We documented the spatial distribution, abundance and molecular diversity of mycorrhizal hyphae and physical and chemical properties of soil-weathered bedrock in a chaparral community that experiences seasonal drought. Because plants in this community were known to rely on bedrock-stored water during the summer, the data were used to evaluate the potential role of mycorrhizal hyphae in accessing bedrock-stored water during summer drought. The granitic bedrock was characterized by factures filled with a disaggregated, sandy loam that acted as conduits for water, and matrices composed of soil-weathered granite that retained the fabric and structure of rock. Mycorrhizal hyphae of six ectomycorrhizal taxa (from the Basidiomycota and Ascomycota), and arbuscular mycorrhizal hyphae (Zygomycota) were recovered from both fracture and matrix compartments to depths greater than 200 cm. Our findings also indicated a potential linkage between the abundance of Ascomycete hyphae, substrate physical (bulk density) and chemical properties (total N, N:P, Ca:Mg), and bedrock moisture content, as well as spatial patterning between hyphae and resources at a scale of 25–45 cm. Such linkages suggest that mycorrhizal fungal hyphae may be part of an adaptive mechanism that enables chaparral plants to survive seasonal drought.  相似文献

Effects of ionophore type and carrier on in vitro ruminal digestion and fermentation patterns of a concentrate substrate were evaluated at various incubation times. Treatments were: control (no ionophore); lasalocid sodium commercial premix (Bov); lasalocid sodium mycelium cake (LasBio); laidlomycin sodium salt (LaidNa); laidlomycin propionate commercial premix (LaidPro); monensin sodium salt (Mon); and monensin sodium commercial premix (Rum). The Bov, LasBio, Mon, and Rum treatments supplied 4 μg of ionophore/mL of culture volume, whereas the LaidNa and LaidPro treatments supplied 1.33 μg of ionophore/mL. Total gas and methane production did not differ among treatments at any of the incubation times (P>0.09). Similarly, in vitro dry matter disappearance (IVDMD) was not affected by treatment (P>0.28) at 6, 18, and 24 h of incubation; however, IVDMD (P=0.03) was greater for ionophores than for the control at 12 h of incubation. Molar proportions of acetate (P<0.01), acetate:propionate (P<0.01), and total volatile fatty acid (VFA) concentrations (P<0.01) were decreased and propionate was increased (P<0.001) for the average of all ionophore-containing substrates compared with the control. Total VFA were decreased by Bov, LaidNa, and Rum, contrasted with their specific counterparts (LasBio, LaidPro, and Mon, respectively; P<0.05). Differences were detected among ionophore types for acetate (lasalocid vs. laidlomycin; P<0.05), propionate (lasalocid vs. monensin; P<0.05), and butyrate (monensin vs. lasalocid or laidlomycin; P<0.05). Capture of metabolic hydrogen in end products of fermentation was greater for ionophore-containing treatments (P<0.01) than for the control. These data suggest limited unique effects of ionophore type or carrier on IVDMD, total gas production, and methane; however, VFA proportions varied among ionophore types and carriers, which deserves further study.  相似文献

Background

Inhaled short acting β2-agonists (SABA), e.g. albuterol, are used for quick reversal of bronchoconstriction in asthmatics. While SABA are not recommended for maintenance therapy, it is not uncommon to find patients who frequently use SABA over a long period of time and there is a suspicion that long term exposure to SABA could be detrimental to lung function. To test this hypothesis we studied the effect of long-term inhaled albuterol stereoisomers on immediate allergic response (IAR) and airway hyperresponsiveness (AHR) in mouse models of asthma.

Methods

Balb/C mice were sensitized and challenged with ovalbumin (OVA) and then we studied the IAR to inhaled allergen and the AHR to inhaled methacholine. The mice were pretreated with nebulizations of either racemic (RS)-albuterol or the single isomers (S)- and (R)-albuterol twice daily over 7 days prior to harvest.

Results

We found that all forms of albuterol produced a significant increase of IAR measured as respiratory elastance. Similarly, we found that AHR was elevated by albuterol. At the same time a mouse strain that is intrinsically hyperresponsive (A/J mouse) was not affected by the albuterol isomers nor was AHR induced by epithelial disruption with Poly-L-lysine affected by albuterol.

