The recently discovered Type VII/Esat-6 secretion systems seem to be widespread among bacteria of the phyla Actinobacteria and Firmicutes. In some species they play an important role in pathogenic interactions with eukaryotic hosts. Several studies have predicted that the locus yukEDCByueBC of the non-pathogenic, Gram-positive bacterium Bacillus subtilis would encode an Esat-6-like secretion system (Ess). We provide here for the first time evidences for the functioning of this secretion pathway in an undomesticated B. subtilis strain. We show that YukE, a small protein with the typical features of the secretion substrates from the WXG100 superfamily is actively secreted to culture media. YukE secretion depends on intact yukDCByueBC genes, whose products share sequence or structural homology with known components of the S. aureus Ess. Biochemical characterization of YukE indicates that it exists as a dimer both in vitro and in vivo. We also show that the B. subtilis Ess essentially operates in late stationary growth phase in absolute dependence of phosphorylated DegU, the response regulator of the two-component system DegS-DegU. Motor sequence learning is known to rely on more than a single process. As the skill develops with practice, two different representations of the sequence are formed: a goal representation built under spatial allocentric coordinates and a movement representation mediated through egocentric motor coordinates. This study aimed to explore the influence of daytime sleep (nap) on consolidation of these two representations. Through the manipulation of an explicit finger sequence learning task and a transfer protocol, we show that both allocentric (spatial) and egocentric (motor) representations of the sequence can be isolated after initial training. Our results also demonstrate that nap favors the emergence of offline gains in performance for the allocentric, but not the egocentric representation, even after accounting for fatigue effects. Furthermore, sleep-dependent gains in performance observed for the allocentric representation are correlated with spindle density during non-rapid eye movement (NREM) sleep of the post-training nap. In contrast, performance on the egocentric representation is only maintained, but not improved, regardless of the sleep/wake condition. These results suggest that motor sequence memory acquisition and consolidation involve distinct mechanisms that rely on sleep (and specifically, spindle) or simple passage of time, depending respectively on whether the sequence is performed under allocentric or egocentric coordinates. 相似文献

Background
The posterior parietal cortex (PPC) is thought to interact with the medial temporal lobe (MTL) to support spatial cognition and topographical memory. While the response of medial temporal lobe regions to topographical stimuli has been intensively studied, much less research has focused on the role of PPC and its functional connectivity with the medial temporal lobe.Methodology/Principle Findings
Here we report a dissociation between dorsal and ventral regions of PPC in response to different types of change in natural scenes using an fMRI adaptation paradigm. During scanning subjects performed an incidental target detection task whilst viewing trial unique sequentially presented pairs of natural scenes, each containing a single prominent object. We observed a dissociation between the superior parietal gyrus and the angular gyrus, with the former showing greater sensitivity to spatial change, and the latter showing greater sensitivity to scene novelty. In addition, we observed that the parahippocampal cortex has increased functional connectivity with the angular gyrus, but not superior parietal gyrus, when subjects view change to the scene content.Conclusions/Significance
Our findings provide support for proposed dissociations between dorsal and ventral regions of PPC and suggest that the dorsal PPC may support the spatial coding of the visual environment even when this information is incidental to the task at hand. Further, through revealing the differential functional interactions of the SPG and AG with the MTL our results help advance our understanding of how the MTL and PPC cooperate to update representations of the world around us. 相似文献994.

