The current standard therapy for chronic HCV infection is a combination of pegylated-interferon (PEG-IFN) and weight-based ribavirin, administered for 24-48 weeks, according to the viral genotype. Although the weekly administration of pegylated interferons provides superior antiviral efficacy over standard interferon alpha, the rate of sustained virological response rarely overpasses 50% in patients infected with HCV genotypes 1 and 4. Consequently, multiple clinical trials with congeners of interferon (consensus interferon, interferon lambda, albinterferon, and controlled-release interferons) are ongoing. Their main advantages consist in maintenance of viral suppression across a longer dosing interval, avoidance of interdose trough and reduced dosing frequencies (twice or even once per month compared to once per week for the actual PEG-IFNs). Along with these superior pharmacokinetic properties, new interferons are expected to have improved side-effect profiles and better tolerability compared with the currently available formulations, providing an option for otherwise difficult to treat, challenging populations. New interferon formulation can be incorporated into future combination with direct acting antivirals, in order to maintain viral suppression over longer periods and minimize the development of viral resistance.  相似文献

Materials and methods: Complete new dentures were made to replace old fractured ‘un’‐reinforced acrylic dentures. The total number of dentures was 30 and woven E‐glass fibre reinforcements were used in maxillary complete dentures. Unidirectional E‐glass fibre reinforcements were used as partial fibre reinforcements in mandibular complete dentures. Ten complete acrylic un‐reinforced dentures were used as control. The follow‐up period was 5 years and the recalls were made at 6 months. Results: After 5 years of wearing the new dentures, the control dentures suffered seven fractures. After 5 years all the mandibular reinforced dentures were in good shape. The maxillary complete reinforced dentures suffered four partial fractures. Fracture lines were restricted by the glass fibre net and the patients could still use their dentures. Conclusion: Pre‐impregnated E‐glass fibre nets and polymer pre‐impregnated E‐glass unidirectional fibres are useful in reinforcing acrylic resin complete dentures especially were heavy occlusal forces are involved. Glass fibre reinforcement will be applied on the tension side in both cases (total fibre reinforcement and partial fibre reinforcement). The reinforcement cannot replace the necessary linings and occlusal adjustments.  相似文献

Mammalian ovarian function is regulated by both hormonal inputs and direct neural influences. Recent studies have shown that, in addition to the extrinsic innervation, the ovaries of nonhuman primates and a strain of rats contain a discrete population of intrinsic neurons. In the present study, we used histological and immunohistochemical approaches to identify the presence of neuronal cell bodies in the fetal and neonatal human ovary. Neurons containing neurofilament immunoreactivity were detected in the hilum and medulla of the ovary at all ages studied, ranging from 24 weeks of gestation to 10 months of postnatal age. Most of them coexpressed the low affinity neurotrophin receptor (p75NTR), and some were catecholaminergic, as determined by their content of immunoreactive tyrosine hydroxylase (TH), the rate-limiting enzyme in catecholamine biosynthesis. The presence of intrinsic neurons in the human ovary, similar to those previously found in other species, indicates that they may be engaged in regulating common, phylogenetically conserved, ovarian functions. It also raises the possibility that their dysfunction may contribute to the manifestation of particular ovarian pathologies.  相似文献

Two Enterococcus strains (E. faecalis and E. faecium) isolated from 2 patients in an intensive care unit (blood and drain, respectively) were analyzed for susceptibility to 4 antibiotics (penicillin, vancomycin, gentamicin, streptomycin) by agar dilution standard method (MICs), time-kill and flow cytometry. We compared the data from classical methods of antibiotic susceptibility detection, that are compulsory 24 hrs long and flow cytometry results at 5 and 24 hrs cultivation. The results from both classical and flow cytometric analyses were highly cogent and revealed the fact that flow cytometry is very useful in early diagnosis of bacterial resistance to antibiotics.  相似文献

The adherence of bacteria to eukaryote cells has been largely investigated as an essential step in the occurrence of bacterial infection. Some clinical and epidemiological studies have revealed the frequent association of certain viral infections with bacterial infections originating in the same ecological niche. Therefore, we investigated the effect of the viral preinfection (ADV4) of some cultivated cells (HEp-2 and IC.SK-27) upon the adherence of staphylococcus to these cells. The analysis of cell adherence within the mentioned conditions, estimated by flow cytometry, allowed of the following conclusions: 1. bacterial adherence to cultivated and virally preinfected cells is augmented by the viral preinfection, and its value on a given cell substrate may characterize a bacterial strain; 2. bacterial adherence to the investigated cell substrates does not correlate with the origin of the tested staphylococcus strains (infections or carriers) and some cell lines can differentiate bacterial strains depending upon the ecological niche or inside it.  相似文献

The balance between pro- and anti-inflammatory signalling is critical to maintain the immune homeostasis under physiological conditions as well as for the control of inflammation in different pathological settings. Recent progress in the signalling pathways that control this balance has led to the development of novel therapeutic agents for diseases characterized by alterations in the activation/suppression of the immune response. Different molecules have a key role in the regulation of the immune system, including the receptors PD-1 (Programmed cell Death 1), CTLA-4 (Cytotoxic T-Lymphocyte Antigen 4) and galectins; or the intracellular enzyme IDO (indoleamine 2,3-dioxygenase). In addition, other molecules as CD69, AhR (Aryl hydrocarbon Receptor), and GADD45 (Growth Arrest and DNA Damage-inducible 45) family members, have emerged as potential targets for the regulation of the activation/suppression balance of immune cells. This review offers a perspective on well-characterized as well as emergent negative immune regulatory molecules in the context of autoimmune inflammatory diseases.  相似文献
