Recent studies on the in vivo roles of the sterol regulatory element binding protein (SREBP) family indicate that SREBP-2 is more specific to cholesterogenic gene expression whereas SREBP-1 targets lipogenic genes. To define the molecular mechanism involved in this differential regulation, luciferase-reporter gene assays were performed in HepG2 cells to compare the transactivities of nuclear SREBP-1a, -1c, and -2 on a battery of SREBP-target promoters containing sterol regulatory element (SRE), SRE-like, or E-box sequences. The results show first that cholesterogenic genes containing classic SREs in their promoters are strongly and efficiently activated by both SREBP-1a and SREBP-2, but not by SREBP-1c. Second, an E-box containing reporter gene is much less efficiently activated by SREBP-1a and -1c, and SREBP-2 was inactive in spite of its ability to bind to the E-box. Third, promoters of lipogenic enzymes containing variations of SRE (SRE-like sequences) are strongly activated by SREBP-1a, and only modestly and equally by both SREBP-1c and -2. Finally, substitution of the unique tyrosine residue within the basic helix-loop-helix (bHLH) portion of nuclear SREBPs with arginine, the conserved residue found in all other bHLH proteins, abolishes the transactivity of all SREBPs for SRE, and conversely results in markedly increased activity of SREBP-1 but not activity of SREBP-2 for E-boxes. These data demonstrate the different specificity and affinity of nuclear SREBP-1 and -2 for different target DNAs, explaining a part of the mechanism behind the differential in vivo regulation of cholesterogenic and lipogenic enzymes by SREBP-1 and -2, respectively.  相似文献

We examined the effects of a limitation of the O2-supply onthe syntheses of bacteriochlorophyll ap and carotenoids in isolatesof aerobic marine bacteria, OCh 101 and OCh 114, grown heterotrophically.Whereas they formed these pigments fairly well under high aerationin the dark, a limitation of the O2-supply resulted in the decreasedsyntheses of bacteriochlorophyll in both strains. Synthesesof carotenoids also were depressed under low aeration but toa lesser extent (especially in OCh 101) than the depressionof bacteriochlorophyll synthesis. Aerobic incubation of a cultureof OCh 101, that previously had been grown semiaerobically,induced the supplementary synthesis of bacteriochlorophyll.This induction was inhibited almost completely by chloram-phenicol.The absorption spectra of suspensions and solvent extracts ofcells grown aerobically or semiaerobically are reported. The thermal denaturation of 7S globulin has been examined as a function of ionic strength from 0 to 4.0 by using immunochemical methods, disc electrophoresis and turbidity. Changes in immunochemical properties were a measure of the extent of denaturation and provided information on the structural changes in 7S globulin at the molecular level. The 7S globulin did not lose its antigenicity even after heating, and both dissociates and aggregates maintained partial cross reactivity against anti-7S. The nature of thermal denaturation was divided into two main types at an ionic strength of approximately 2.0. At an ionic strength near zero, stable dissociation products were actually formed, and at ionic strengths from 0.1 to 2.0, the presence of salts increased in aggregate formation. The stabilizing effect of salt on thermal denaturation appeared at 2.0 ~ 2.2 μ. In the range of 3.5 ~ 4.0 μ, 7S globulin was heated to 100°C for 5min without changes in both antigenic specificity and electrophoresis pattern.  相似文献

The relationship between sulfation and polymerization in chondroitinsulfate (CS) biosynthesis has been poorly understood. In thisstudy, we investigated the specificity of bovine serum UDP-GalNAc:CS ß-GalNAc transferase responsible for chain elongationusing structurally defined acceptor substrates. They consistedof tetra- and hexasaccharide-serines that were chemically synthesizedand various regular oligosaccharides with a GlcA residue atthe nonreducing terminus, prepared from chondroitin and CS usingtesticular hyaluronidase. In this study, we investigated the preparation of polysaccharide supramolecular films through the formation of inclusion complexes by amylose in vine-twining polymerization using carboxymethyl cellulose-graft-poly(?-caprolactone) (CMC-g-PCL) as a new guest polymer. First, hydrogels were prepared by phosphorylase-catalyzed enzymatic polymerization in the presence of CMC-g-PCL according to the vine-twining polymerization manner. The XRD result of a powdered sample obtained by lyophilization of the resulting hydrogel indicated the presence of inclusion complexes of amylose with the PCL graft-chains between intermolecular (CMC-g-PCL)s, which acted as supramolecular cross-linking points for the hydrogelation. Then, the supramolecular films were obtained by adding water to the powdered samples, followed by drying. The mechanical properties of the selected films examined by tensile testing were superior to those of a CMC film. The effect of the supramolecular cross-linking structures on the mechanical properties of the films was evaluated further by several investigations.  相似文献
