New core and seismic data suggest that widespread reef building started on Lang Bank by 8,900 CalBP and was dominated by Acropora palmata for the next three millennia. Accretion rates averaged 5.81 m ky?1, a rate that was sufficient for reefs to keep pace with rising sea level on the bank throughout their history. Seismic data show a deep platform interior that was flooded well in advance of reef building along the elevated rim. As a result, those reefs were buffered from sediment stress by their higher positions and active water flow to the west. A. palmata disappeared from the shallow margin by 6,350 yr ago, and reef building on Lang Bank largely ceased by 5,035 CalBP. The reasons for these dramatic events are unclear. Water depth over the reefs was generally shallower than when they started to build, and sea level was slowing dramatically. The new data described here show that reefs flourished on Lang Bank throughout the hiatus suggested by earlier studies (10–7 kyrs BP), and the ultimate demise of shelf-edge reefs is clearly not associated with either poor water quality or sudden sea-level rise. In addition, accretion rates from eastern St. Croix and throughout the Caribbean were well below the high values (≥10 m ky?1) that have been widely assumed. These data collectively argue against models that require extreme environmental or oceanographic phenomena to drown reefs on Lang Bank where reef building was too fast to be outpaced by Holocene sea-level rise. This also bears on more generalized Caribbean models that depend on the presumed reef history on eastern St. Croix.  相似文献

Under magnetic resonance (MR) guidance, high intensity focused ultrasound (HIFU) is capable of precise and accurate delivery of thermal dose to tissues. Given the excellent soft tissue imaging capabilities of MRI, but the lack of data on the correlation of MRI findings to histology following HIFU, we sought to examine tumor response to HIFU ablation to determine whether there was a correlation between histological findings and common MR imaging protocols in the assessment of the extent of thermal damage. Female FVB mice (n = 34), bearing bilateral neu deletion tumors, were unilaterally insonated under MR guidance, with the contralateral tumor as a control. Between one and five spots (focal size 0.5 × 0.5 × 2.5 mm3) were insonated per tumor with each spot receiving approximately 74.2 J of acoustic energy over a period of 7 seconds. Animals were then imaged on a 7T MR scanner with several protocols. T1 weighted images (with and without gadolinium contrast) were collected in addition to a series of T2 weighted and diffusion weighted images (for later reconstruction into T2 and apparent diffusion coefficient maps), immediately following ablation and at 6, 24, and 48 hours post treatment. Animals were sacrificed at each time point and both insonated/treated and contralateral tumors removed and stained for NADH-diaphorase, caspase 3, or with hematoxylin and eosin (H&E). We found the area of non-enhancement on contrast enhanced T1 weighted imaging immediately post ablation correlated with the region of tissue receiving a thermal dose CEM43 ≥ 240 min. Moreover, while both tumor T2 and apparent diffusion coefficient values changed from pre-ablation values, contrast enhanced T1 weighted images appeared to be more senstive to changes in tissue viability following HIFU ablation.  相似文献

Simulating factors affecting human athletic performance, including fatigue, requires a dynamic model of the bioenergetic capabilities of the athlete. To address general cases, the model needs inputs, outputs, and states with a set of differential equations describing how the inputs affect the states and outputs as functions of time. We improve an existing phenomenological muscle model, removing unnecessarily fast dynamic behavior, adding force–velocity dependence, and generalizing it to task level activities. This makes it more suitable for simulating and calculating optimal strategies of athletic events of medium duration (longer than a sprint but shorter than a marathon). To examine the validity and limitations of the model, parameters have been identified from numerical fits to published experimental data.  相似文献