Mononuclear Mo-containing enzymes of the xanthine oxidase (XO) family catalyze the oxidative hydroxylation of aldehydes and heterocyclic compounds. The molybdenum active site shows a distorted square-pyramidal geometry in which two ligands, a hydroxyl/water molecule (the catalytic labile site) and a sulfido ligand, have been shown to be essential for catalysis. The XO family member aldehyde oxidoreductase from Desulfovibrio gigas (DgAOR) is an exception as presents in its catalytically competent form an equatorial oxo ligand instead of the sulfido ligand. Despite this structural difference, inactive samples of DgAOR can be activated upon incubation with dithionite plus sulfide, a procedure similar to that used for activation of desulfo-XO. The fact that DgAOR does not need a sulfido ligand for catalysis indicates that the process leading to the activation of inactive DgAOR samples is different to that of desulfo-XO. We now report a combined kinetic and X-ray crystallographic study to unveil the enzyme modification responsible for the inactivation and the chemistry that occurs at the Mo site when DgAOR is activated. In contrast to XO, which is activated by resulfuration of the Mo site, DgAOR activation/inactivation is governed by the oxidation state of the dithiolene moiety of the pyranopterin cofactor, which demonstrates the non-innocent behavior of the pyranopterin in enzyme activity. We also showed that DgAOR incubation with dithionite plus sulfide in the presence of dioxygen produces hydrogen peroxide not associated with the enzyme activation. Objective
We studied whether the change in antibiotic susceptibility testing (AST) guidelines from CLSI to EUCAST influenced cumulative antibiograms in a tertiary care hospital in Switzerland.Methods
Antibiotic susceptibilities of non-duplicate isolates collected within a one-year period before (period A) and after (period B) changing AST interpretation from CLSI 2009 to EUCAST 1.3 (2011) guidelines were analysed. In addition, period B isolates were reinterpreted according to the CLSI 2009, CLSI 2013 and EUCAST 3.1 (2013) guidelines.Results
The majority of species/drug combinations showed no differences in susceptibility rates comparing periods A and B. However, in some gram-negative bacilli, decreased susceptibility rates were observed when comparing CLSI 2009 with EUCAST 1.3 within period B: Escherichia coli / cefepime, 95.8% (CLSI 2009) vs. 93.1% (EUCAST 1.3), P=0.005; Enterobacter cloacae / cefepime, 97.0 (CLSI 2009) vs. 90.5% (EUCAST 1.3), P=0.012; Pseudomonas aeruginosa / meropenem, 88.1% (CLSI 2009) vs. 78.3% (EUCAST 1.3), P=0.002. These differences were still evident when comparing susceptibility rates according to the CLSI 2013 guideline with EUCAST 3.1 guideline. For P. aeruginosa and imipenem, a trend towards a lower antibiotic susceptibility rate in ICUs compared to general wards turned into a significant difference after the change to EUCAST: 87.9% vs. 79.8%, P=0.08 (CLSI 2009) and 86.3% vs. 76.8%, P=0.048 (EUCAST 1.3).Conclusions
The change of AST guidelines from CLSI to EUCAST led to a clinically relevant decrease of susceptibility rates in cumulative antibiograms for defined species/drug combinations, particularly in those with considerable differences in clinical susceptibility breakpoints between the two guidelines. 相似文献996.

Cinnamic acid and its hydroxylated derivatives (p-coumaric, caffeic, ferulic and sinapic acids) are known allelochemicals that affect the seed germination and root growth of many plant species. Recent studies have indicated that the reduction of root growth by these allelochemicals is associated with premature cell wall lignification. We hypothesized that an influx of these compounds into the phenylpropanoid pathway increases the lignin monomer content and reduces the root growth. To confirm this hypothesis, we evaluated the effects of cinnamic, p-coumaric, caffeic, ferulic and sinapic acids on soybean root growth, lignin and the composition of p-hydroxyphenyl (H), guaiacyl (G) and syringyl (S) monomers. To this end, three-day-old seedlings were cultivated in nutrient solution with or without allelochemical (or selective enzymatic inhibitors of the phenylpropanoid pathway) in a growth chamber for 24 h. In general, the results showed that 1) cinnamic, p-coumaric, caffeic and ferulic acids reduced root growth and increased lignin content; 2) cinnamic and p-coumaric acids increased p-hydroxyphenyl (H) monomer content, whereas p-coumaric, caffeic and ferulic acids increased guaiacyl (G) content, and sinapic acid increased sinapyl (S) content; 3) when applied in conjunction with piperonylic acid (PIP, an inhibitor of the cinnamate 4-hydroxylase, C4H), cinnamic acid reduced H, G and S contents; and 4) when applied in conjunction with 3,4-(methylenedioxy)cinnamic acid (MDCA, an inhibitor of the 4-coumarate:CoA ligase, 4CL), p-coumaric acid reduced H, G and S contents, whereas caffeic, ferulic and sinapic acids reduced G and S contents. These results confirm our hypothesis that exogenously applied allelochemicals are channeled into the phenylpropanoid pathway causing excessive production of lignin and its main monomers. Rio Grande water is intensively managed and regulated by