Summary Relationships between root zone temperature, concentrations and uptake rates of NH 4 + and NO 3 were studied in non-mycorrhizal roots of 4-year-old Norway spruce under controlled environmental conditions. Additionally, in a forest stand NH 4 + and NO 3 uptake rates along the root axis and changes in the rhizosphere pH were measured. In the concentration (Cmin) range of 100–150 M uptake rates of NH 4 + were 3–4 times higher than those of NO 3 The preference for NH 4 + uptake was also reflected in the minimum concentration (Cmin) values. Supplying NH4NO3, the rate of NO 3 uptake was very low until the NH 4 + concentrations had fallen below about 100 M. The shift from NH 4 + to NO 3 uptake was correlated with a corresponding shift from net H+ production to net H+ consumption in the external solution. The uptake rates of NH 4 + were correlated with equimolar net production of H+. With NO 3 nutrition net consumption of H+ was approximately twice as high as uptake rates of NO 3 In the forest stand the NO 3 concentration in the soil solution was more than 10 times higher than the NH 4 + concentration (<100 M), and the rhizosphere pH of non-mycorrhizal roots considerably higher than the bulk soil pH. The rhizosphere pH increase was particularly evident in apical root zones where the rates of water and NO 3 uptake and nitrate reductase activity were also higher. The results are summarized in a model of water and nutrient transport to, and uptake by, non-mycorrhizal roots of Norway spruce in a forest stand. Model calculations indicate that delivery to the roots by mass flow may meet most of the plant demand of nitrogen and calcium, and that non-mycorrhizal root tips have the potential to take up most of the delivered nitrate and calcium.  相似文献

Heteroplastic combinations were made between Xenopus laevis presumptive neural plate and competent ectoderm of Xenopus borealis . Primarily induced presumptive neural plate cells ( Xenopus laevis ) can easily be distinguished from Xenopus borealis cells by specific quinacrine fluorescence of the nuclei. It was clearly shown that presumptive neural plate, which has primarily been induced by the underlying chordamesoderm exerts homoiogenetic inducing activity on competent ectoderm. The inducing activity is increased in pieces of presumptive neural plates, when the superficial layer has been removed from the adjacent deep layers. The enhancement can be explained by the fact that the removal of the superficial layer acting as barrier allows the inducing stimulus to be easily propagated from the apical (distal) side of the deep layers of the presumptive neural plate.  相似文献

Summary The afferent connections of the paraventricular nucleus (PVN) of the domestic mallard (Pekin duck), Anas platyrhynchos, were demonstrated by means of microiontophoretic injection of horseradish peroxidase (HRP). To place the HRP injection exactly into the PVN, its location was identified prior to the injection by observing antidiuretic reactions to electrostimulations within the rostral hypothalamus of conscious, hydrated animals. Antidiuresis was induced only when electrostimulation was applied to a distinct hypothalamic area. Two different patterns of antidiuresis were observed: (i) an immediate reduction in rate of production of urine, and (ii) antidiuresis preceded by a period of increase in production of urine. Repeated stimulation of the same site with the same parameters resulted in decreasing antidiuretic effects. At the site where stimulation had elicited the most pronounced antidiuresis of either response type, HRP was injected microiontophoretically.Histological examination after 3–8 days of survival revealed delicate injection sites located exclusively in the periventricular portion of the PVN. Adjacent to the dorsal portion of the PVN retrogradely labeled tanycytes and intraependymal neurons were scattered in the ventricular wall. As demonstrated in neurohistological and electron-microscopic investigations, this ependymal region exhibits a particular arrangement of tanycytes and small neurons (10–15 m in diameter), some of which belong to the neurosecretory type.Additional HRP-labeled neuronal perikarya afferent to the PVN were demonstrated in the contralateral PVN, and on the ipsilateral side in the lateral septum, lateral hypothalamic area and locus coeruleus. Within the nuclei of the solitary tract, stained nerve cells were found ipsilateral as well as contralateral to the injection site.Several of the neurons demonstrated may be considered as candidates for the transmission of signals originating from various receptive structures relevant for the control of avian salt- and water-balance. The physiological results conform to the concept that neurons of the PVN influence urine formation by controlling the release of arginine-vasotocin (AVT). Nuclear genes coding for the Mr 17000, 14000 and 11000 subunits of the ubiquinol-cytochrome c reductase complex (complex III) in yeast have been isolated from a clone bank of yeast nuclear DNA by use of a mRNA hybridization-competition assay. This is based on our observations that levels of mRNAs for these subunits are much reduced during glucose repression and in cytoplasmic petite mutants and the procedure should be of general application for the isolation of other inducible or repressible genes coding for mRNAs present at low levels in the cell. A first characterization of the clones is presented. The genes are not closely linked in the genome and those coding for Mr 14000 and 11000 subunits are present in unique genomic environments, which suggests that there are only single copies of each in the nuclear genome.  相似文献

