根据已克隆的刺五加鲨烯合酶(squalene synthase,SS)、鲨烯环氧酶(squalene epoxidase,SE)和β-香树酯醇合成酶(β-amyrin synthase,bAS)基因序列信息设计引物,通过半定量RT-PCR分析了SS、SE和bAS基因在刺五加不同生长发育时期和不同器官中表达量的变化.结果表明,SS、SE和bAS基因在各生长发育时期和各器官中均有表达,但表达量差异显著(P<0.05),三者均在盛花期表达量最高,之后降低,进入果实成熟期后SS和bAS的表达量迅速回升,SE无显著变化.SS和bAS在叶片和根中的表达量较高,SE表达量的最大值出现在叶片和幼茎中.刺五加SS、SE和bAS基因的表达间存在显著的正相关关系(P<0.05).研究结果为进一步分析关键酶基因对刺五加三萜皂苷生物合成的影响奠定了基础. 相似文献

Increasing evidence shows that protein phosphorylation on serine (Ser), threonine (Thr) and tyrosine (Tyr) residues is a major regulatory post-translational modification in the bacteria. To reveal the phosphorylation state in the Gram-negative pathogenic bacterium Helicobacter pylori, we carried out a global and site-specific phosphoproteomic analysis based on TiO(2) -phosphopeptide enrichment and high-accuracy LC-MS/MS determination. Eighty-two phosphopeptides from 67 proteins were identified with 126 phosphorylation sites, among which 79 class I sites were determined to have a distribution of 42.8:38.7:18.5% for the Ser/Thr/Tyr phosphorylation, respectively. The H. pylori phosphoproteome is characterized by comparably big size, high ratio of Tyr phosphorylation, high abundance of multiple phosphorylation sites in individual phosphopeptides and over-representation of membrane proteins. An interaction network covering 28 phosphoproteins was constructed with a total of 163 proteins centering on the major H. pylori virulence factor VacA, indicating that protein phosphorylation in H. pylori may be delicately controlled to regulate many aspects of the metabolic pathways and bacterial virulence. 相似文献

Antiretroviral protease inhibitors significantly potentiated the sensitivity of chloroquine-resistant malaria parasites to the antimalarial drug in vitro and in vivo. Ritonavir was found to be potent in potentiating CQ antimalarial activities in both -resistant and -sensitive lines. The mechanism by which the APIs modulate the CQ resistance in malaria parasites was further investigated. CQ-resistant parasites showed increased intracellular glutathione levels in comparison with the CQ-sensitive parasites. Treatment with APIs significantly reduced the levels of GSH and glutathione S-transferase activities in CQ-resistant parasites. Ritonavir also decreased glutathione reductase activities and glutathione peroxidase activities in CQ-resistant parasite line. Taken together, these results demonstrate that parasite GSH and GST may play an important role in CQ resistance and APIs are able to enhance the sensitivity of CQ-resistant malaria parasite to the drug by influencing the levels of GSH and the activities of the related enzymes. 相似文献

Neuropeptide FF (NPFF) belongs to a neuropeptide family including two precursors (pro-NPFFA and pro-NPFFB) and two receptors (NPFF1 and NPFF2). Very recently, the novel compound RF9 was reported as the truly selective antagonist on NPFF receptors. The present study examined the effects of RF9 on the hypothermia and anti-morphine action induced by NPFF in mice. (1) RF9 injected into the third ventricle was devoid of any residual agonist activity, but it completely antagonized the hypothermic effects of NPFF (30 or 45 nmol) after cerebral administration in mice; (2) RF9 did not alter the tail-flick latency and morphine analgesia in nociceptive test, however, co-administration of RF9 prevented the anti-morphine action of intracerebroventricularly applied NPFF (10 nmol, i.c.v.) in the mouse tail-flick assay. Collectively, our data indicate that RF9, behaving as a truly pure NPFF receptors antagonist, prevents NPFF-induced drops of the body temperature and morphine analgesia in mice. In addition, it further confirms that the hypothermia and anti-morphine action of NPFF are mediated directly by NPFF receptors. 相似文献

本文主要应用单细胞培养技术培育紫菜新品系,以建立紫菜细胞工程快速育苗系统。通过酶解技术分离出紫菜叶状体细胞,并进行多克隆,从中获得4个细胞纯系植株(HA,HB,HC,HD)。对该4个细胞株系进行了纯系培养,并对其细胞苗和丝状体的生长速度和抗高温(19℃,21℃,23℃,25℃)性进行了测定。结果显示,船株系具有较高的抗高温性和最快生长速度。在1998年到2000年间,在江苏省启东县海丰海区对HB株系进行了海上栽培实验,结果显示,实验组的产量高于当地对照组产量,证明了条斑紫菜船株系不仅具有较高的抗高温性,而且也有较快生长速度。本实验说明应用细胞工程进行条斑紫菜育种是一条很好的快速育种途径。 相似文献

A genetic linkage map of rice was constructed using a double haploid (DH) population from "Gui 630” (Oryza sativa subsp, indica)/"02428" (O. sativa subsp, japonica, wide compatibility variety) and RFLP markers. It consists of 233 loci and covers rice genomes about 2070 cM (centimorgan), and compares well with the other published rice maps. 25 RFLP markers, 2 telomeres and sh-2 (shattering ability) gene were first located on the molecular map of rice. RFLPs between "Gui 630' and "02428' mainly came from base substitution and a few DNA construction variance, not distributed evenly among chromosomes and on chromosome. This was probably resulted from the difference genetic stability among chromosomes and regions, in exchanging recombination ability in different segments of chromosome. 相似文献

将已克隆的棉花杜松烯合成酶的cDNA(cadC14)插入到植物表达载体pBI121中,构建含CaMV35S启动子驱动下的杜松烯合成酶基因的植物表达载体pBIC14。用含pBIC14质粒的发根农杆菌(Agrobacteriumrhizogenes)15834感染青蒿(ArtemisiaannuaL.)叶片并诱导发根,共建立121个生长迅速的发根系。经浓度为20mg/L的Kan筛选,获得12个抗Kan阳性根系。PCR和Southernbloting分析表明,外源杜松烯合成酶基因已整合到青蒿基因组中,其转基因频率为3%。RTPCR分析表明,外源杜松烯合成酶基因在C37根系中,在转录水平上已有表达。 相似文献
