Solid-state 13C nuclear magnetic resonance spectroscopy has been used to characterize the C in samples of the food (wood), gut contents and faeces from the wood-feeding termite, Microcerotermes parvus; soil in the guts and mound material from the soil-feeding termite, Thoracotermes macrothorax; and the food and faeces from the litter-feeding, coprophagous larvae of the dipteran fly, Bibio marci. Spectra from the wood-feeding termite indicated preferential loss of polysaccharide and accumulation of lignin with some modification to the O-aromatic-C and methoxyl-C (O-methyl-C) components during passage through the gut. Spectra for the soil-feeding termite indicated little change in the distribution of 13C between resonances following passage through the gut, except for some evidence of preferential polysaccharide loss. Interpretation of the spectra from these organisms was restricted by the relatively low C content of the soils and mound material, and by the large contribution to the NMR spectra from the gut tissue rather than the gut contents. Spectra for the litter-feeding dipteran larvae indicated preferential feeding on the polysaccharide-rich component of the litter and then overall loss of polysaccharide-C and accumulation of both aromatic-C and methoxyl-C in the gut. These changes were greater for the second passage than for the first passage through the gut, suggesting that principally mechanical and physical changes occurred initially and that chemical digestion was prevalent during the second passage.  相似文献

Effect of prolonged, heavy exercise on pulmonary gas exchange in athletes   总被引:1,自引:0,他引:1  
During maximalexercise, ventilation-perfusion inequality increases, especially inathletes. The mechanism remains speculative. Wehypothesized that, if interstitial pulmonary edema is involved, prolonged exercise would result in increasing ventilation-perfusion inequality over time by exposing the pulmonary vascular bed to highpressures for a long duration. The response to short-term exercise wasfirst characterized in six male athletes [maximal O2 uptake(O2 max) = 63 ml · kg1 · min1] by using 5 minof cycling exercise at 30, 65, and 90%O2 max. Multiple inert-gas, blood-gas, hemodynamic, metabolic rate, and ventilatory data were obtained. Resting log SD of the perfusion distribution (logSD) was normal [0.50 ± 0.03 (SE)] and increased with exercise (logSD = 0.65 ± 0.04, P < 0.005), alveolar-arterialO2 difference increased (to 24 ± 3 Torr), and end-capillary pulmonary diffusion limitation occurred at 90%O2 max. The subjectsrecovered for 30 min, then, after resting measurements were taken,exercised for 60 min at ~65%O2 max.O2 uptake, ventilation, cardiacoutput, and alveolar-arterial O2difference were unchanged after the first 5 min of this test, but logSD increased from0.59 ± 0.03 at 5 min to 0.66 ± 0.05 at 60 min(P < 0.05), without pulmonary diffusion limitation. LogSD was negativelyrelated to total lung capacity normalized for body surface area(r = 0.97,P < 0.005 at 60 min). These data are compatible with interstitial edema as a mechanism and suggest that lungsize is an important determinant of the efficiency of gas exchangeduring exercise.

The cellular response to environmental signals is largely dependent upon the induction of responsive protein kinase signaling pathways. Within these pathways, distinct protein-protein interactions play a role in determining the specificity of the response through regulation of kinase function. The interferon-induced serine/threonine protein kinase, PKR, is activated in response to various environmental stimuli. Like many protein kinases, PKR is regulated through direct interactions with activator and inhibitory molecules, including P58IPK, a cellular PKR inhibitor. P58IPK functions to represses PKR-mediated phosphorylation of the eukaryotic initiation factor 2α subunit (eIF-2α) through a direct interaction, thereby relieving the PKR-imposed block on mRNA translation and cell growth. To further define the molecular mechanism underlying regulation of PKR, we have utilized an interaction cloning strategy to identify a novel cDNA encoding a P58IPK-interacting protein. This protein, designated P52rIPK, possesses limited homology to the charged domain of Hsp90 and is expressed in a wide range of cell lines. P52rIPK and P58IPK interacted in a yeast two-hybrid assay and were recovered as a complex from mammalian cell extracts. When coexpressed with PKR in yeast, P58IPK repressed PKR-mediated eIF-2α phosphorylation, inhibiting the normally toxic and growth-suppressive effects associated with PKR function. Conversely, introduction of P52rIPK into these strains resulted in restoration of both PKR activity and eIF-2α phosphorylation, concomitant with growth suppression due to inhibition of P58IPK function. Furthermore, P52rIPK inhibited P58IPK function in a reconstituted in vitro PKR-regulatory assay. Our results demonstrate that P58IPK is inhibited through a direct interaction with P52rIPK which, in turn, results in upregulation of PKR activity. Taken together, our data describe a novel protein kinase-regulatory system which encompasses an intersection of interferon-, stress-, and growth-regulatory pathways.  相似文献

As part of a functional genomic study of scrapie pathogenesis we have generated a complex non-redundant microarray from several tissues by the selection of unique clones and the incremental augmentation of the expressed sequence tag sets. The base set of about 15,000 expressed sequence tags had minimum redundancy and originated from bovine brain. The aim was to augment this set with clones from scrapie-infected brain and spleen and uninfected spleen with minimum redundancy. This was achieved by macroarray hybridization to identify unique sequences in the ovine library. Quality sequence of 4052 unique sheep clones were obtained from the sequencing of only 5000 which have been submitted to the dbEST database at GenBank.  相似文献

