The majority of new HIV infections occur in women as a result of heterosexual intercourse, overcoming multiple innate barriers to infection within the mucosa. However, the avenues through which infection is established, and the nature of bottlenecks to transmission, have been the source of considerable investigation and contention. Using a high dose of a single round non-replicating SIV-based vector containing a novel dual reporter system, we determined the sites of infection by the inoculum using the rhesus macaque vaginal transmission model. Here we show that the entire female reproductive tract (FRT), including the vagina, ecto- and endocervix, along with ovaries and local draining lymph nodes can contain transduced cells only 48 hours after inoculation. The distribution of infection shows that virions quickly disseminate after exposure and can access target cells throughout the FRT, with an apparent preference for infection in squamous vaginal and ectocervical mucosa. JRFL enveloped virions infect diverse CD4 expressing cell types, with T cells resident throughout the FRT representing the primary target. These findings establish a new perspective that the entire FRT is susceptible and virus can reach as far as the ovary and local draining lymph nodes. Based on these findings, it is essential that protective mechanisms for prevention of HIV acquisition must be present at protective levels throughout the entire FRT to provide complete protection. 相似文献

There is increasing evidence to suggest that late chronotypes are at increased risk for depression. The putative psychological mechanisms underpinning this risk, however, have not been fully explored. The aim of the present study was to examine whether, similar to acutely depressed patients and other “at risk” groups, late chronotype individuals display biases in tasks assaying emotional face recognition, emotional categorisation, recognition and recall and attention. Late chronotype was associated with increased recognition of sad facial expressions, greater recall and reduced latency to correctly recognise previously presented negative personality trait words and reduced allocation of attentional resources to happy faces. The current results indicate that certain negative biases in emotional processing are present in late chronotypes and may, in part, mediate the vulnerability of these individuals to depression. Prospective studies are needed to establish whether the cognitive vulnerabilities reported here predict subsequent depression. 相似文献

Marsupial x eutherian cell hybrids would be very useful for studies of mammalian genetics and cell biology. A critical step in the formation of such hybrids is the fusion of cells to form heterokaryons. We have examined many different combinations of marsupial and eutherian cells for their ability to fuse, and we have found that all combinations yielded heterokaryons, but with different frequencies, depending on the cell types used. Ranked in order of decreasing ability to fuse with eutherian cells, the marsupial cell types were; established lines, primary diploid fibroblasts and lymphocytes. In all fusion experiments there was a marked preference for the formation of homokaryons compared with heterokaryons.
