Alzheimer’s disease (AD) is characterized by the appearance of amyloid‐β plaques, neurofibrillary tangles, and inflammation in brain regions involved in memory. Using mass spectrometry, we have quantified the phosphoproteome of the CK‐p25, 5XFAD, and Tau P301S mouse models of neurodegeneration. We identified a shared response involving Siglec‐F which was upregulated on a subset of reactive microglia. The human paralog Siglec‐8 was also upregulated on microglia in AD. Siglec‐F and Siglec‐8 were upregulated following microglial activation with interferon gamma (IFNγ) in BV‐2 cell line and human stem cell‐derived microglia models. Siglec‐F overexpression activates an endocytic and pyroptotic inflammatory response in BV‐2 cells, dependent on its sialic acid substrates and immunoreceptor tyrosine‐based inhibition motif (ITIM) phosphorylation sites. Related human Siglecs induced a similar response in BV‐2 cells. Collectively, our results point to an important role for mouse Siglec‐F and human Siglec‐8 in regulating microglial activation during neurodegeneration.  相似文献

Alginate is an important polysaccharide that is commonly used as a gelling agent in foods, cosmetics and healthcare products. Currently, all alginate used commercially is extracted from brown seaweed. However, with environmental changes such as increasing ocean temperature and the increasing number of biotechnological uses of alginates with specific properties, there is an emerging need for more reliable and customizable sources of alginate. An alternative to seaweed for alginate production is Pseudomonas aeruginosa, a common Gram-negative bacterium that can form alginate-containing biofilms. However, P. aeruginosa is an opportunistic pathogen that can cause life-threatening infections in immunocompromised patients. Therefore, we sought to engineer a non-pathogenic P. aeruginosa strain that is safe for commercial production of alginate. Using a homologous recombination strategy, we sequentially deleted five key pathogenicity genes from the P. aeruginosa chromosome, resulting in the marker-free strain PGN5. Intraperitoneal injection of mice with PGN5 resulted in 0% mortality, while injection with wild-type P. aeruginosa resulted in 95% mortality, providing evidence that the systemic virulence of PGN5 is highly attenuated. Importantly, PGN5 produces large amounts of alginate in response to overexpression of MucE, an activator of alginate biosynthesis. The alginate produced by PGN5 is structurally identical to alginate produced by wild-type P. aeruginosa, indicating that the alginate biosynthetic pathway remains functional in this modified strain. The genetic versatility of P. aeruginosa will allow us to further engineer PGN5 to produce alginates with specific chemical compositions and physical properties to meet different industrial and biomedical needs.  相似文献

Soil legacy effects are commonly highlighted as drivers of plant community dynamics and species co‐existence. However, experimental evidence for soil legacy effects of conditioning plant communities on responding plant communities under natural conditions is lacking. We conditioned 192 grassland plots using six different plant communities with different ratios of grasses and forbs and for different durations. Soil microbial legacies were evident for soil fungi, but not for soil bacteria, while soil abiotic parameters did not significantly change in response to conditioning. The soil legacies affected the composition of the succeeding vegetation. Plant communities with different ratios of grasses and forbs left soil legacies that negatively affected succeeding plants of the same functional type. We conclude that fungal‐mediated soil legacy effects play a significant role in vegetation assembly of natural plant communities.  相似文献

Aim: To investigate the relationship between glaucoma and intraocular pressure (IOP), intracranial pressure (ICP), trans-laminar cribrosa pressure difference (TLCPD), and the newly proposed fractional pressure reserve (FPR). Methods: Ten articles were analyzed by meta-analysis, and subgroup analysis of three factors was conducted. Results: The patients with normal tension glaucoma (NTG) and open-angle glaucoma (OAG) have higher TLCPD and lower ICP than healthy subjects, with obvious heterogeneity. The greater heterogeneity may arise from different ICP measurements and from different countries or regions. FPR performs better than TLCPD in some ways. Conclusion: Both FPR and TLCPD can be used as indicators of NTG and OAG, and FPR is more effective than TLCPD in comparisons between different experiments or individuals.  相似文献

Revegetation represents an effective measure for preventing soil erosion on the Loess Plateau. However, the effects of revegetation‐induced changes in soil and root properties on soil resistance to concentrated flow erosion (SRC) remain unclear. This study sampled soils and roots across a 25‐year chronosequence from farmland to grasslands of different ages (3, 7, 10, 18, and 25 years) to quantify variations in soil and root properties (soil bulk density, SBD; soil disintegration rate, SDR; saturated hydraulic conductivity, SHC; organic matter content, OMC; water‐stable aggregate, WSA; mean weight diameter, MWD; root mass density, RMD; root length density, RLD; and root surface area density, RSAD) and their effects on SRC. Farmland and grassland SRCs were obtained using a hydraulic flume. Soil properties and root density gradually improved with restoration time. In terms of the comprehensive soil property index calculated via principal component analysis, grassland values were 0.66 to 1.94 times greater than farmland values. Grassland SRCs increased and gradually stabilized (>18 years) over time and were 1.60 to 8.26 times greater than farmland SRC. SRC improvement was significantly related to increases in OMC, SHC, WSA, and MWD and decreases in SBD and SDR over time. SRC was effectively simulated by the Hill curve of RMD, RLD, and RSAD. SDR, SHC, and RMD (0.5–1.0 mm) affected SRC the most. This study scientifically describes how revegetation improves soil quality and soil resistance to flow erosion, and suggests that vegetations rich in 0.5–1.0 mm roots should be preferred during revegetation.  相似文献
