Global cerebral ischemia and subsequent reperfusion induce early impairment of the vasodilator responses to hypercapnia and vasoactive substances. Nitric oxide (NO) is involved in the regulation of cerebral blood flow (CBF) in both health and disease. The present study was designed to assess possible changes in the cerebrovascular reactivity to NO donors induced by cerebral ischemia-reperfusion in goats. Female goats (n = 9) were subjected to 20 min global cerebral ischemia under halothane/N2O anesthesia. Sixteen additional goats were sham-operated as a control group. One week later the effects of ischemia-reperfusion on relaxations to NO donors sodium nitroprusside (SNP), diethylamine/NO (DEA/NO), diethylenetriamine/NO (DETA/NO), and spermine/NO (SPER/NO) were studied in rings of middle cerebral artery (MCA) isolated in an organ bath for isometric tension recording. SNP, DEA/NO, DETA/NO, and SPER/NO induced concentration-dependent relaxations of MCA precontracted with KCl (DEA/NO > SPER/NO > SNP > DETA/NO) or with endothelin-1 (DEA/NO > SNP > SPER/NO > DETA/NO). Relaxations were always higher in endothelin-1-precontracted arteries. One week after cerebral ischemia concentration-response curves to SNP and DEA/NO were displaced to the right, indicating a reduction in relaxant potency of NO donors. The classical nitrovasodilator SNP and NONOates induce relaxation of isolated goat MCA which is partially inhibited by arterial depolarization. Global cerebral ischemia followed by reperfusion induces delayed impairment of the relaxant effects of NO on cerebrovascular smooth muscle, which results in reduced vasodilatory potency of NO donors in large cerebral arteries. 相似文献

Owing that Oreochromis niloticus is resistant to different aquatic environments, four contaminated sites were selected: decentralized engineered ecosystem (DEE) and conventional wastewater treatment plants (WWTP); urban lagoon; and gasoline-contaminated marshland. Endocrine disruption was assessed by alkali-labile phosphate (ALP) quantification, genotoxicity by micronuclei frequency, and morphological changes by hepatosomatic and gonadosomatic indexes. The ALP baseline of non-exposed O. niloticus males was low, compared with other fish species in the literature. Endocrine disruption was observed in fish exposed to raw wastewater of WWTP and DEE, discharge point of channeled river in the urban lagoon, and water from gasoline-contaminated marshland. After treatment in the DEE, the endocrine disruption effect was removed. The micronuclei frequency corroborated with the ALP results in most cases and proved to be a useful tool to assess genotoxicity in fish. The exposure time of 15 days was not enough to provoke morphological effects in most samples. However, in all gasoline-contaminated samples, the fishes showed an increase in the hepatosomatic index followed by a decrease in the gonadosomatic index. The tested biomarkers showed feasibility, sensibility, reproducibility, and were complementary in the assessment of chronic ecotoxicity; therefore, we recommend them to compose a suitable protocol for aquatic monitoring programs. 相似文献

During abundance surveys of the nine-armed sea star Luidia senegalensis (Lamark, 1816) in shallow waters north of Cubagua Island, Venezuela, novel locomotive behaviour was recorded in one sea star. The animal was observed to ‘walk’ with four arms in contact with the benthos whilst the remaining five were held erect. This previously unreported means of locomotion may be: for chemosensory detection of the surroundings; to alter the characteristic track pattern left in sediments as a means to avoid detection by predators; or, to lift the arms to attract commensal cleaners, thus eliminating epibiont commensals that may occupy the ambulacral groove. Whilst the predators of L. senegalensis remain unknown, earlier observations of serranid fish biting this species may support the notion of this behaviour as a predator avoidance strategy. 相似文献

Megazol (7) is a 5-nitroimidazole that is highly active against Trypanosoma
cruzi and Trypanosoma brucei, as well as drug-resistant
forms of trypanosomiasis. Compound 7 is not used clinically due to its mutagenic and
genotoxic properties, but has been largely used as a lead compound. Here, we compared
the activity of 7 with its 4H-1,2,4-triazole bioisostere (8) in
bloodstream forms of T. brucei and T. cruzi and
evaluated their activation by T. brucei type I nitroreductase
(TbNTR) enzyme. We also analysed the cytotoxic and genotoxic
effects of these compounds in whole human blood using Comet and fluorescein
diacetate/ethidium bromide assays. Although the only difference between 7 and 8 is
the substitution of sulphur (in the thiadiazole in 7) for nitrogen (in the triazole
in 8), the results indicated that 8 had poorer antiparasitic activity than 7 and was
not genotoxic, whereas 7 presented this effect. The determination of Vmax indicated
that although 8 was metabolised more rapidly than 7, it bounds to the
TbNTR with better affinity, resulting in equivalent kcat/KM
values. Docking assays of 7 and 8 performed within the active site of a homology

Yarrowia lipolytica is a dimorphic oleaginous non-conventional yeast widely used as a powerful host for expressing heterologous proteins, as well as a promising source of engineered cell factories for various applications. This microorganism has a documented use in Feed and Food and a GRAS (generally recognized as safe) status. Moreover, in vivo studies demonstrated a beneficial effect of this yeast on animal health. However, despite the focus on Y. lipolytica for the industrial manufacturing of heterologous proteins and for probiotic effects, its potential for oral delivery of recombinant therapeutic proteins has seldom been evaluated in mammals. As the first steps towards this aim, we engineered two Y. lipolytica strains, a dairy strain and a laboratory strain, to produce the model fluorescent protein mCherry. We demonstrated that both Y. lipolytica strains transiently persisted for at least 1 week after four daily oral administrations and they maintained the active expression of mCherry in the mouse intestine. We used confocal microscopy to image individual Y. lipolytica cells of freshly collected intestinal tissues. They were found essentially in the lumen and they were rarely in contact with epithelial cells while transiting through the ileum, caecum and colon of mice. Taken as a whole, our results have shown that fluorescent Y. lipolytica strains constitute novel tools to study the persistence and dynamics of orally administered yeasts which could be used in the future as oral delivery vectors for the secretion of active therapeutic proteins in the gut. 相似文献

Premise
Plant lineages differ markedly in species richness globally, regionally, and locally. Differences in whole-genome characteristics (WGCs) such as monoploid chromosome number, genome size, and ploidy level may explain differences in global species richness through speciation or global extinction. However, it is unknown whether WGCs drive species richness within lineages also in a recent, postglacial regional flora or in local plant communities through local extinction or colonization and regional species turnover.Methods
We tested for relationships between WGCs and richness of angiosperm families across the Netherlands/Germany/Czechia as a region, and within 193,449 local vegetation plots.Results
Families that are species-rich across the region have lower ploidy levels and small monoploid chromosomes numbers or both (interaction terms), but the relationships disappear after accounting for continental and local richness of families. Families that are species-rich within occupied localities have small numbers of polyploidy and monoploid chromosome numbers or both, independent of their own regional richness and the local richness of all other locally co-occurring species in the plots. Relationships between WGCs and family species-richness persisted after accounting for niche characteristics and life histories.Conclusions
Families that have few chromosomes, either monoploid or holoploid, succeed in maintaining many species in local communities and across a continent and, as indirect consequence of both, across a region. We suggest evolutionary mechanisms to explain how small chromosome numbers and ploidy levels might decrease rates of local extinction and increase rates of colonization. The genome of a macroevolutionary lineage may ultimately control whether its species can ecologically coexist.499.