Conclusions

We conclude that long term inhalation treatment with either isomer of albuterol is capable of precipitating IAR and AHR in allergically inflamed airways but not in intrinsically hyperresponsive mice or immunologically naïve mice. Because (S)-albuterol, which lacks affinity for the β2-receptor, did not differ from (R)-albuterol, we speculate that isomer-independent properties of the albuterol molecule, other than β2-agonism, are responsible for the effect on AHR.  相似文献

Measures of population productivity and individual survival are essential for effective conservation management and scientific investigations. In studies of birds, individuals are often marked with unique colour band combinations to enable estimates of population vital rates. If birds are banded in the nest this must be done at the correct stage in nestling development. If nestlings are banded too early it can lead to leg damage. If they are banded too late it can cause early fledging and reduced survival. Here, we use photographs to guide estimation of the best stage to band North Island robin (Petroica longipes), or toutouwai, nestlings, which is typically at 9–12 days of age.  相似文献

We investigated the effects of Nigella sativa on apoptosis and gamma-aminobutyric acid (GABAA) receptor density in cerebral cortical and hippocampal neurons in a pentylenetetrazol (PTZ)-induced kindling model in rats. The PTZ kindling model was produced by injecting PTZ in subconvulsive doses to rats on days 1, 3, 5, 8, 10, 12, 15, 17, 19, 22 and 24 of the study into animals of PTZ treated (PTZ) and PTZ + N. sativa treated (PTZ + NS) groups. Clonic and tonic seizures were induced by injecting a convulsive dose of PTZ on day 26 of the study. Rats in the PTZ + NS group were treated also with a 10 mg/kg methanolic extract of N. sativa 2 h before each PTZ injection. Rats in the control group were treated with 4 ml/kg saline. The number of neurons that expressed GABAA receptors in the hippocampus and cerebral cortex of rats in the PTZ and PTZ + NS groups increased significantly. There was no significant difference in the number of GABAA receptors between the PTZ and PTZ + NS groups. GABAA receptor density of the neurons in the cerebral cortex, but not hippocampus, was increased in PTZ group compared to controls. We observed a significant increase in the number of apoptotic neurons in the cerebral cortex of rats of both the PTZ and PTZ + NS groups compared to controls. We observed a significant decrease in the number of the apoptotic neurons in the cerebral cortex of rats in the PTZ + NS group compared to the PTZ group. N. sativa treatment ameliorated the PTZ induced neurodegeneration in the cerebral cortex as reflected by neuronal apoptosis and neuronal GABAA receptor frequency.  相似文献

Although two cryptic pipistrelle bat species, Pipistrellus pipistrellus and Pipistrellus pygmaeus , belong among the most common bat species in Europe, it is still unclear whether they can migrate over long distances between summer and winter roosts. Long-distance migratory species may be expected to show low levels of genetic structuring in large areas due to regular mixing of the gene pool by mating that occurs during migration and/or hibernation. Conversely, the dispersal of gametes in sedentary species is spatially restricted, populations are more genetically structured, and isolation by relatively short distance is visible. By analysing diversity of highly variable microsatellites within and among summer colonies of both studied species in central Europe, we found that differentiation between populations is very weak. Both classical F ST and Bayesian clustering approach failed to detect genetic structure among colonies and there was no significant isolation-by-distance pattern. The analyses of relatedness, however, revealed that individuals within colonies are more related than random suggesting philopatry of at least one sex. The results were very similar for the two species. Abstract:  A new method for the extraction of calcified and/or partly pyritized macrofossils has been developed. This method is based upon the differential speeds for the dissolving of microcrystalline and macrocrystalline calcite in 38% sulphuric acid. The effectiveness of the sulphuric acid treatment is also influenced by the volume of clay minerals in the host rock. Therefore, this method is highly applicable for the extraction of macrofossils from marlstones, marly limestones, and other lithified calcareous sediments. The main advantages of this method, when compared with other chemical methods, are (1) the short treatment time, (2) the capability of dissolving the sediment on the fossil's surface, and (3) its efficiency in dissolving calcareous rocks with low porosity. This method has been successfully applied to Upper Cretaceous macrofossils from the Bohemian Cretaceous Basin. The surface of extracted macrofossils remained undamaged, exhibiting minute skeletal details; perhaps even encrusters and bioerosions.  相似文献