Chromatographic methods suitable for the resolution of 24,25-dihydroxyvitamin D3, 24,25-dihydroxyvitamin D2, 25-hydroxyvitamin D3-26,23 lactone, and 25,26-dihydroxyvitamin D2 are described. These four metabolites comigrated in high-pressure liquid chromatography on silicic acid columns developed in 11:89 isopropanol:hexane. Adequate resolution was achieved by subjecting the four-metabolite complex to high-pressure liquid chromatography column developed in 2:98 isopropanol:methylene chloride. This additional chromatographic step, coupled with modifications of assay procedures previously described, allowed for the estimation of plasma concentrations of vitamin D2, vitamin D3, 25-hydroxyvitamin D2, 25-hydroxyvitamin D3, 24,25-dihydroxyvitamin D2, 24,25-dihydroxyvitamin D3, 25,26 dihydroxyvitamin D2, 25,26-dihydroxyvitamin D3, 25-hydroxyvitamin D3-26,23 lactone, and 1,25-dihydroxyvitamin D (1,25-dihydroxyvitamin D2 plus 1,25-dihydroxyvitamin D3). The samples automatically were introduced onto the high-pressure liquid chromatography columns with a Waters 710A “intelligent” processor. The metabolites were automatically collected with the aid of a programmable timer that advanced a fraction collector at predetermined intervals. The assays were used to determine the plasma vitamin D and vitamin D metabolite concentrations in five species of adult farm animals.  相似文献

Abstract: Plasma membranes were isolated from C6 glioblastoma cells by two methods. In the first method cells were treated with concanavalin A and lysed in hypotonic medium. After partial separation of plasma membranes from other cell material, the lectin was displaced with a-methyl-D-mannoside. In the second method untreated cells or cells iodinated in a lactoperoxidase-catalyzed reaction were homogenized in isotonic medium. Membrane fractions obtaincd by either homogenization procedure were further purified by rate zonal and equilibrium centrifugations into linear density gradients. Disruption of the glioblastoma cell membrane gives rise to heterogeneous assemblies of mem- brane fragments. Two populations of plasma membranes were isolated from untreated and from iodinated cells: a "lighter")membrane fraction characterized by relatively lower sedimentation velocity and buoyant density, and a "heavier" membrane fraction of relatively faster sedimentation velocity and higher buoyant density. Both fractions showed electrophoretic patterns similar to those of 125I-labeled cell surface proteins. Their specific (Na++ K+)-ATPase activity was seven- to eightfold the homogenate activity (recovery, 13.1%). Both fractions were, however, still contaminated by smooth endo- plasmic reticulum, as judged from the activity 0: NADPH-dependent cytochrome c reductase (recovery, 2.4%). It is suggested that plasma membrane fragments present in the two fractions might differ in the organization of their structures, e.g., membrane vesicle intactness and membrane orientation.  相似文献

Summary A new system of dispersal units has been elaborated, based on weight and morphological features functional in dispersal. Weight was divided into eight classes and the functional morphological features were selected in such a way that their effectiveness could be tested by experiments.The spectra of weight and dispersal adaptations of dispersal units sampled in Euphorbio-Pinetum nigrae and Fumano-Stipetum habitats south of Vienna are calculated with this system and then compared.The results show that both communities can be characterized with such spectra. It is also possible with these spectra to make statements about the ecological and social position of the association in a succession.Nomenclature follows Ehrendorfer (1973).  相似文献

A new metabolite of vitamin D3 has been isolated from the plasma of vitamin D3 treated cows and has been generated from 25(S),26-dihydroxyvitamin D3 with homogenates of vitamin D deficient chick kidney. This metabolite has been identified as 1,25,26-trihydroxyvitamin D3 by comigration with synthetic 1,25(S),26-trihydroxyvitamin D3 in four chromatographic systems, ultraviolet spectroscopy, mass spectrometry, and high-pressure liquid chromatography and mass spectrometry of derivatives. 1,25(S),26-Trihydroxyvitamin D3 is one-tenth as effective as 1,25-dihydroxyvitamin D3 in binding to the chick intestinal cytosol 1,25-dihydroxyvitamin D receptor. Either 25(S),26-dihydroxyvitamin D3 or 1,25-dihydroxyvitamin D3 can serve as precursor for in vitro production of 1,25,26-trihydroxyvitamin D3 by chick kidney tissue.  相似文献
