Microbes in the deep vadose zone play an essential role in the mitigation of nitrate leaching; however, limited information is available on the mechanisms of microbial denitrification due to sampling difficulties. We experimentally studied the factors that affect denitrification in soils collected down to 10.5 meters deep along the soil profile. After an anoxic pre‐incubation, denitrification rates moderately increased and the N2O/(N2O + N2) ratios declined while the microbial abundance and diversity did not change significantly in most of the layers. Denitrification rate was significantly enhanced and the abundance of the denitrification genes was simultaneously elevated by the increased availability of organic carbon in all studied layers, to a greater extent in the subsurface layers than in the surface layers, suggesting the severe scarcity of carbon in the deep vadose zone. The genera Pseudomonas and Bacillus, which are made up of a number of species that have been previously identified as denitrifiers in soil, were the major taxa that respond to carbon addition. Overall, our results suggested that the limited denitrification in the deep vadose zone is not because of the lack of denitrifiers, but due to the low abundance of denitrifiers which is caused by low carbon availability.  相似文献

Tumour necrosis factor alpha (TNFα) and interferon gamma (IFNγ) were originally found to be produced by inflammatory cells and play important roles in the immune system and surveillance of tumour growth. By activating distinct signalling pathways of nuclear factor‐κB (NF‐κB), mitogen‐activated protein kinase (MAPK), and JAK/STAT, TNFα and IFNγ were reported to effectively trigger cell death and perform powerful anti‐cancer effects. In this review, we will discuss the new advancements of TNFα and IFNγ in anti‐cancer therapy.  相似文献

Micro/macrowire intracranial EEG (iEEG) signals recorded from implanted micro/macroelectrodes in epileptic patients have received great attention and are considered to include much information of neuron activities in seizure transition compared to scalp EEG from cortical electrodes. Microelectrode is contacted more close to neurons than macroelectrode and it is more sensitive to neuron activity changes than macroelectrode. Microwire iEEG recordings are inevitably advantageous over macrowire iEEG recordings to reveal neuronal mechanisms contributing to the generation of seizures. In this study, we investigate the seizure generation from microwire iEEG recordings and discuss synchronization of microwire iEEGs in four frequency bands: alpha (1−30 Hz), gamma (30−80 Hz), ripple (80–250 Hz), and fast ripple (>250 Hz) via two measures: correlation and phase synchrony. We find that an increase trend of correlation or phase synchrony exists before the macroseizure onset mostly in gamma and ripple bands where the duration of the preictal states varied in different seizures ranging up to a few seconds (minutes). This finding is contrast to the well-known result that a decrease of synchronization in macro domains exists before the macroseizure onset. The finding demonstrates that it is only when the seizure has recruited enough surrounding brain tissue does the signal become strong enough to be observed on the clinical macroelectrode and as a result support the hypothesis of progressive coalescence of microseizure domains. The potential ramifications of such an early detection of microscale seizure activity may open a new window on treatment by making possible disruption of seizure activity before it becomes fully established.  相似文献

Many metabolites in plant are highly polar and ionic. Their analysis using gas chromatography–mass spectrometry and liquid chromatography–mass spectrometry can be problematic. Therefore a capillary electrophoresis–mass spectrometry (CE–MS) method with charge-driven separation characteristic was developed to investigate polar metabolites in tobacco. To obtain as many features as possible, extraction of polar metabolites was optimized by the design of experiments and evaluated by univariate statistics. Method validation was carried out to evaluate the analytical characteristics including calibration curve, precision, sample stability and extraction reproducibility. The developed method was successfully applied in studying 30 tobacco leaves obtained from Yunnan and Guizhou provinces in China. A total of 154 polar metabolites were identified based on available database. Multivariate pattern recognition clearly revealed the metabolic differences between the two geographic areas and 43 significantly different metabolites were defined by the non-parametric hypothesis test (Mann–Whitney U test) and false discovery rate. Some key metabolites involved in photosynthesis such as ribulose 1,5-disphosphate, fructose 1,6-diphosphate, glycine, betaine, GABA and serine were found to be susceptible to environmental conditions. This study shows that the metabolic profiling based on CE–MS can clearly discriminate tobacco leaves of different geographical origins and understand the relationship between plant metabolites and their geographical origins.  相似文献

以东居延海为研究对象,利用遥感技术目视解译ETM影像,提取东居延海2002—2012年各月湖面面积。通过水文保证率法确定不同保证率下的湖面面积,结合额济纳旗气象站观测的风速、相对湿度、气温、水汽压、降水量等气象数据估算湖泊蒸发耗水量和湖泊降水补给量,根据湖泊渗漏系数估算湖泊渗漏量,最后运用水平衡原理构建湖泊生态需水模型,估算了东居延海在湖面面积保证率为50%、75%、95%时各月月均和年均生态需水量,其中年均生态需水量分别为1.78×108、1.60×108、1.03×108m3,约占莺落峡年均径流量的9.66%、8.66%、5.59%,约占正义峡年均径流量的16.27%、14.60%、9.42%,约占狼心山年均径流量的30.81%、27.65%、17.84%。  相似文献

