In the SV40-transformed mouse embryo fibroblast cell line SVT2/S, genomic rearrangements involving the SV40 DNA and flanking host sequences were identified by Southern blot hybridization using viral DNA as probe. No rearrangements of SV40 DNA integrated into nonpermissive mouse cells have been previously described. The standard arrangement found in the majority of subclones was mapped with 20 restriction enzymes, 10 of which cleave sites within the SV40 DNA. A single copy of a defective integrated viral genome is present, in which the late region is missing from about nucleotide 200 clockwise to about nucleotide 1750. The rest of the viral genome including the origin of replication and T antigen binding region is present and colinear with SV40 DNA, except for an internal repeat of about 1750 bp located between nucleotides 2750 and 4500. Rearrangements were found in 4 out of 20 random subclones of the parental SVT2/S cell line and 3 of the 4 continued to rearrange. The thioguanine-resistant cell line 281-1-4, derived from SVT2/S, remained stable on subculture but a chloramphenicol-resistant mutant, 107-6-4, derived from 281-1-4, was highly unstable. In 107-6-4, unique rearrangements were found in 6 of 31 subclones of a population that had undergone abut 25 doublings from a single-cell isolate. The high rate of rearrangement and the sporadic expression of rearrangement potential are characteristic of the transposable controlling elements discovered by McClintock. 相似文献

A model is developed to predict the changes in total capacitance (i.e. total charge stored divided by surface membrane potential) of the tubular system of muscle fibers. The tubular system is represented as a punctated disc and the area of membrane across which current flows is represented as a punctated annulus, the capacitance of the muscle fiber being proportional to this area. The area can be determined from a distributed model of the tubular system, in which the only resistance to radial current flow is presumed to be in the lumen of the tubules. Calculations are made of the variation of capacitance expected as the conductivity of the bathing solution is varied. These calculations include the effects of fixed charge in the tubular lumen and the effects of changes in the shape and volume of the tubular system in solutions of low conductivity. The calculated results fail to fit comparable experimental data, although they do qualitatively account for the known variation of the radial spread of contraction with conductivity of the bathing medium. It is pointed out that the existence of a significant "access resistance" at the mouth of the tubules might explain the discrepancy between theory and experiment. 相似文献

The coagulation of isoelectric egg albumin solutions, on exposure to ultraviolet radiation, involves three distinct processes, (1) the light denaturation of the albumin molecule, (2) a reaction between the light denatured molecule and water which may be similar to heat denaturation but occurs at a lower temperature, and (3), the flocculation of the denatured molecules to form a coagulum. The light denaturation is unimolecular, independent of temperature, and occurs over a wide pH range. The reaction between the light denatured molecule and water has a temperature coefficient of 10+ and occurs rapidly at 40°C., a temperature at which heat denaturation is inappreciable. 相似文献

Evidence is brought forward to show that at concentrations of urea high enough to split the egg albumin molecule the solubility changes produced by urea are profoundly modified. The degree of precipitation after dialysis is the net result of two changes produced by the urea: the first, normally spoken of as denaturation, which makes the protein insoluble in dilute solution and the second, a splitting of the molecule which makes it soluble. These two reactions may proceed independently and simultaneously or the second reaction may follow the first, taking place in the denatured molecule only. In view of the decrease in the opalescence with time, the latter process is more probable. Both of these reactions have positive temperature coefficients, but as the concentration of urea increases the second reaction is more affected by increase in temperature than the first, and consequently the resulting opalescence decreases rather than increases with temperature. This accounts for and explains reports of negative temperature coefficients of denaturation, when denaturation is measured by the amount of insoluble material found on dilution. The occurrence of these two reactions, one leading to an increase and the other to a decrease in the amount of insoluble protein, should be taken into account when denaturation changes in egg albumin with urea are studied. 相似文献