We have localized and identified five rat hepatocyte plasma membrane proteins using hybridoma technology in combination with morphological and biochemical methods. Three different membrane preparations were used as immunogens: isolated hepatocytes, a preparation of plasma membrane sheets that contained all three recognizable surface domains of the intact hepatocyte (sinusoidal, lateral, and bile canalicular), and a glycoprotein subfraction of that plasma membrane preparation. We selected monoclonal IgGs that were hepatocyte specific and localized them using both immunofluorescence on 0.5-micron sections of frozen liver and immunoperoxidase at the ultrastructural level. One antigen (HA 4) was localized predominantly to the bile canalicular surface, whereas three (CE 9, HA 21, and HA 116) were localized predominantly to the lateral and sinusoidal surfaces. One antigen (HA 16) was present in all three domains. Only one antigen (HA 116) could be detected in intracellular structures both in the periphery of the cell and in the Golgi region. The antigens were all integral membrane proteins as judged by their stability to alkaline extraction and solubility in detergents. The apparent molecular weights of the antigens were established by immunoprecipitation and/or immunoblotting. In a related study (Bartles, J.R., L.T. Braiterman, and A.L. Hubbard, 1985, J. Cell. Biol., 100:1126-1138), we present biochemical confirmation of the domain-specific localizations for two of the antigens, HA 4 and CE 9, and demonstrate their suitability as endogenous domain markers for monitoring the separation of bile canalicular and sinusoidal lateral membrane on sucrose density gradients.  相似文献

We have used a combined biochemical and morphological approach to establish the suitability of certain endogenous and exogenous domain markers for monitoring the separation of rat hepatocyte plasma membrane domains in sucrose density gradients. As endogenous domain markers, we employed two of the integral plasma membrane protein antigens, HA 4 and CE 9, localized to the bile canalicular and sinusoidal/lateral domains, respectively, of the hepatocyte plasma membrane in rat liver tissue (Hubbard, A. L., J. R. Bartles, and L. T. Braiterman, 1985, J. Cell Biol., 100:1115-1125). We used immunoelectron microscopy with a colloidal gold probe to demonstrate that HA 4 and CE 9 retained their domain-specific localizations on isolated hepatocyte plasma membrane sheets. When the plasma membrane sheets were vesiculated by sonication and the resulting vesicles were centrifuged to equilibrium in sucrose density gradients, quantitative immunoblotting revealed that the vesicles containing HA 4 and those containing CE 9 exhibited distinct density profiles. The density profile for the bile canalicular vesicles (marked by HA 4) was characterized by a single peak at a density of 1.10 g/cm3. The density profile for the sinusoidal/lateral vesicles (marked by CE 9) was bimodal, with a peak in the body of the gradient at a density of 1.14 g/cm3 and a smaller amount in the pellet (density greater than or equal to 1.17 g/cm3). We used this sucrose gradient fractionation as a diagnostic procedure to assign domain localizations for several other hepatocyte plasma membrane antigens and enzyme activities. In addition, we used the technique to demonstrate that 125I-wheat germ agglutinin, introduced during isolated liver perfusion at 4 degrees C, can serve as an exogenous domain marker for the sinusoidal domain of the rat hepatocyte plasma membrane.  相似文献

