The human pituitary tumor-transforming gene is an oncogenic protein which serves as a central hub in the cellular signaling network of medulloblastoma. The protein contains two vicinal PxxP motifs at its C terminus that are potential binding sites of peptide-recognition SH3 domains. Here, a synthetic protocol that integrated in silico analysis and in vitro assay was described to identify the SH3-binding partners of pituitary tumor-transforming gene in the gene expression profile of medulloblastoma. In the procedure, a variety of structurally diverse, non-redundant SH3 domains with high gene expression in medulloblastoma were compiled, and their three-dimensional structures were either manually retrieved from the protein data bank database or computationally modeled through bioinformatics technique. The binding capability of these domains towards the two PxxP-containing peptides m1p: 161LGPPSPVK168 and m2p: 168KMPSPPWE175 of pituitary tumor-transforming gene were ranked by structure-based scoring and fluorescence-based assay. Consequently, a number of SH3 domains, including MAP3K and PI3K, were found to have moderate or high affinity for m1p and/or m2p. Interestingly, the two overlapping peptides exhibits a distinct binding profile to these identified domain partners, suggesting that the binding selectivity of m1p and m2p is optimized across the medulloblastoma expression spectrum by competing for domain candidates. In addition, two redesigned versions of m1p peptide ware obtained via a structure-based rational mutation approach, which exhibited an increased affinity for the domain as compared to native peptide.  相似文献

A bifunctional hybrid electrolyte composed of mesoporous silica nanosheets and liquid electrolyte is achieved for lithium–sulfur (Li–S) batteries. This hybrid electrolyte possesses abundant mesopores (2.8 nm), thin feature (20 µm), and high ionic conductivity (1.17 × 10?1 mS cm?1) as well as a low interfacial resistance with electrodes. Such unique features not only enable the efficient inhibition of the growth of lithium dendrites, but also significantly prevents polysulfide shuttling. Consequently, a Li–S battery with this hybrid electrolyte exhibits a relatively high reversible capacity and good capacity retention.  相似文献

Polymer‐based dielectrics are attracting increasing attention due to their high‐density energy storage. However, mitigating the heat generation in real capacitors has been a challenge. Here an electrothermal breakdown phase‐field model is developed to fundamentally understand the thermal effects on the dielectric breakdown of polymer‐based dielectrics in real capacitor configurations including the increase in the dielectric loss and the decrease in the breakdown strength. While both enhancing the thermal conductivity and reducing the electrical conductivity of the polymer nanocomposites can reduce the thermal effects, it is found that reducing the electrical conductivity is more effective. This work is expected to not only stimulate attention to the thermal effects in polymer‐based dielectrics but also provide a fundamental guidance to mitigate the heat‐induced deterioration of breakdown strength.  相似文献

为了促进亮氨酸脱氢酶在Bacillus subtilis中高效表达,采取在质粒pMA5自带的启动子P_(HpaⅡ)之后分别添加诱导型和组成型启动子,考察双启动子对酶表达的影响。选取2种诱导型启动子(P_(grac)、P_(gl-M1))和4种组成型启动子(P_(43)、P_(laps)、P_(HpaⅡ)、P_(amyQ))进行构建表达,其中组成型启动子P_(amyQ)与P_(HpaⅡ)构成的双启动子效果最好,有效地将胞外活性提高到31. 24U/ml,是单启动子PHpaⅡ的3. 4倍。在以上最优双启动子的基础上分别融合Sec途径和Tat途径的4种信号肽,但信号肽与双启动子共同作用并没有获得更高的酶活性。选用酶活最高的双启动子突变菌株Bacillus subtilis168/pW6(P_(HpaⅡ)-P_(amyQ))进行7. 5L发酵罐补料发酵产酶研究,LeuDH酶活达到217. 96U/ml,是摇瓶水平的6. 97倍,对工业上产亮氨酸脱氢酶有一定参考价值。  相似文献