international and interstate compacts, Native American treaties, local water rights, and federal, state, and local agencies. Legislation and engineering projects in the early twentieth century brought about water impoundment projects and channelization of the Rio Grande which led to the eventual loss of floodplain habitats. In particular, current water management practices in the Middle Rio Grande (MRG) have altered the natural flood regime altering the riparian community and floodplain dynamics which may be causing the demise of many fish species by altering food web processes. The Rio Grande silvery minnow (Hybognathus amarus), a federally endangered species, has been classified as an herbivore, detritivore, or carnivore. During low flow conditions H. amarus is primarily an algivore; however, during flood conditions, hydrodynamic scouring reduces or eliminates benthic algal food sources. The objective of this study was to identify and characterize food resources and trophic interactions for H. amarus on a restored floodplain during an extended flood-pulse release from reservoirs using stable isotope analyses (δ13C and δ15N) and paleolimnology techniques. Results from stable isotope ratios indicate that H. amarus obtained carbon primarily from chironomids while aquatic invertebrates (including chironomids) obtained their carbon from macrophytes. Results from the GLIMMIX procedure indicate that the range of isotopic signatures for prey items was much broader at parallel habitats (i.e. floodplain flow parallel to main stem flow) than perpendicular (i.e. floodplain flow perpendicular to main stem flow) or leeward habitats (i.e. leeward sides of island where flow was near zero) indicating a wider selection of food resources. This study suggests that increased duration of floodplain inundation in the MRG provides vital habitats for spawning, nursery, and recruitment of threatened and endangered fish species. A combination of allochthonous and autochthonous resources best describes the nutrient and energy transfers for the Los Lunas, NM restored floodplain. 相似文献

Human exposure to electromagnetic fields produced by two wearable antennas operating in the 2.4 GHz frequency band was assessed by computational tools. Both antennas were designed to be attached to the skin, but they were intended for different applications. The first antenna was designed for off-body applications, i.e. to communicate with a device placed outside the body, while the second antenna model was optimized to communicate with a device located inside the body. The power absorption in human tissues was determined at several locations of adult male and female body models. The maximum specific absorption rate (SAR) value obtained with the off-body antenna was found on the torso of the woman model and was equal to 0.037 W/kg at 2.45 GHz. SAR levels increased significantly for the antenna transmitting inside the body. In this case, SAR values ranged between 0.23 and 0.45 W/kg at the same body location. The power absorbed in different body tissues and total power absorbed in the body were also calculated; the maximum total power absorbed was equal to 5.2 mW for an antenna input power equal to 10 mW. Habitat destruction and overexploitation are the main threats to biodiversity and where they co-occur, their combined impact is often larger than their individual one. Yet, detailed knowledge of the spatial footprints of these threats is lacking, including where they overlap and how they change over time. These knowledge gaps are real barriers for effective conservation planning. Here, we develop a novel approach to reconstruct the individual and combined footprints of both threats over time. We combine satellite-based land-cover change maps, habitat suitability models and hunting pressure models to demonstrate our approach for the community of larger mammals (48 species > 1 kg) across the 1.1 million km2 Gran Chaco region, a global deforestation hotspot covering parts of Argentina, Bolivia and Paraguay. This provides three key insights. First, we find that the footprints of habitat destruction and hunting pressure expanded considerably between 1985 and 2015, across ~40% of the entire Chaco – twice the area affected by deforestation. Second, both threats increasingly acted together within the ranges of larger mammals in the Chaco (17% increase on average, ± 20% SD, cumulative increase of co-occurring threats across 465 000 km2), suggesting large synergistic effects. Conversely, core areas of high-quality habitats declined on average by 38%. Third, we identified remaining priority areas for conservation in the northern and central Chaco, many of which are outside the protected area network. We also identify hotspots of high threat impacts in central Paraguay and northern Argentina, providing a spatial template for threat-specific conservation action. Overall, our findings suggest increasing synergistic effects between habitat destruction and hunting pressure in the Chaco, a situation likely common in many tropical deforestation frontiers. Our work highlights how threats can be traced in space and time to understand their individual and combined impact, even in situations where data are sparse. 相似文献
