In Xanthomonas campestris pv. campestris (Xcc), the proteins encoded by the rpf (regulator of pathogenicity factor) gene cluster produce and sense a fatty acid signal molecule called diffusible signalling factor (DSF, 2(Z)‐11‐methyldodecenoic acid). RpfB was reported to be involved in DSF processing and was predicted to encode an acyl‐CoA ligase. We report that RpfB activates a wide range of fatty acids to their CoA esters in vitro. Moreover, RpfB can functionally replace the paradigm bacterial acyl‐CoA ligase, Escherichia coli FadD, in the E. coli ß‐oxidation pathway and deletion of RpfB from the Xcc genome results in a strain unable to utilize fatty acids as carbon sources. An essential RpfB function in the pathogenicity factor pathway was demonstrated by the properties of a strain deleted for both the rpfB and rpfC genes. The ΔrpfB ΔrpfC strain grew poorly and lysed upon entering stationary phase. Deletion of rpfF, the gene encoding the DSF synthetic enzyme, restored normal growth to this strain. RpfF is a dual function enzyme that synthesizes DSF by dehydration of a 3‐hydroxyacyl‐acyl carrier protein (ACP) fatty acid synthetic intermediate and also cleaves the thioester bond linking DSF to ACP. However, the RpfF thioesterase activity is of broad specificity and upon elimination of its RpfC inhibitor RpfF attains maximal activity and its thioesterase activity proceeds to block membrane lipid synthesis by cleavage of acyl‐ACP intermediates. This resulted in release of the nascent acyl chains to the medium as free fatty acids. This lack of acyl chains for phospholipid synthesis results in cell lysis unless RpfB is present to counteract the RpfF thioesterase activity by catalysing uptake and activation of the free fatty acids to give acyl‐CoAs that can be utilized to restore membrane lipid synthesis. Heterologous expression of a different fatty acid activating enzyme, the Vibrio harveyi acyl‐ACP synthetase, replaced RpfB in counteracting the effects of high level RpfF thioesterase activity indicating that the essential role of RpfB is uptake and activation of free fatty acids. 相似文献

Objectives
To explore the relationship between non-alcoholic fatty liver disease (NAFLD) and the metabolic syndrome (MetS), and evaluate the value of NAFLD as a marker for predicting the risk of MetS in a large scale prospective cohort from northern urban Han Chinese population.Materials and Methods
A total of 17,920 MetS-free at baseline cohort members was included in the current study between 2005 and 2011. The baseline characteristics of the cohort were compared by NAFLD status at baseline, MetS status after follow-up. Cox proportional hazards models were used to estimate the unadjusted or adjusted hazard ratios (HRs) for NAFLD at baseline predicting the risk of MetS.Results
2,183 (12.18%) new cases of MetS occurred between 2005 and 2011. In unadjusted model, HRs (95% CIs) for NAFLD predicting MetS was 3.65 (3.35, 3.97). After adjusting the confounding factors of age, gender, the metabolic factors, smoke and exercise, the HRs (95% CIs) was 1.70 (1.55, 1.87). Gender difference was observed, adjusted HRs (95% CIs) of NAFLD for predicting MetS were 2.06(1.72, 2.46) and 1.55(1.39, 1.72) in female and male population, respectively. Moreover, 163 participants developed MetS among participants without any MetS component at baseline, and its adjusted HRs was still significant, 1.87 (1.12, 3.13).Conclusion
The present study indicates that NAFLD is an independent risk factor for predicting the risk of MetS in northern urban Han Chinese population, and the people with NAFLD should initiate weight and dietary control to prevent the occurrence of MetS. 相似文献28.
Quetiapine (Que), a commonly used atypical antipsychotic drug (APD), can prevent myelin from breakdown without immune attack. Multiple sclerosisis (MS), an autoimmune reactive inflammation demyelinating disease, is triggered by activated myelin-specific T lymphocytes (T cells). In this study, we investigated the potential efficacy of Que as an immune-modulating therapeutic agent for experimental autoimmune encephalomyelitis (EAE), a mouse model for MS. Que treatment was initiated on the onset of MOG(35-55) peptide induced EAE mice and the efficacy of Que on modulating the immune response was determined by Flow Cytometry through analyzing CD4(+)/CD8(+) populations and the proliferation of effector T cells (CD4(+)CD25(-)) in peripheral immune organs. Our results show that Que dramatically attenuates the severity of EAE symptoms. Que treatment decreases the extent of CD4(+)/CD8(+) T cell infiltration into the spinal cord and suppresses local glial activation, thereby diminishing the loss of mature oligodendrocytes and myelin breakdown in the spinal cord of EAE mice. Our results further demonstrate that Que treatment decreases the CD4(+)/CD8(+) T cell populations in lymph nodes and spleens of EAE mice and inhibits either MOG(35-55) or anti-CD3 induced proliferation as well as IL-2 production of effector T cells (CD4(+)CD25(-)) isolated from EAE mice spleen. Together, these findings suggest that Que displays an immune-modulating role during the course of EAE, and thus may be a promising candidate for treatment of MS. 相似文献

研究功能多样性与物种多样性关系及其随环境梯度的变化规律,有助于理解生物在群落中的共存机制;然而,二者间关系的研究在淡水生态学中尚鲜见报道。通过对新薛河典型河段(A缓流河段、B断流河段、C有机污染河段、D对照河段、E人为干扰河段)底栖动物季节性调查,就物种多样性和功能多样性时空动态及关系进行了研究。结果表明:在空间序列上,物种多样性指数在B河段均最低,表明间歇性断流对物种多样性影响重大。功能丰富度在D河段最高,A河段最低;功能均匀度在A河段高于其他河段;功能分离度在A、B河段最高,D河段最低。在时间序列上,物种丰富度和Shannon指数均值在10月份最低,4月份最高;均匀度指数在12月份最低,10月份最高。3个功能多样性指数于各季节间差异显著、相互独立,主要受水文条件和底栖动物生活史影响。相关分析表明,功能多样性指数间无显著相关性;功能丰富度同物种丰富度和Shannon指数相关显著,功能均匀度同物种均匀度相关显著。逐步回归分析发现,功能丰富度受物种丰富度和Shannon指数影响显著,功能均匀度受物种均匀度影响显著;功能多样性和物种多样性指数间拟合度总体不高。研究结果进一步表明:相对物种多样性,功能多样性对生境梯度变化响应更加全面。 相似文献

群落时空格局研究是后续深入开展相关研究的基础。基于2012年山东省新薛河底栖动物调查数据,就新薛河上游典型溪流生境底栖动物群落结构进行了比较研究。结果表明:共采集底栖动物108种,隶属10纲,74属;短脉纹石蛾(Cheumatopsyche sp.)、长钝直突摇蚊(Orthocaldius vaillanti)、拟长跗摇蚊属一种(Paratanytarsus sp.E)、Neozarelia sp.为研究区域优势类群,相对丰度分别为25.1%、9.8%、9.0%、8.6%。各河段优势物种组成、密度、生物量、生物多样性均存在显著差异。非度量多维标度排序和多响应置换过程分析从群落层面验证了不同河段群落结构的差异;且发现,流量越小,群落分化越明显。通过指示物种和双向聚类分析,进一步明确了物种组成对群落结构的影响。总之,不同生境底栖动物物种组成和群落结构存在较大差异,栖境多样性对生物多样性的维持和保护具有重要意义。 相似文献