Effective treatment of rheumatoid arthritis can be mediated by native chicken type II collagen (nCCII), recombinant peptide containing nCCII tolerogenic epitopes (CTEs), or a therapeutic DNA vaccine encoding the full-length CCOL2A1 cDNA. As recombinant CCII (rCCII) might avoid potential pathogenic virus contamination during nCCII preparation or chromosomal integration and oncogene activation associated with DNA vaccines, here we evaluated the importance of propeptide and telopeptide domains on rCCII triple helix molecular assembly. We constructed pC- and pN-procollagen (without N- or C-propeptides, respectively) as well as CTEs located in the triple helical domain lacking both propeptides and telopeptides, and expressed these in yeast Pichia pastoris host strain GS115 (his4, Mut+) simultaneously with recombinant chicken prolyl-4-hydroxylase α and β subunits. Both pC- and pN-procollagen monomers accumulated inside P. pastoris cells, whereas CTE was assembled into homotrimers with stable conformation and secreted into the supernatants, suggesting that the large molecular weight pC-or pN-procollagens were retained within the endoplasmic reticulum whereas the smaller CTEs proceeded through the secretory pathway. Furthermore, resulting recombinant chicken type II collagen pCα1(II) can induced collagen-induced arthritis (CIA) rat model, which seems to be as effective as the current standard nCCII. Notably, protease digestion assays showed that rCCII could assemble in the absence of C- and N-propeptides or telopeptides. These findings provide new insights into the minimal structural requirements for rCCII expression and folding.  相似文献

Pentatricopeptide repeat (PPR) gene family plays an essential role in the regulation of plant growth and organelle gene expression. Some PPR genes are related to fertility restoration in plant, but there is no detailed information in Gossypium. In the present study, we identified 482 and 433 PPR homologues in Gossypium raimondii (\(\hbox {D}_{5}\)) and G. arboreum (\(\hbox {A}_{2}\)) genomes, respectively. Most PPR homologues showed an even distribution on the whole chromosomes. Given an evolutionary analysis to PPR genes from G. raimondii (\(\hbox {D}_{5}\)), G. arboreum (\(\hbox {A}_{2}\)) and G. hirsutum genomes, eight PPR genes were clustered together with restoring genes of other species. Most cotton PPR genes were qualified with no intron, high proportion of \(\upalpha \)-helix and classical tertiary structure of PPR protein. Based on bioinformatics analyses, eight PPR genes were targeted in mitochondrion, encoding typical P subfamily protein with protein binding activity and organelle RNA metabolism in function. Further verified by RNA-seq and quantitative real-time PCR (qRT-PCR) analyses, two PPR candidate genes, Gorai.005G0470 (\(\hbox {D}_{5}\)) and Cotton_A_08373 (\(\hbox {A}_{2}\)), were upregulated in fertile line than sterile line. Aims

Soil fungal pathogens can result in the failure of seedling establishment, but the effects of fungicide applications on seed/seedling survival have differed among studies. We assumed that the variation may relate to seed dormancy/germination characteristics and hypothesized that nondormant germinating seeds are more likely to be killed by fungal pathogens than dormant seeds.

Methods

Dormant and nondormant seeds of Stipa bungeana and Lespedeza davurica were inoculated with a pathogenic fungus Fusarium tricinctum under laboratory and field conditions. The outcomes of seed/seedling fate and other parameters were evaluated.

Results

In the laboratory, nondormant seeds inoculated with F. tricinctum developed white tufts of mycelium on the radicles of germinating seeds causing them to quickly die, but dormant seeds remained intact. In contrast, in the field inoculation with F. tricinctum did not cause higher mortality of nondormant than dormant seeds but resulted in higher percentages of seedling death before they emerged from soil than the controls.

Conclusions

Our results suggest that dormancy protects seeds from being attacked by some pathogens by preventing germination, but the protection is lost once germination has commenced. Further study involving various plant species with more seeds is needed to assess the generality of this pathogen-seed interaction hypothesis.
