。78只子代饲养至3月龄,仍有67只存活,存活率为85.9 %。通过本实验,得出改进达乌尔黄鼠实验室条件下繁殖的主要措施如下:(1)繁殖期母鼠及发育期幼鼠,除饲喂标准大鼠饲料外,适量添加幼犬粮、胡萝卜,以补充营养;(2)出眠的雄性达乌尔黄鼠放入18℃房间暂养2周,有助于其性腺发育,促进繁殖成功;(3)“双笼配对”能有效地降低外界干扰,减少动物的杀婴行为。

Heart failure(HF) is a major clinical concern owing to its high prevalence and high mortality. Metabolomics, an effective approach to predict diagnostic biomarkers and to explore the altered metabolic pathways in pathogenesis, has been extensively applied in evaluating the course of diseases. In this study, we used this approach to analyse the abundance of metabolites, with liquid chromatograph-mass spectrometer, in plasma samples from rats with transverse aortic constriction(TAC) and patients at different stages of HF. We compared the metabolic parameters within and between TAC rats and patients. An apparent metabolic shift was observed in rats, from compensated hypertrophy stage to decompensated hypertrophy stage, and in patients with HF,from stage A to stage B and subsequently stage C. Diagnostic biomarkers were predicted by comparing the variable importance in the projection scores and fold change analysis within and between rats and patients. Enrichment pathway analysis and network analysis provided an overview of the largely disturbed metabolic pathways, and those interfered at different stages and across species were confirmed. The significantly changed metabolites and pathways revealed the underlying mechanisms of HF pathogenesis, hinted at novel potential biomarkers, and provided potential therapeutic intervention targets for HF. 相似文献

Plant growth depressions in arbuscular mycorrhizal symbioses: not just caused by carbon drain? 总被引:1,自引:0,他引:1
* This study investigated effects of plant density and arbuscular mycorrhizal (AM) colonization on growth and phosphorus (P) nutrition of a cultivar of wheat (Triticum aestivum) that often shows early AM-induced growth depressions. * Two experiments were conducted. Expt 1 had three plant densities and one soil P concentration. Expt 2 had two plant densities and two P concentrations. Plants were grown in calcareous P-fixing soil, inoculated with Glomus intraradices or Gigaspora margarita, or noninoculated (nonmycorrhizal (NM)). Glomus intraradices colonized well and caused a growth depression only in Expt 1. Gigaspora margarita caused large growth depressions in both experiments even though it colonized poorly. * The results showed that growth depressions were mitigated by changes in relative competition for soil P by NM and AM plants, and probably by decreasing carbon costs of the fungi. * The different effects of the two fungi appear to be attributable to differences in the balance between P uptake by the fungal pathway and direct uptake via the roots. These differences may be important in other AM symbioses that result in growth depressions. The results show that mycorrhizal growth responses of plants grown singly may not apply at the population or community level. 相似文献

本文报道一种适合疫苗生产的大规模纯化Vero细胞乙脑疫苗的方法。原疫苗经适当浓缩和去除DNA处理后,用不连续蔗糖梯度(36%和60%)。32600g,速率区带离心4h。纯化后疫苗的效力比中国参考疫苗高出6倍以上,补体结合抗原比中国参考疫苗高4~8倍。总蛋白含量低于30μg/mL,牛血清含量降至0.5μg/mL以下,细胞残余DNA低于100pg/0.5mL。用此法连续制备三批纯化疫苗,其纯度和效力均高于日本鼠脑纯化疫苗。此法对于制备其它纯化Vero细胞疫苗也具有一定的参考意义。 相似文献

copper, zinc, iron, and other nine trace elements (TEs) in 30 patients with hepatocellular carcinoma (HCC) were determined from the HCC itself and the surrounding liver parenchyma (SLP) using particle induced X-ray emission (PIXE). The result showed that the level of Zn, Fe, Co, Ca, Mn, Rb, and Cl in HCC significantly differed from that in the SLP (P<0.01), while no notable change has been observed in content of Cu (P=0.9). However, in relation to tumor size, an imbalance Cu distribution was observed. The content of Cu present in small HCC (<35 mm in average diameter) was greater than that in SLP (P<0.4), and when HCC progressed to large one (>35 mm in average diameter), hepatic Cu was no longer accumulated. Other elements such as Zn and Fe, showed no change in concentration relative to tumor size. The significance of the critical HCC size where a change from accumulation to loss in Cu contents is seen is also discussed. 相似文献