Heme oxygenase (HO) represents an intrinsic antiinflammatory system based on its ability to inhibit expression of proinflammatory cytokines. The constitutive isoform heme oxygenase-2 (HO-2) has high expression and activity in cerebral microvascular endothelial cells (CMVEC). This study was undertaken to evaluate the role of HO-2 in regulation of TLR4/MyD88-dependent signaling and to study the effect of HO-2 on the expression and secretion of the proinflammatory cytokines tumor necrosis factor α (TNF-α) and Interleukin-6 (IL6) in CMVEC. HO-2 short hairpin RNA (shRNA) and HO-2 overexpression plasmids were used to observe the effect of HO-2 on proinflammatory cytokines in CMVEC in vitro, and the results showed that the messenger RNA (mRNA) and protein levels of TNF-α and IL6 were increased and decreased, respectively, compared with control groups. LPS-stimulated TNF-α and IL6 mRNA and protein were also reduced in CMVEC treated with an inhibitor of TLR4 signaling, CLI-095, or HO-2 overexpression. CLI-095 and HO-2 overexpression both reduced TLR4 expression in CMVEC, and HO-2 shRNA blocked these effects of CLI-095. CLI-095 and HO-2 overexpression potently suppressed TLR4/MyD88-dependent proinflammatory cytokine expression in CMVEC. These results suggest that HO-2 plays an important role in protecting CMVEC against cytokine-mediated inflammation.  相似文献

Previous studies indicate that the H3 influenza virus has the ability to establish infection upon interspecies transmission and poses a threat to mammals. Therefore, it is important to enhance the surveillance of H3 avian influenza viruses (AIVs). In this study, A/duck/Shanghai/C84/2009(H3N2) (C84) was isolated from a live poultry market in Shanghai, China. Using PCR and sequencing analyses, we obtained the whole-genome sequence of this virus. The H3N2 virus proved to be a novel multiple-gene reassortant AIV whose genes were derived from H3N2, H4N6, H6N2, and H9N2. Knowledge regarding the complete genome sequence of the C84 virus will be useful for epidemiological surveillance.  相似文献

Cerebral glucose metabolism is a reliable index of neural activity and may provide evidence for brain function in healthy adults. We studied the correlation between cerebral glucose metabolism and age under the resting-state in both sexes with position emission tomography. Statistical test of age effect on cerebral glucose metabolism was performed using the statistical parametric mapping software with a voxel-by-voxel approach ( family wise error corrected, -voxel threshold). The subjects consisted of 108 females (mean S.D. = 4510 years) and 126 males (mean S.D. = 4911 years). We showed here that brain activity in the frontal and temporal lobes in both sexes decreased significantly with normal aging. The glucose metabolism in the caudate bilaterally showed a negative correlation with age in males, but not in females. Few regions in males were shown with an increased glucose metabolism with age. Although the mechanisms of brain aging are still unknown, a map of brain areas susceptible to age was described in this report.  相似文献

Background: Helicobacter pylori infection is a major cause of gastritis and gastric carcinoma. Aspirin has anti‐inflammatory and antineoplastic activity. The aim of the present study was to determine the effects of aspirin on H. pylori‐induced gastritis and the development of heterotopic proliferative glands. Methods: H. pylori strain SS1 was inoculated into the stomachs of Mongolian gerbils. Two weeks after inoculation, the animals were fed with the powder diets containing 0 p.p.m. (n = 10), 150 p.p.m. (n = 10), or 500 p.p.m. (n = 10) aspirin. Mongolian gerbils were killed after 36 weeks of infection. Uninfected Mongolian gerbils (n = 10) were used as controls. Histologic changes, epithelial cell proliferation and apoptosis, and prostaglandin E2 (PGE2) levels of gastric tissue were determined. Results: H. pylori infection induced gastric inflammation. Administration of aspirin did not change H. pylori‐induced gastritis, but alleviated H. pylori‐induced hyperplasia and the development of heterotopic proliferative glands. Administration of aspirin accelerated H. pylori‐associated apoptosis but decreased H. pylori‐associated cell proliferation. In addition, the increased gastric PGE2 levels due to H. pylori infection were suppressed by treatment with aspirin, especially at the dose of 500 p.p.m. Conclusions: Aspirin alleviates H. pylori‐induced hyperplasia and the development of heterotopic proliferative glands. Moreover, aspirin increases H. pylori‐induced apoptosis. We demonstrated the antineoplastic activities of aspirin in H. pylori‐related gastric carcinogenesis.  相似文献
