Many structural, signaling, and adhesion molecules contain tandemly
repeated amino acid motifs. The alpha-actinin/spectrin/dystrophin
superfamily of F-actin-crosslinking proteins contains an array of triple
alpha-helical motifs (spectrin repeats). We present here the complete
sequence of the novel beta-spectrin isoform beta(Heavy)- spectrin (beta H).
The sequence of beta H supports the origin of alpha- and beta-spectrins
from a common ancestor, and we present a novel model for the origin of the
spectrins from a homodimeric actin-crosslinking precursor. The pattern of
similarity between the spectrin repeat units indicates that they have
evolved by a series of nested, nonuniform duplications. Furthermore, the
spectrins and dystrophins clearly have common ancestry, yet the repeat unit
is of a different length in each family. Together, these observations
suggest a dynamic period of increase in repeat number accompanied by
homogenization within each array by concerted evolution. However, today,
there is greater similarity of homologous repeats between species than
there is across repeats within species, suggesting that concerted evolution
ceased some time before the arthropod/vertebrate split. We propose a
two-phase model for the evolution of the spectrin repeat arrays in which an
initial phase of concerted evolution is subsequently retarded as each new
protein becomes constrained to a specific length and the repeats diverge at
the DNA level. This evolutionary model has general applicability to the
origins of the many other proteins that have tandemly repeated motifs.

In hardwood subtropical forests of southern Florida, nonnative vines have been hypothesized to be detrimental, as many species form dense “vine blankets” that shroud the forest. To investigate the effects of nonnative vines in post‐hurricane regeneration, we set up four large (two pairs of 30 X 60 m) study areas in each of three study sites. One of each pair was unmanaged and the other was managed by removal of nonnative plants, predominantly vines. Within these areas, we sampled vegetation in 5 X 5 m plots for stems 2 cm DBH (diameter at breast height) or greater and in 2 X 0.5 m plots for stems of all sizes. For five years, at annual censuses, we tagged and measured stems of vines, trees, shrubs and herbs in these plots. For each 5 X 5 m plot, we estimated percent coverage by individual vine species, using native and nonnative vines as classes. We investigated the hypotheses that: (1) plot coverage, occurrence and recruitment of nonnative vines were greater than that of native vines in unmanaged plots; (2) the management program was effective at reducing cover by nonnative vines; and (3) reduction of cover by nonnative vines improved recruitment of seedlings and saplings of native trees, shrubs, and herbs. In unmanaged plots, nonnative vines recruited more seedlings and had a significantly higher plot‐cover index, but not a higher frequency of occurrence. Management significantly reduced cover by nonnative vines and had a significant overall positive effect on recruitment of seedlings and saplings of native trees, shrubs and herbs. Management also affected the seedling community (which included vines, trees, shrubs, and herbs) in some unanticipated ways, favoring early successional species for a longer period of time. The vine species with the greatest potential to “strangle” gaps were those that rapidly formed dense cover, had shade tolerant seedling recruitment, and were animal‐dispersed. This suite of traits was more common in the nonnative vines than in the native vines. Our results suggest that some vines may alter the spatiotemporal pattern of recruitment sites in a forest ecosystem following a natural disturbance by creating many very shady spots very quickly. 相似文献

Certain mutations in the unc-105 II gene of the nematode Caenorhabditis elegans have dominant effects on morphology and behavior: animals become small, severely hypercontracted and paralyzed. These unc-105 mutants revert both spontaneously and with mutagens at high frequencies to a wild-type phenotype. Most of the reversion events are intragenic, apparently because the null (loss-of-function) phenotype of unc-105 is wild type. One revertant defined an extragenic suppressor locus, sup-20 X. Such suppressor alleles of sup-20 are rare, and the apparent null phenotype of sup-20 is embryonic lethality. By constructing animals genetically mosaic for sup-20, we have shown that the primary effect of sup-20 is in muscle cells. In addition to mutations in sup-20, other mutations causing muscle defects, such as unc-54 and unc-22 mutations, suppress the hypercontracted phenotype of unc-105. The ease of identifying nonhypercontracted revertants of unc-105 mutants greatly facilitates the isolation of new mutants defective in muscle structure and function. 相似文献

To explore the nature of cell lineage modifications that have occurred during evolution, the gonadal cell lineages of the nematode Panagrellus redivivus have been determined and compared to the known gonadal lineages of Caenorhabditis elegans (J. Kimble and D. Hirsh, 1979, Develop. Biol.70, 396–417). Essentially invariant lineages generate the 143 somatic cells of the male gonad and at least 326 somatic cells of the female gonad of P. redivivus. The basic program of gonadogenesis is strikingly similar among both sexes of both species. For example, the early division patterns of the somatic gonad precursors Z1 and Z4 are almost identical. Later division patterns are more divergent and, in a few cases, generate structures that are species specific. In general, similar cell types are produced after similar patterns of cell divisions. Differences among the Z1 and Z4 cell lineages appear to reflect phylogenetic modifications of a common developmental program. The nature of these differences suggests that the evolution of cell lineages involves four distinct classes of alterations: switches in the fate of a cell to that normally associated with another cell; reversals in the polarity of the lineage generated by a blast cell; alterations in the number of rounds of cell division; and an “altered segregation” of developmental potential, so that a potential normally associated with one cell instead becomes associated with its sister. A number of cell deaths occur during gonadogenesis in P. redivivus. The death of Z4.pp, a cell that controls the development of the posterior ovary in C. elegans, probably prevents the development of a posterior ovary in P. redivivus and hence is responsible for the gross difference in the morphologies of the gonads of the P. redivivus female and the C. elegans hermaphrodite. As exemplified by the death of Z4.pp, an alteration in the fate of a “regulatory cell” could facilitate rapid and/or discontinuous evolutionary change. 相似文献

