摘要:【目的】筛选一株可以将琼胶转化为新琼寡糖的菌株,并对该菌株进行鉴定。【方法】从紫菜生长区域采集紫菜和该区域海水,用含1‰琼胶的培养基富集培养,逐级稀释涂布、平板划线进行初筛,液体培养进行复筛,DNS法测定琼胶降解产物中还原糖的含量。通过16S rDNA序列分析,结合菌体形态、菌落特征及生理生化特性,确立该菌的系统发育学地位。【结果】从紫菜振荡液中筛选出一株可以产琼胶酶的菌株HJPHYXJ-1,该菌属于革兰氏阴性菌,16S rDNA序列同源性与需钠弧菌(Vibrio natriegens)的达到了99%,结合形态特征和生理生化实验结果鉴定该菌为需钠弧菌。HPLC法测定酶解产物为新琼寡糖。【结论】HJPHYXJ-1被筛选用于转化琼胶,酶解产物的聚合度在2-12 之间,是以二糖为单位的新琼寡糖,该菌产生的酶为β-琼胶酶。 相似文献

Human papilloma virus type 16 (HPV16) E7 is a viral oncoprotein that is believed to play a major role in cervical neoplasia.
A novel antagonist peptide against HPV16 E7 was previously selected by phage display screening and the selected peptide was
found to have anti-tumor efficacy against HPV16-positive cervical carcinoma through induction of cell cycle arrest. In the
current study, to further elucidate the mechanisms of the antagonist peptide, the effects of the peptide on apoptosis are
investigated by RT-PCR, Western blotting, MTT assay, TUNEL staining, Annexin V apoptosis assay, flow cytometry, and animal
experiments. The antagonist peptide showed obvious anti-tumor efficacy through apoptosis induction, both in HPV16-positive
cervical cancer cell lines and tumor xenografts. Our results also revealed that the peptide induced accumulation of cellular
p53 and p21, and led to HPV16 E7 protein degradation. In the case of mRNA levels, it resulted in unaltered p53 and HPV16 E7
expression, but increased expression of p21. In contrast, the induction of apoptosis and p53 reactivation effects by the selected
peptide were abolished after E7 knocked down with siRNA. These results demonstrate that the selected peptide can induce E7
degradation and lead to marked apoptosis in HPV16-related cancer cells by activating cellular p53 and its target genes, such
as p21. Furthermore, the evident therapeutic efficacy obtained from the subcutaneous tumor model experiments in nude mice
suggests a therapeutic potential for HPV16-related cancers of the selected peptide. Therefore, this specific peptide may be

The castor-bean endosperm-the best-studied material of reserve lipid hydrolysis in seed germination-was previously shown to have an acid lipase and an alkaline lipase having reciprocal patterns of development during germination. We studied oil seeds from 7 species, namely castor bean (Ricinus communis L.), peanut (Arachis hypogaea L.), sunflower (Helianthus annus L.), cucumber (Cucumis sativus L.), cotton (Gossypisum hirsutum L.), corn (Zea mays. L.) and tomato (Lycopersicon esculentum Mill.). The storage tissues of all these oil seeds except castor bean contained only alkaline lipase activity which increased drastically during germination. The pattern of acid and alkaline lipases in castor bean does not seem to be common in other oil seeds. The alkaline lipase of peanut cotyledons was chosen for further study. On sucrose gradient centrifugation of cotyledon homogenate from 3-d-old seedlings, about 60% of the activity of the enzyme was found to be associated with the glyoxysomes, 15% with the mitochondria, and 25% with a membrane fraction at a density of 1.12 g cm-3. The glyoxysomal lipase was associated with the organelle membrane, and hydrolyzed only monoglyceride whereas the mitochondrial and membrane-fraction enzymes degraded mono-, di- and triglycerides equally well. Thus, although the lipase in the glyoxysomes had the highest activity, it had to cooperate with lipases in other cellular compartments for the complete hydrolysis of reserve triglycerides. 相似文献

We describe the synthesis, characterization, and reactivity of several Ru(II) complexes of the type cis-L2Ru(Z)n+, where L is an α-diimine [e.g. 2,2′-bipyridine (bpy) or 1,10-phenanthroline (phen)] ligand and Z is a bis-coordinated scorpionate ligand such as tris-(1-pyrazolyl)methane (HC(pz)3, PZ=1-pyrazolyl; n=2) or tetrakis-(1-pyrazolyl)borate anion (B(pz)4 −; n=1). The complexes each exhibit strong visible absorption assigned as a π*(L)←dπ(Ru) metal-to-ligand charge-transfer (MLCT) transition characteristic of the cis-L2Ru2+ kernel. A corresponding MLCT excited state emission is observed in room temperature CH3CN solution, although emission energies, lifetimes, and quantum yields are reduced relative to Ru(bpy)3 2+. Electronic spectra and cyclic voltammetry measurements indicate that the relative π-acceptor abilities of the coordinated Z are: Z=(1H-pyrazolyl)2(pz)2B(pz)2<(pyridine)2<(pz)2CH(pz). Uncoordinated pz groups of cis-(bpy)2Ru(pz)2B(pz)2 + can be reacted to form a sterically hindered, localized-valence (Kcom33 l mol−1) cis,cis-(bpy)2RuII(pz)2B(pz)2RuII(bpy)2 3+ dimer. The dimer properties are interpreted by comparison to the known cis-(bpy)2RuII(pz)2RuII(bpy)2 2+ analog. The dimer is photoreactive and undergoes an asymmetrical photocleavage in CH3CN (yielding cis-(bpy)2RuIII(pz)2B(pz)2 2+ and cis-(bpy)2RuII(CH3CN)2 2+), similar to the corresponding thermal reaction observed for the mixed-valence cis-(bpy)2RuII(pz)2RuIII(bpy)2 3+ system. 相似文献

