Summary Acid phosphatases are localized on the internal strands of sieve-tube members in the secondary phloem of Tilia americana. Companion cells and certain strand parenchyma cells associated with sieve-tube members show a high activity of acid phosphatases.This research has been supported by NSF Grant GB-615.  相似文献

The polymorphic phase behavior of aqueous dispersions of 1-oleoyl-, 1-linoleoyl-, and 1-linolenoyl-sn-3-glycerophosphoethanolamine (1-C18:1c-PE, 1-C18:2c-PE, and 1-C18:3c-PE, respectively) has been investigated by 31P NMR, small-angle and wide-angle X-ray diffraction, and freeze-fracture techniques in response to changes in temperature and pH. Between -20 and 0 degrees C at pH 7, NMR and X-ray data indicate that 1-C18:1c-PE adopts a lamellar phase. Above 20 degrees C, the X-ray diffraction from 1-C18:1c-PE reveals no long-range lattice order, whereas the NMR data indicate lamellar structure to 90 degrees C. Freeze-fracture electron microscopy shows that 1-C18:1c-PE at pH 8.2 forms closed multilamellar vesicles upon dispersion and also that large unilamellar vesicles produced by extrusion techniques (LUVETs) can be made from 1-C18:1c-PE at pH 7. Such LUVETs can trap [3H]inulin and support a K+ diffusion potential for up to 4 h. At pH 8.5 and above, 1-C18:1c-PE forms optically clear, fluid dispersions with NMR and X-ray characteristics consistent with a micellar (noninverted) phase structure. Attempts to prepare LUVETs from 1-C18:1c-PE at pH 9 result in structures that can neither trap [3H]inulin nor support a membrane potential.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

A new melanocyte-stimulating peptide has been isolated from acid extracts of frozen human pituitary glands by salt/ethanol fractionation, Sephadex G-75 gel filtration and DEAE- and cM-cellulose ion-exchange chromatography. The peptide is glycosylated, has an N-terminal tryptophan residue and an apparent mol.wt. of 16000 as estimated by sodium dodecyl sulphate/polyacrylamide-gel electrophoresis. Its amino acid analysis closely resembles residues Trp-105 to Gln-29 predicted for the common precursor protein of bovine corticotropin and beta-lipotropin by Nakanishi, Inoue, Kita, Nakamura, Chang, Cohen & Numa [(1979) Nature (London) 278, 423-427]. This fragment is expected to have melanotropin activity due to the tetrapeptide -His-Phe-Arg-Trp- (residues -51 to -48) of the predicted sequence of the common precursor. It was found to have a molar potency of 1 X 10(-5) relative to alpha-melanotropin in the frog skin bioassay. These characteristics are consistent with the isolated melanotropin peptide being a non-corticotropin, non-lipotropin peptide of the human common precursor protein of corticotropin and lipotropin. The peptide neither potentiates the adrenal weight-maintenance activity of corticotropin-(1-24)-tetracosapeptide when administered to hypophysectomized rats, nor stimulates release of non-esterified fatty acids from isolated rat epididymal cells. A second N-terminal-tryptophan glycopeptide was also isolated, which had an amino-acid composition similar to that predicted for the bovine common precursor protein, residues Trp-105 to Gly-35.  相似文献

The molecular and isotopic compositions of lipid biomarkers of cultured Aquificales genera have been used to study the community and trophic structure of the hyperthermophilic pink streamers and vent biofilm from Octopus Spring. Thermocrinis ruber, Thermocrinis sp. strain HI 11/12, Hydrogenobacter thermophilus TK-6, Aquifex pyrophilus, and Aquifex aeolicus all contained glycerol-ether phospholipids as well as acyl glycerides. The n-C(20:1) and cy-C(21) fatty acids dominated all of the Aquificales, while the alkyl glycerol ethers were mainly C(18:0). These Aquificales biomarkers were major constituents of the lipid extracts of two Octopus Spring samples, a biofilm associated with the siliceous vent walls, and the well-known pink streamer community (PSC). Both the biofilm and the PSC contained mono- and dialkyl glycerol ethers in which C(18) and C(20) alkyl groups were prevalent. Phospholipid fatty acids included both the Aquificales n-C(20:1) and cy-C(21), plus a series of iso-branched fatty acids (i-C(15:0) to i-C(21:0)), indicating an additional bacterial component. Biomass and lipids from the PSC were depleted in (13)C relative to source water CO(2) by 10.9 and 17.2 per thousand, respectively. The C(20-21) fatty acids of the PSC were less depleted than the iso-branched fatty acids, 18.4 and 22.6 per thousand, respectively. The biomass of T. ruber grown on CO(2) was depleted in (13)C by only 3.3 per thousand relative to C source. In contrast, biomass was depleted by 19.7 per thousand when formate was the C source. Independent of carbon source, T. ruber lipids were heavier than biomass (+1.3 per thousand). The depletion in the C(20-21) fatty acids from the PSC indicates that Thermocrinis biomass must be similarly depleted and too light to be explained by growth on CO(2). Accordingly, Thermocrinis in the PSC is likely to have utilized formate, presumably generated in the spring source region.  相似文献

In 1997, Oregon enacted rules that define an unacceptable population-level risk as a >10% chance of >20% of the total local population receiving an exposure greater than the toxicity reference value. This rule applies to populations of plants and animals not listed as threatened or endangered. An operational procedure was developed to perform such population-level ecological risk assessments. This case study describes how this procedure was used to develop site-specific groundwater remedial action objectives for resident aquatic populations at a site in Northwest Oregon, where an upland petroleum pipeline released gasoline-range hydrocarbons (TPH-G) into soils and groundwater immediately upgradient of the Tualatin River. With an interim response in place, the goal was to proactively establish a remedial action objective that would equate to an acceptable risk level for populations of aquatic receptors potentially threatened by future groundwater discharges to the river. Approximately 10 and 32% dilutions of the water-soluble fraction of pure product produced an acceptable risk level for zooplankton and forage fish populations, respectively. The proposed population-level remedial action objective, based on zooplankton as the most sensitive receptor, was a TPH-G groundwater concentration of 17.4?mg/L.  相似文献
