We examined whether nitric oxide (NO) generated from neuronal NO synthase (nNOS) contributes to the reduced ability of the newborn to autoregulate retinal blood flow (RBF) and choroidal blood flow (ChBF) during acute rises in perfusion pressure. In newborn pigs (1-2 days old), RBF (measured by microsphere) is autoregulated over a narrow range of perfusion pressure, whereas ChBF is not autoregulated. N(G)-nitro-L-arginine methyl ester (L-NAME) or specific nNOS inhibitors 7-nitroindazole, 3-bromo-7-nitroindazole, and 1-(2-trifluoromethyl-phenyl)imidazole as well as ganglionic blocker hexamethonium, unveiled a ChBF autoregulation as observed in juvenile (4- to 6-wk old) animals, whereas autoregulation of RBF in the newborn was only enhanced by L-NAME. All NOS inhibitors and hexamethonium prevented the hypertension-induced increase in NO mediator cGMP in the choroid. nNOS mRNA expression and activity were three- to fourfold higher in the choroid of newborn pigs than in tissues of juvenile pigs. It is concluded that increased production of NO from nNOS curtails ChBF autoregulation in the newborn and suggests a role for the autonomic nervous system in this important hemodynamic function, whereas, for RBF autoregulation, endothelial NOS seems to exert a more important contribution in limiting autoregulation.  相似文献

To react with peptides, nitric oxide.NO has to be activated by oxidation, or by coupling with superoxide (O.-2) thereby producing peroxynitrite. In the course of.NO oxidation,.NO2 free radicals and N2O3 may be formed. Using gamma-irradiation methods, we characterized the products formed by these nitrogen oxides with angiotensin II. Angiotensin II is specifically nitrated at its tyrosinyl residue by.NO2 or peroxynitrite. Equimolecular amounts of each reagent in K+/Pi solutions at pH 7.4 led to 56% and 5% nitration yields, respectively. Nitrogen oxides produced by autoxidation of.NO, as well as.NO2 under.NO, reacted only with the arginine residue, giving a mixture of peptides containing citrulline, a N-(hydroxylamino-cyanamido-) instead of guanido group, and a conjugated diene derived from an arginine side-chain. However, nitrosation reactions by N2O3 occurred only when the initial concentration of.NO2 was 10 times that able to react with angiotensin II. Thus, in this case.NO appears to protect against.NO2 action.  相似文献

Protoplasts were isolated by enzyme digestion from leaf of Japanese butterbur (Petasites japonicus). The enzyme incubation mixture consisted of 4% (W/V)cellulase RS, 2% (W/V) hemicellulase, 1% (W/V) pectinase-dissolved Y-23 and polygalacturonase in a solution of 0. 5 mol/L mannitol at pH 5.7 . In the basic medium of 1/4 MS inorganic salts and 1/2 MS vitamins supplemented with 2 mg/L NAA, 0. 2–0. 5 mg/L BA, 0. 5 mol/L mannitol and 10 g/L sucrose, the cells divided luxuriantly. Two and ten compounds were isplated from the water portion and the ethereal soluble lipophilic fraction of Ganoderma lucidum respectively. On the basis of their chemical properties and co-TLC and IR spectral analysis, the two compounds from the water portion were identified as choline (Ⅰ) and betains (Ⅱ), ten compounds from thereal soluble fraction as tetra- cosanoic acid, stearic acid, palmitic acid, ergosta-7, 22-diene-3β-ol, nonadecanoic acid, benenic acid, tetracosane, hentriacontane, ergosterol and β-sitosterol.
