为了探讨评估结直肠癌预后指标。应用催化信号放大-原位杂交技术。研究90例结直肠癌组织中CD15和nm23H1的mRNA表达,并结合随访资料分析,结果表明,在结直肠癌中CD15和nm23H1的mRNA阳性表达分别为84.4%和66.7%。CD15mRNA高表达及nm23H1mRNA低表达与结直肠癌浸润程度,淋巴结转移和肝脏转移状况密切相关,提示CD15和nm23H1的mRNA表达均可作为预测结直肠癌侵袭转移和客观评估患者预后的生物学指标。 相似文献

Differences between the prion protein and its homolog Doppel: a partially structured state with implications for scrapie formation 总被引:2,自引:0,他引:2
The key event in the pathogenesis of prion diseases is a conformational change in the prion protein (PrP). Models for conversion of PrP(C) into PrP(Sc) typically implicate an, as yet, unidentified intermediate. In an attempt to identify such an intermediate, we used native-state hydrogen exchange monitored with NMR. Although we were unable to detect an intermediate directly, we observed substantial protection above that expected based upon measurements of the global stability of PrP (>2 kcal mol(-1) super protection). This super protection implicates either structure in the denatured state or the presence of an intermediate. Similar experiments with Doppel, a homolog of PrP that does not form infectious prions, failed to demonstrate such super protection. This suggests that the partially structured state of PrP encompassing portions of the B and C helices, may be a significant factor in the ability of PrP to convert from PrP(C) to PrP(Sc). 相似文献

The test on the preference of Thrips palmi to eight color sticky cards was carried out in an eggplant field. The thrip had the strongest preference to blue sticky card (P<0.01), and its preference order to the other 7 color cards was as follows: blue, turguoise, yellow, deep blue, green, orange, red and black. Results on the trapping effect of blue sticky card from east, south, west and north directions show that most thrips were trapped from the north,which existed a significant difference with those from the other three directions (P<0.05). Five blue sticky cards were set up at the heights of 73.9,101.7,129.5,157.3 and 185.1 cm above the ground to trap thrips, when the average height of eggplant was approximately 70 cm. More thrips were trapped of 73.9 and 101.7 cm height, which existed significant difference with those at the other three heights. During 5 continuous 3 hour spans from 5 am to 8 pm, the numbers of female, male and total adult thrips trapped were not significantly different. 相似文献

为了探讨评估肾细胞癌(RCC)预后指标,应用免疫组化法研究60例RCC中细胞粘附分子CD15和Bcl-2蛋白表达及其相关关系。结果表明,在RCC中CD15和Bcl-2表达阳性率分别为70.0%和61.7%。CD15高表达与RCC分级、分期和淋巴结转移有关。Bcl-2低表达与RCC分级有关,与其分期和淋巴结转移无关。CD15表达与Bcl-2表达呈负相关。结果提示,CD15表达是RCC发生、发展和转移的指标,Bcl-2是RCC发生的早期事件。 相似文献

MicroRNAs (miRNAs) are small non-coding RNAs that regulate protein-coding gene expression primarily found in plants and animals. Fungi produce microRNA-like RNAs (milRNAs) that are structurally similar to miRNAs and functionally important in various biological processes. The fungus Fusarium oxysporum f. sp. cubense (Foc) is the causal agent of Banana Fusarium vascular wilt that threatens global banana production. It remains uncharacterized about the biosynthesis and functions of milRNAs in Foc. In this study, we investigated the biological function of milRNAs contributing to Foc pathogenesis. Within 24 hours post infecting the host, the Argonaute coding gene FoQDE2, and two Dicer coding genes FoDCL1 and FoDCL2, all of which are involved in milRNA biosynthesis, were significantly induced. FoQDE2 deletion mutant exhibited decreased virulence, suggesting the involvement of milRNA biosynthesis in the Foc pathogenesis. By small RNA sequencing, we identified 364 small RNA-producing loci in the Foc genome, 25 of which were significantly down-regulated in the FoQDE2 deletion mutant, from which milR-87 was verified as a FoQDE2-depedent milRNA based on qRT-PCR and Northern blot analysis. Compared to the wild-type, the deletion mutant of milR-87 was significantly reduced in virulence, while overexpression of milR-87 enhanced disease severity, confirming that milR-87 is crucial for Foc virulence in the infection process. We furthermore identified FOIG_15013 (a glycosyl hydrolase-coding gene) as the direct target of milR-87 based on the expression of FOIG_15013-GFP fusion protein. The FOIG_15013 deletion mutant displayed similar phenotypes as the overexpression of milR-87, with a dramatic increase in the growth, conidiation and virulence. Transient expression of FOIG_15013 in Nicotiana benthamiana leaves activates the host defense responses. Collectively, this study documents the involvement of milRNAs in the manifestation of the devastating fungal disease in banana, and demonstrates the importance of milRNAs in the pathogenesis and other biological processes. Further analyses of the biosynthesis and expression regulation of fungal milRNAs may offer a novel strategy to combat devastating fungal diseases. 相似文献
