Background

Balloon injury (BI) of the rat carotid artery (CCA) is widely used to study intimal hyperplasia (IH) and decrease in lumen diameter (LD), but CCA's small diameter impedes the evaluation of endovascular therapies. Therefore, we validated BI in the aorta (AA) and iliac artery (CIA) to compare it with CCA.

Methods

Rats underwent BI or a sham procedure (control). Light microscopic evaluation was performed either directly or at 1, 2, 3, 4 and 16 weeks follow-up. The area of IH and the change in LD (LD at 16 weeks minus LD post BI) were compared.

Results

In the BI-groups the area of IH increased to 0.14 ± 0.08 mm2 (CCA), 0.14 ± 0.03 mm2 (CIA) and 0.12 ± 0.04 mm2 (AA) at 16 weeks (NS). The LD decreased with 0.49 ± 0.07 mm (CCA), compared to 0.22 ± 0.07 mm (CIA) and 0.07 ± 0.10 mm (AA) at 16 weeks (p < 0.05). The constrictive vascular remodelling (CVR = wall circumference loss combined with a decrease in LD) was -0.17 ± 0.05 mm in CIA but absent in CCA and AA. No IH, no decrease in LD and no CVR was seen in the control groups.

Conclusions

BI resulted in: (1.) a decrease in LD in CCA due to IH, (2.) a decrease in LD in CIA due to IH and CVR, (3.) no change in LD in AA, (4.) Comparable IH development in all arteries, (5.) CCA has no vasa vasorum compared to CIA and AA, (6.) The CIA model combines good access for 2 F endovascular catheters with a decrease in LD due to IH and CVR after BI.  相似文献

In sub-Saharan Africa, HIV-1 infection frequently occurs in the context of other coinfecting pathogens, most importantly, Mycobacterium tuberculosis and malaria parasites. The consequences are often devastating, resulting in enhanced morbidity and mortality. Due to the large number of confounding factors influencing pathogenesis in coinfected people, we sought to develop a nonhuman primate model of simian immunodeficiency virus (SIV)-malaria coinfection. In sub-Saharan Africa, Plasmodium falciparum is the most common malaria parasite and is responsible for most malaria-induced deaths. The simian malaria parasite Plasmodium fragile can induce clinical symptoms, including cerebral malaria in rhesus macaques, that resemble those of P. falciparum infection in humans. Thus, based on the well-characterized rhesus macaque model of SIV infection, this study reports the development of a novel rhesus macaque SIV-P. fragile coinfection model to study human HIV-P. falciparum coinfection. Using this model, we show that coinfection is associated with an increased, although transient, risk of both HIV and malaria transmission. Specifically, SIV-P. fragile coinfected macaques experienced an increase in SIV viremia that was temporarily associated with an increase in potential SIV target cells and systemic immune activation during acute parasitemia. Conversely, primary parasitemia in SIV-P. fragile coinfected animals resulted in higher gametocytemia that subsequently translated into higher oocyst development in mosquitoes. To our knowledge, this is the first animal model able to recapitulate the increased transmission risk of both HIV and malaria in coinfected humans. Therefore, this model could serve as an essential tool to elucidate distinct immunological, virological, and/or parasitological parameters underlying disease exacerbation in HIV-malaria coinfected people.  相似文献

Threats to a species' persistence are likely to change as conservation measures reduce some threats, while natural and anthropogenic changes increase others. Despite a variety of potential underlying mechanisms, extinction threats will be manifested through one of the 3 components of population dynamics: reducing population growth potential, increasing population variability, or lowering the population ceiling. Consequently, effective management can be guided by monitoring programs and population models that examine each of these components. We examined the potential for a coupled monitoring and modeling effort to guide management of species-at-risk while accounting for evolving risks using the case study of the threatened San Clemente sage sparrow (Amphispiza belli clementeae). Originally listed due to a low population ceiling imposed by severe habitat loss, we found that the major threat to San Clemente sage sparrow persistence has shifted to low population growth potential driven by high juvenile mortality. We further found that successful mitigation of high juvenile mortality will shift the primary threat to drought frequency, which is predicted to increase on San Clemente Island as a consequence of global climate change. The latter shift is a consequence of the boom-bust ecology exhibited by San Clemente sage sparrows in response to rainfall—likely a common characteristic of short-lived terrestrial vertebrates in arid environments. Our ability to successfully recover this species hinges on a comprehensive monitoring and modeling program incorporating all 3 components of population dynamics informing changes in management priorities to reflect shifting threats. Our study indicates that the next critical step to recovering sage sparrows is to understand and mitigate the causes of high juvenile mortality. In response to these predictions, the United States Navy has funded a radio-telemetry study to determine the cause(s) of juvenile mortalities. © 2011 The Wildlife Society.  相似文献