We report observational data on behavioral laterality in 10 captive bonobos (Pan paniscus)at the San Diego Zoo. The unimanual measures include carrying, leading limb in locomotion, self-touching, face-touching, reaching, and gestures. We also recorded bimanual feeding in these subjects. A significant population level left-hand bias exists for carrying. Right-hand biases occur for leading limb in locomotion and gestures. During bimanual feeding, the bonobos hold food items with the left hand while feeding with the right hand. Overall, bonobos exhibit behavioral asymmetries that are similar to previous findings in other pongid ape species. The asymmetries in gestures and bimanual feeding represent novel findings with theoretical implications for the origins of tool use and language.  相似文献

Senescence is a highly orchestrated developmental stage in the life cycle of plants. The onset of senescence is tightly controlled by signaling cascades that initiate changes in gene expression and the synthesis of new proteins. This complement of new proteins includes hydrolytic enzymes capable of executing catabolism of macromolecules, which in turn sets in motion disassembly of membrane molecular matrices, leading to loss of cell function and, ultimately, complete breakdown of cellular ultrastructure. A distinguishing feature of senescence that sets it apart from other types of programmed cell death is the recovery of carbon and nitrogen from the dying tissue and their translocation to growing parts of the plant such as developing seeds. For this to be accomplished, the initiation of senescence and its execution have to be meticulously regulated. For example, the initiation of membrane disassembly has to be intricately linked with the recruitment of nutrients because their ensuing translocation out of the senescing tissue requires functional membranes. Molecular mechanisms underlying this linkage and its integration with the catabolism of macromolecules in senescing tissues are addressed.  相似文献

An artificial neural network is used to classify environments, including climate, terrain and soil variables, according to their suitability for fifteen structural/environmental forest classes in the Wet Tropics Bioregion of north-east Queensland. We map the environments characteristic of these forest classes in four climate regimes (the present and three past climate scenarios), quantify the changes in area of these environments in response to past regional changes in climate and identify areas that would have been environmentally suitable for rainforests at last glacial maximum (glacial refugia). We also identify areas that would have been suitable for upland and highland rainforest classes during the warmest parts of the interglacial (interglacial refugia) and map locations that consistently remain favourable to specific forest classes despite large changes in climate.In the climate of the last glacial maximum (LGM), rainforest environments are predicted in three relatively distinct refugia in the northern, central and southern Wet Tropics. Only three percent of the total area contains lowland, Mesophyll Vine Forest and the majority of the area of the rainforest refugia supports upland rainforest classes. In the cool, wet climate of the Pleistocene/Holocene transition (PHT), rainforest environments expand to form a more or less continuous block from the northern limits of the region to the Walter Hill Range, except for discontinuous patches extending through the Seaview and Paluma Ranges in the south. During the Holocene climatic optimum (HCO), rainforest environments become more fragmented, especially in the south. Lowland rainforest environments are very extensive in this climate while upland rainforest classes are restricted to what we term “interglacial refugia”.Estimated distributions and stable locations (consistently predicted in all four climate scenarios) for the various rainforest environment classes are our main, novel contribution. Each forest environment responds individualistically to climate change. Our results confirm the highly dynamic nature of the Wet Tropics landscape and present a much more detailed picture of landscape change since the late Pleistocene than previously has been available. This mapping exercise should be useful in the future for analyses of present-day biogeographic patterns. We argue that empirical modelling approaches have an important role in palaeoecology and global change research that is complementary to the developing mechanistic methods.  相似文献

This study addressed whether personality ratings using a 43 adjective questionnaire based on the Five-Factor Model generalized from a sample of 202 zoo-housed chimpanzees to a sample of 175 chimpanzees housed in Yerkes National Primate Research Center. Mean interrater reliabilities of adjectival ratings were lower for the chimpanzees housed in Yerkes. In addition, rank order of the interrater reliabilities of items differed between settings. To compare factor structure, we first examined whether we could replicate the original six factor structure found in an earlier study of 100 zoo chimpanzees using principal factors analysis in the Yerkes sample and 102 new zoo chimpanzees. The dominance, extraversion, conscientiousness, and agreeableness factors were clearly replicated in the Yerkes sample and the 102 new zoo chimpanzees. The Neuroticism and Openness factors did not replicate in the Yerkes sample, but they also did not replicate in the new zoo chimpanzees. These findings suggest the need to sample more adjectives representing neuroticism and openness in future versions of the questionnaire. We next sought to determine whether factor structure, as determined by principal components analysis, remained invariant across the two settings. This analysis revealed dominance, extraversion, conscientiousness, and agreeableness factors in both settings and a high level of congruence between the zoo and Yerkes samples for these factors. Finally, we tested whether factor scores in the two samples were similarly related to age and sex. With the exception of differences in age effects for dominance and agreeableness, age, and sex effects were consistent across samples. These findings suggest that, whereas there may be differences in the ease with which ratings are made, personality structure, and development are largely consistent across widely differing settings.  相似文献