Bladder cancer is one of the most common malignant tumors in the urinary system. The development and improvement of treatment efficiency require the deepening of the understanding of its molecular mechanism. This study investigated the role of ALPK2, which is rarely studied in malignant tumors, in the development of bladder cancer. Our results showed the upregulation of ALPK2 in bladder cancer, and data mining of TCGA database showed the association between ALPK2 and pathological parameters of patients with bladder cancer. In vitro and in vivo experiments demonstrated that knockdown of ALPK2 could inhibit bladder cancer development through regulating cell proliferation, cell apoptosis, and cell migration. Additionally, DEPDC1A is identified as a potential downstream of ALPK2 with direct interaction, whose overexpression/downregulation can inhibit/promote the malignant behavioral of bladder cancer cells. Moreover, the overexpression of DEPDC1A can rescue the inhibitory effects of ALPK2 knockdown on bladder cancer. In conclusion, ALPK2 exerts a cancer-promoting role in the development of bladder cancer by regulating DEPDC1A, which may become a promising target to improve the treatment strategy of bladder cancer.Subject terms: Cancer models, Bladder cancer 相似文献

Insect pathogenic fungi like Beauveria bassiana have been developed as environmentally friendly biocontrol agents against arthropod pests. However, restrictive environmental factors, including solar ultraviolet (UV) radiation frequently lead to inconsistent field performance. To improve resistance to UV damage, we used Agrobacterium-mediated transformation to engineer B. bassiana with an exogenous tyrosinase gene. The results showed that the mitotically stable transformants produced larger amounts of yellowish pigments than the wild-type strain, and these imparted significantly increased UV-resistance. The virulence of the transgenic isolate was also significantly increased against the silkworm Bombyx mori and the mealworm Tenebrio molitor. This study demonstrated that genetic engineering of B. bassiana with a tyrosinase gene is an effective way to improve fungal tolerance against UV damage. 相似文献

The global asymptotic stability of impulsive stochastic Cohen–Grossberg neural networks with mixed delays and reaction–diffusion terms is investigated. Under some suitable assumptions and using Lyapunov–Krasovskii functional method, we apply the linear matrix inequality technique to propose some new sufficient conditions for the global asymptotic stability of the addressed model in the stochastic sense. The mixed time delays comprise both the time-varying and continuously distributed delays. The effectiveness of the theoretical result is illustrated by a numerical example. 相似文献

Evaluation of cross-sectional geometry and mass density distributions of humans and laboratory animals using computerized tomography 总被引:1,自引:0,他引:1
The purpose of this study was to determine the cross-sectional geometry and mass density distribution of a young porcine subject using the X-ray computerized tomographic (CT) method and to perform a comparative study of anatomical features of this subject and a 3 yr old female child specimen. The cross-sectional CT scans of the porcine subject were obtained at 1 cm intervals. The outlines of each cross section and of selected anatomical components within each section were obtained by standard picture processing techniques. The mass and inertia tensor for each cross section and for each anatomical structure in a section were computed based on the CT numbers. The porcine subject was then sacrificed, frozen, sectioned and photographed. These sectional photographs were then compared with those obtained from the CT method. Tabulated cross-sectional mass and inertia tensor obtained from CT scans of the porcine subject were also used to compare with similar results derived from previously completed CT scans of the 3 yr old female child specimen. In particular, the comparisons were made on the location of the center of gravity and the inertia tensor in the head, neck, head and neck and cervical spine regions. Some immediate applications of this data are inputs to finite element models, lumped parameter biodynamic models, computer simulation of vehicle crash victims, and dummy design. 相似文献

【目的】研究可降解成年泌乳奶牛粪中主要酸臭物的微生物群落的组成及动态变化。【方法】利用牛粪堆肥环境中的微生物进行了发酵优化、菌种驯化以及酸臭有机物降解规律的研究,结合r DNA高通量测序技术对有益微生物的组成及相对生物量进行了分析。【结果】实验发现,奶牛排泄物中的臭味来源主要为短链有机酸,堆肥自然环境中的微生物可以有效地对有机酸等污染物进行去除,经从低到高浓度的有机酸臭物(W/V,0.1%–0.2%)驯化发酵后,培养物中原核微生物以芽孢杆菌居多,而真核微生物主要由红曲霉及粉状毕赤酵母组成。【结论】进一步推测这几种微生物是耐受并降解有机酸臭物的优势微生物,可以应用于奶牛养殖过程中酸臭排泄物的生物控制。 相似文献

This study was carried out to investigate whether shark hepatic stimulator substance (HSS) can prevent acute liver injury and affect mitochondrial function and antioxidant defenses in a rat model of thioacetamide (TAA)-induced liver injury. The acute liver injury was induced by two intraperitoneal injections of TAA (400 mg/kg) in a 24 h interval. In the TAA plus shark HSS group, rats were treated with shark HSS (80 mg/kg) 1 h prior to each TAA injection. In this group, serum liver enzyme activities were significantly lower than those in the TAA group. The mitochondrial respiratory control ratio was improved, and the mitochondrial respiratory enzyme activities were increased in the TAA plus shark HSS group. The mitochondrial antioxidant enzyme activities and glutathione level were higher in the TAA plus shark HSS group than in the TAA group. These results suggest that the protective effect of shark HSS against TAA-induced acute liver injury may be a result of the restoration of the mitochondrial respiratory function and antioxidant defenses and decreased oxygen stress. 相似文献