Our study has examined the synthesis of platelet activating factor (PAF; 1-0-alkyl-2-acetyl-sn-glycero-3-phosphocholine) and of structurally related molecules by an enriched preparation (greater than 70%) of the human lung mast cell (HLMC) in response to immunologic stimulation. Upon activation with anti-IgE, HLMC incorporated exogenously provided acetate into a phospholipid that migrated with authentic PAF on TLC. The formation of this product in HLMC occurred concomitantly with histamine and leukotriene C4 release. Further analysis of this phospholipid revealed that 1-acyl-2-acetyl-sn-glycero-3-phosphocholine (GPC) and not 1-alkyl-2-acetyl-GPC was the major 1-radyl-2-acetyl-GPC subclass formed during cell activation. The presence of 1-alkyl-2-acetyl-GPC was confirmed by negative ion chemical ionization mass spectrometry. In addition to this product, anti-IgE-stimulated HLMC synthesized relatively small quantities of another 2-acetylated phospholipid migrating on TLC between phosphatidylcholine and phosphatidylinositol. The chromatographic characteristics of this product suggested that it is a subclass of 1-radyl-2-acetyl-sn-glycero-3-phosphoethanolamine. The catabolism of both 1-acyl-2-acetyl-GPC and 1-alkyl-2-acetyl-GPC was next examined to determine if the predominant formation of 1-acyl-2-acetyl-GPC over 1-alkyl-2-acetyl-GPC were metabolized by the HLMC at similar rates. There was, however, a qualitative difference in the metabolic products derived from the two phospholipids. 1-Alkyl-2-acetyl-GPC was rapidly inactivated by removal of the acetate moiety at the sn-2 position followed by rapid reacylation with arachidonate. By contrast, 1-acyl-2-acetyl-GPC was catabolized mainly by removal of the fatty acyl moiety at the sn-1 position. These data demonstrate the natural occurrence of PAF and at least two structurally similar molecules in anti-IgE stimulated HLMC. Furthermore, an analog containing an ester linkage at the sn-1 position, 1-acyl-2-acetyl-GPC, appears to be the major acetylated product synthesized under these conditions.  相似文献

During ripening of bananas (Musa spp. [AAA group, Cavendish subgroup]), there is a massive conversion of starch to sucrose. Also during ripening there is a rise in respiration known as the respiratory climacteric. In this study changes in carbohydrate content, activities of starch and sucrose metabolizing enzymes, and respiration were measured to assess their potential interrelationships. Sucrose phosphate synthase activity increased dramatically during the first 4 days after initiation of ripening by ethylene treatment. Starch concentration decreased and sucrose concentration increased during this time period. Developmental changes in sucrose phosphate synthase activity were measured with limiting substrate (plus Pi) and saturating substrate concentrations. Activities were not parallel under the two assay conditions, providing tentative evidence that kinetically different forms of the enzyme may exist at different stages of ripening. Sucrose accumulation rate was most highly correlated with sucrose phosphate synthase activity assayed with limiting substrate concentrations (plus Pi). The cumulative amount of CO2 respired during ripening was positively correlated with sugar accumulation (R2 = 0.97). From this linear regression it was calculated that a constant 0.605 millimoles of CO2 was evolved per mole of sucrose formed throughout ripening. Using this quantity, the percentage of the total respiratory ATP produced which was required for the conversion of starch to sucrose was calculated assuming different models for carbon export from the amyloplast. The results suggest that sucrose biosynthesis during ripening constitutes a significant sink for respiratory ATP.  相似文献

Rates of primary and bacterial secondary production in Lake Arlington, Texas, were determined. The lake is a warm (annual temperature range, 7 to 32°C), shallow, monomictic reservoir with limited macrophyte development in the littoral zone. Samples were collected from six depths within the photic zone from a site located over the deepest portion of the lake. Primary production and bacterial production were calculated from NaH14CO3 and [methyl-3H]thymidine incorporation, respectively. Peak instantaneous production ranged between 14.8 and 220.5 μg of C liter−1 h−1. There were two distinct periods of high rates of production. From May through July, production near the metalimnion exceeded 100 μg of C liter−1 h−1. During holomixis, production throughout the water column was in excess of 100 μg of C liter−1 h−1 and above 150 μg of C liter−1 h−1 near the surface. Annual areal primary production was 588 g of C m−2. Bacterial production was markedly seasonal. Growth rates during late fall through spring were typically around 0.002 h−1, and production rates were typically 5 μg of C liter−1 h−1. Growth rates were higher during warmer parts of the year and reached 0.03 h−1 by August. The maximum instantaneous rate of bacterial production was approximately 45 μg of C liter−1 h−1. Annual areal bacterial production was 125 g of C m−2. Temporal and spatial distributions of bacterial numbers and activities coincided with temporal and spatial distributions of primary production. Areal primary and bacterial secondary production were highly correlated (r = 0.77, n = 15, P < 0.002).  相似文献