Many butterfly populations are monitored by counting the number of butterflies observed while walking transects during the butterfly’s flight season. Methods for estimating population abundance from these transect counts are appealing because they allow rare populations to be monitored without capture–recapture studies that could harm fragile individuals. An increasingly popular method for estimating abundance from transect counts relies on strong assumptions about the counting process and the processes that govern butterfly population dynamics. Here, we study the statistical performance of this method when underlying model assumptions are violated. We find that estimates of population size are robust to departures from underlying model assumptions, but that the uncertainty in these estimates (i.e., confidence intervals) is substantially underestimated. Alternative bootstrap and Bayesian methods provide better measures of the uncertainty in estimated population size, but are conditional upon knowledge of butterfly detectability. Because of these requirements, a mixed approach that combines data from small capture–recapture studies with transect counts strikes the best balance between accurate monitoring and minimal injury to individuals. Our study is motivated by monitoring studies for St. Francis satyr (Neonympha mitchelli francisci), a rare and relatively immobile butterfly occurring only in the sandhills region of south-central North Carolina, USA.  相似文献

The p16(INK4a) tumor suppressor gene is a mediator of cellular senescence and has been suggested to be a biomarker of 'molecular' age in several tissues including T cells. To determine the association of both active and suppressed HIV infection with T-cell aging, T-cell p16(INK4a) expression was compared between 60 HIV+ suppressed subjects, 23 HIV+ untreated subjects, and 18 contemporaneously collected HIV-negative controls, as well as 148 HIV-negative historical samples. Expression did not correlate with chronologic age in untreated HIV+ patients, consistent with an effect of active HIV replication on p16(INK4a) expression. In patients on cART with suppressed viral loads, however, p16(INK4a) levels were similar to uninfected controls and correlated with chronologic age, with a trend toward an inverse correlation with CD4 count. These data show that p16(INK4a) is a reliable biomarker of T-cell aging in HIV+ patients with suppressed viral loads and suggest that poor CD4 cell recovery on cART may be associated with increased T-cell expression of p16(INK4a) , a marker of cellular senescence.  相似文献

Connecting isolated patches of habitat in fragmented landscapes with corridors is a popular conservation strategy. This strategy is also controversial in large part because of uncertainty about what characteristics of a species and its environment promote corridor use. In this article we address the question, For what types of species will populations benefit from corridors? We asked this question using a model of two logistically growing populations connected by migration in which both emigration and migration success were determined by the presence or absence of a corridor. We found that in the short run (e.g., during recovery from disaster), corridors are most effective for species with fast-growing populations that have low survivorship when dispersing through unsuitable (matrix) habitat. We also found that emigration rates and habitat-specific mortality rates are key determinants of the effects of corridors on population size. In the long term, corridors are most likely to benefit species with slow-growing populations that have low survivorship when dispersing through matrix habitat. Our results confirm the major conclusions from previous empirical studies of corridor benefits. However, most studies fail to consider the most appropriate questions to determine the potential benefits of habitat corridors. First, what is the time scale of the conservation goal? Corridors have positive effects on different suites of species in the short and long term. Second, is the major threat of local extinction due to sustained population decline or boom-bust cycles? Third, what is the migration rate through the matrix? Fourth, what fraction of migrants dispersing through the matrix successfully immigrate to another patch?  相似文献

In this case report the occurrence of a catheter-induced coronary artery dissection is described. In our patient, angiography showed a mushroom-shaped exudate above the left main coronary artery. Intravascular ultrasound revealed a circular dissection with a huge false lumen connected to the true lumen by a small intimal tear. A brief review of the literature on catheter-induced coronary dissection is included. We believe that this case report provides a good illustration of the need for careful reviewing of indications for angiography. Although procedural risks are low, angiography remains an invasive diagnostic test with the potential to cause severe complications.  相似文献

Summary Free-living nematodes may be attacked and eaten by soil mites which are normally considered fungivores or saprophages. Three species in the genus Tyrophagus, common inhabitants of grassland soils and also common pests in stored products, museums and laboratories, are predators of nematodes. All active stages of the mites will voraciously consume nematodes. When offered nematodes and a choice of other food (baker's yeast and algae), 11% of the Tyrophagus putrescentiae, 23% of the T. zachvatkini, and 56% of the T. similis tested fed on nematodes. Tyrophagus zechvatkini and similis were reared on a diet consisting entirely of nematodes, and developed at rates similar to a fungal diet and produced viable offspring. Agar cultures of Aphelenchus avenae which were inoculated with five mating pairs of Tyrophagus zachvatkini had populations that were one-third less than mite-free controls. Observations indicate that nematodes may be attacked by tyrophagid mites when in a dry, anhydrobiotic state.  相似文献

Activation of highly specific biochemical processes by simple chemical agents is demonstrated for morphogenesis (anlage and development of female gametophyte in cereal) and mitosis (in cell cultures and animal and plant tissues). The effects of these agents are tissue-specific. Structure--activity relationship is analyzed in this group of compounds. Thus, the phenomenon reveals the exact pathways of the influence of allelopathic and anthropogenic chemical agents on evolution of plant biocenoses.  相似文献