Previous studies have shown that the development of the vulva of the C. elegans hermaphrodite involves six multipotential hypodermal cells as well as the gonadal anchor cell, which induces vulval formation. Our further examination of the interactions among these seven cells has led to the following model. Each hypodermal precursor cell becomes determined to adopt one of its three potential fates; each of these fates is to generate a particular cell lineage. In the absence of cellular interactions each precursor cell will generate the nonvulval cell lineage; an inductive signal from the anchor cell is required for a precursor cell to generate either of the two types of vulval cell lineages. The inductive signal is spatially graded, and the potency of the signal specifies which lineage is expressed by each of the tripotential precursor cells. 相似文献

Background
Despite recent achievements to reduce child mortality, neonatal deaths continue to remain high, accounting for 41% of all deaths in children under five years of age worldwide, of which over 90% occur in low- and middle-income countries (LMICs). Infections are a leading cause of death and limitations in care seeking for ill neonates contribute to high mortality rates. As estimates for care-seeking behaviors in LMICs have not been studied, this review describes care seeking for neonatal illnesses in LMICs, with particular attention to type of care sought.Methods and Findings
We conducted a systematic literature review of studies that reported the proportion of caregivers that sought care for ill or suspected ill neonates in LMICs. The initial search yielded 784 studies, of which 22 studies described relevant data from community household surveys, facility-based surveys, and intervention trials. The majority of studies were from South Asia (n = 17/22), set in rural areas (n = 17/22), and published within the last 4 years (n = 18/22). Of the 9,098 neonates who were ill or suspected to be ill, 4,320 caregivers sought some type of care, including care from a health facility (n = 370) or provider (n = 1,813). Care seeking ranged between 10% and 100% among caregivers with a median of 59%. Care seeking from a health care provider yielded a similar range and median, while care seeking at a health care facility ranged between 1% and 100%, with a median of 20%. Care-seeking estimates were limited by the few studies conducted in urban settings and regions other than South Asia. There was a lack of consistency regarding illness, care-seeking, and care provider definitions.Conclusions
There is a paucity of data regarding newborn care-seeking behaviors; in South Asia, care seeking is low for newborn illness, especially in terms of care sought from health care facilities and medically trained providers. There is a need for representative data to describe care-seeking patterns in different geographic regions and better understand mechanisms to enhance care seeking during this vulnerable time period. MicroRNAs (miRNAs) are highly expressed in vertebrate neural tissues, but the contribution of specific miRNAs to the development and function of different neuronal populations is still largely unknown. We report that miRNAs are required for terminal differentiation of olfactory precursors in both mouse and zebrafish but are dispensable for proper function of mature olfactory neurons. The repertoire of miRNAs expressed in olfactory tissues contains over 100 distinct miRNAs. A subset, including the miR-200 family, shows high olfactory enrichment and expression patterns consistent with a role during olfactory neurogenesis. Loss of function of the miR-200 family phenocopies the terminal differentiation defect observed in absence of all miRNA activity in olfactory progenitors. Our data support the notion that vertebrate tissue differentiation is controlled by conserved subsets of organ-specific miRNAs in both mouse and zebrafish and provide insights into control mechanisms underlying olfactory differentiation in vertebrates. 相似文献

Heparin and its derivatives bind to HIV-1 recombinant envelope glycoproteins, rather than to recombinant HIV-1 receptor, CD4 总被引:1,自引:0,他引:1
We have employed a direct radiolabel binding assay to investigate the
interaction between3H-heparin and recombinant envelope glycoproteins,
rgp120s, derived from several different isolates of HIV-1. Comparable
dose-dependent binding is exhibited by rgp120s from isolates IIIB, GB8, MN
and SF-2. Under identical experimental conditions the binding of3H- heparin
to a recombinant soluble form of the cellular receptor for gp120, CD4, is
negligible. The binding of3H-heparin to rgp120 is competed for by excess
unlabeled heparin and certain other, but not all, glycosaminoglycan and
chemically modified heparins. Of a range of such polysaccharides tested,
ability to compete with3H-heparin for binding was strictly correlated with
inhibition of HIV-1 replication in vitro. Those possessing potent
anti-HIV-1 activity were effective competitors, whereas those having no or
little anti-HIV-1 activity were poor competitors. Scatchard analysis
indicates that the K d of the interaction between heparin and rgp120 is 10
nM. Binding studies conducted in increasing salt concentrations confirm
that the interaction is ionic in nature. Synthetic 33-35 amino acid
peptides based on the sequence of the V3 loop of gp120 also bind to heparin
with high affinity. V3 loop peptides that are cyclized due to terminal
cysteine residues show more selective binding than their uncyclized
counterparts. Overall, these data demonstrate further that heparin exerts
its anti-HIV-1 activity by binding to the envelope glycoprotein of HIV-1,
rather than its cellular receptor, CD4. This study confirms that the V3
loop of gp120 is the site at which heparin exerts its anti- HIV-1 activity.
Moreover, it reveals that high affinity binding to heparin is shared by all
four rgp120s examined, despite amino acid substitutions within the V3 loop.
