The continuous assembly and disassembly of focal adhesions is required for efficient cell spreading and migration. The G-protein-coupled receptor kinase-interacting protein 1 (GIT1) is a multidomain protein whose dynamic localization to sites of cytoskeletal remodeling is critically involved in the regulation of these processes. Here we provide evidence that the subcellular localization of GIT1 is regulated by protein kinase D3 (PKD3) through direct phosphorylation on serine 46. GIT1 phosphorylation on serine 46 was abrograted by PKD3 depletion, thereby identifying GIT1 as the first specific substrate for this kinase. A GIT1 S46D phosphomimetic mutant localized to motile, paxillin-positive cytoplasmic complexes, whereas the phosphorylation-deficient GIT1 S46A was enriched in focal adhesions. We propose that phosphorylation of GIT1 on serine 46 by PKD3 represents a molecular switch by which GIT1 localization, paxillin trafficking, and cellular protrusive activity are regulated. 相似文献

The diaminopimelic acid pathway of lysine biosynthesis has been suggested to provide attractive targets for the development of novel antibacterial drugs. Here we report the characterization of two enzymes from this pathway in the human pathogen Pseudomonas aeruginosa, utilizing structural biology, biochemistry and genetics. We show that tetrahydrodipicolinate N-succinyltransferase (DapD) from P. aeruginosa is specific for the L-stereoisomer of the amino substrate L-2-aminopimelate, and its D-enantiomer acts as a weak inhibitor. The crystal structures of this enzyme with L-2-aminopimelate and D-2-aminopimelate, respectively, reveal that both compounds bind at the same site of the enzyme. Comparison of the binding interactions of these ligands in the enzyme active site suggests misalignment of the amino group of D-2-aminopimelate for nucleophilic attack on the succinate moiety of the co-substrate succinyl-CoA as the structural basis of specificity and inhibition. P. aeruginosa mutants where the dapA gene had been deleted were viable and able to grow in a mouse lung infection model, suggesting that DapA is not an optimal target for drug development against this organism. Structure-based sequence alignments, based on the DapA crystal structure determined to 1.6 Å resolution revealed the presence of two homologues, PA0223 and PA4188, in P. aeruginosa that could substitute for DapA in the P. aeruginosa PAO1ΔdapA mutant. The potential of an organic monolith with incorporated titanium dioxide (TiO(2)) and zirconium dioxide (ZrO(2)) nanoparticles was evaluated for the selective enrichment of phosphorylated peptides from tryptic digests. A pipette tip was fitted with a monolith based on divinylbenzene (DVB) of highly porous structure, which allows sample to pass through the monolithic bed. The enrichment of phosphopeptides was enhanced by increasing the pipetting cycles during the sample preparation and a higher recovery could be achieved with adequate buffer systems. A complete automated process was developed for enrichment of phosphopeptides leading to high reproducibility and resulting in a robust method designed to minimize analytical variance while providing high sensitivity at high sample throughput. The effect of particle size on the selectivity of phosphopeptides was investigated by comparative studies with nano- and microscale TiO(2) and ZrO(2) powders. Eleven phosphopeptides from alpha-casein digest could be recovered by an optimized mixture of microscale TiO(2)/ZrO(2) particles, whereas nine additional phosphopeptides could be retained by the same mixture of nano-structured material. When compared to conventional immobilized metal-ion affinity chromatography and commercial phosphorylation-enrichment kits, higher selectivity was observed in case of self fabricated tips. About 20 phosphopeptides could be retained from alpha-casein and five from beta-casein digests by using TiO(2) and ZrO(2) based extraction tips. Further selectivity for phosphopeptides was demonstrated by enriching a digest of in vitro phosphorylated extracellular signal regulated kinase 1 (ERK1). Host caste recognition may be important for the dispersal of phoretic mites associated with social insects. All developmental
stages of the mite Parasitellus fucorum (Acari: Mesostigmata: Parasitidae) live in the nests of bumblebees (Hymenoptera: Apidae: Bombus). Dispersal occurs by specialised phoretic instars, deutonymphs, which attach to adult bumblebees. Since bumblebee colonies
are annual and only young queens overwinter, deutonymphs that are able to discriminate between bumblebee castes and preferentially
attach to queens should be favoured by selection. In the field, deutonymphs of P. fucorum were found to be phoretic on bumblebee workers and queens, and in behavioural experiments all castes proved to be attractive
as carriers for the mites. However, they preferred queens that had hibernated as carriers when they could choose between workers
and queens. In a further experiment, when given a choice, deutonymphs switched from males to young queens but never transferred
from a queen to a male. These results suggest that deutonymphs preferentially attach to queens but may also use other castes
for transport. Those dispersing on workers and males may try to switch to queens later. Host-switching is possible during
copulation and on flowers, where bees of all castes forage.

A data base was generated for quantifying effects of thermal time (degree-days) on the appearance of new leaves, the expansion of such leaves to maximum area, their death, the appearance of new internodes below the node associated with such leaves, and the extension of these internodes to maximum length. The data base for a list of crop (agronomic and horticultural), weed, and native Tallgrass Prairie plants has been summarized, with equations for the above events as a function of degree days, with appropriate base temperatures and maximum cut-off temperatures, in a Java applet which is available at a website with the URL . Associated graphical plots such as shown in this paper are also given. Branching behavior was accounted for. These events predict the effect of thermal time on leaf age and its height in the plant canopy, both important factors needed for upscaling functions for leaf behavior to those for behavior of the plant canopy. The data base is evolving to include coefficients for other species. Coefficients are used to predict the leaf area index of the canopy, which is important for predicting evapotranspiration from the crop and the protection of the soil from erosion. 相似文献

A case of primary nonsecretory plasmacytoma of the spleen is reported. On laparotomy and splenectomy a 920 g spleen was removed,
measuring 16×14×6 cm. The cut surface of the entire spleen showed that the tumour occupied most of the splenic tissue. A bone
marrow aspirate and trephine, skeletal survey showed no signs of myeloma. Biopsy of the liver and regional lymph nodes was
normal. Immunocytochemistry of the splenic tumour showed positivity for pan-B and plasma cell markers. After splenectomy the

Graphitic nanofibres (GNFs), 100–200 nm in diameter and 5–20 μm in length have been modified in order to yield different affinities
(Cu2+ and Fe3+ loaded immobilized metal affinity chromatography (IMAC) as well as cation and anion exchange materials) for the extraction
of a range of biomolecules by their inherited hydrophobicity and the hydrophilic chemical functionalities, obtained by derivatization.
Modified GNFs have for the first time been employed as carrier materials for protein profiling in material-enhanced laser
desorption/ionization (MELDI) for the enrichment and screening of biofluids. For that purpose, the derivatized GNF materials
have comprehensively been characterized regarding surface area, structural changes during derivatization, IMAC, as well as
ion exchange and protein-loading capacity and recovery. GNF derivatives revealed high protein-binding capacity (2,000 μg ml−1 for insulin) and ideal sensitivities, resulting in a detection limit of 50 fmol μl−1 (for insulin), which is crucial for the detection of low abundant species in biological samples. Compared to other MELDI
carrier materials, sensitivity was enhanced on GNF derivatives, which might be ascribed to the fact that GNFs support desorption
