New insights into membrane fouling in a submerged anaerobic membrane bioreactor based on characterization of cake sludge and bulk sludge 总被引:1,自引:0,他引:1
A laboratory-scale submerged anaerobic membrane bioreactor (SAnMBR) treating thermomechanical pulping whitewater was operated for over 7 months to investigate and compare the characteristics of cake sludge and bulk sludge during stable state operation period. Serial analysis showed that cake sludge had a smaller particle size distribution (PSD), much higher specific filtration resistance (1.34 × 1014 m/kg), 1.5 times higher bound EPS and significantly different microbial community as compared with bulk sludge. Further analysis indicated that small flocs, bound EPS and inorganic materials play important role in cake formation process. The formed cake layer was found to have a heterogeneous structure. The results obtained in this study indicated that cake formation process started from attachment of small flocs and/or specific bacterial clusters which colonize the surface of the membrane and provide enhanced conditions that allow for cake formation to progress. 相似文献

A two-continuous mesophilic (37 ± 2 °C) UASB system with step-feed was investigated as an attractive optimization strategy for enhancing COD and total phenols removal of the system and improving aerobic biodegradability of real coal gasification wastewater. Through the step-feed period, the maximum removal efficiencies of COD and total phenols reached 55-60% and 58-63% respectively in the system, at an influent flow distribution ratio of 0.2 and influent COD concentration of 2500 mg/L; the corresponding efficiencies were at low levels of 45-50% and 43-50% respectively at total HRT of 48 h during the single-feed period. The maximum specific methanogenic activity and substrate utilization rate were 592 ± 16 mg COD-CH4/(gVSS d) and 89 ± 12 mg phenol/(gVSS d) during the step-feed operation. After the anaerobic digestion with step-feed, the aerobic effluent COD concentration decreased from 270 ± 9 to 215 ± 10 mg/L. The results suggested that step-feed enhanced the degradation of refractory organics in the second reactor. 相似文献

Elucidating kinetic processes of protein–protein interactions (PPI) helps to understand how basic building blocks affect overall behavior of living systems. In this study, we used structure‐based properties to build predictive models for kinetic constants of PPI. A highly diverse PPI dataset, protein–protein kinetic interaction data and structures (PPKIDS), was built. PPKIDS contains 62 PPI with complex structures and kinetic constants measured experimentally. The influence of structural properties on kinetics of PPI was studied using 35 structure‐based features, describing different aspects of complex structures. Linear models for the prediction of kinetic constants were built by fitting with selected subsets of structure‐based features. The models gave correlation coefficients of 0.801, 0.732, and 0.770 for koff, kon, and Kd, respectively, in leave‐one‐out cross validations. The predictive models reported here use only protein complex structures as input and can be generally applied in PPI studies as well as systems biology modeling. Our study confirmed that different properties play different roles in the kinetic process of PPI. For example, kon was affected by overall structural features of complexes, such as the composition of secondary structures, the change of translational and rotational entropy, and the electrostatic interaction; while koff was determined by interfacial properties, such as number of contacted atom pairs per 100 Å2. A strain of the cyanobacterium Arthrospira was isolated from Lake Chahannaoer in northern China and was characterized according to microscopic morphology, photosynthetic oxygen-evolving activity, growth rate, and nutritional profile. Compared with thermophilic Arthrospira species occurring naturally in tropical and subtropical lakes, this isolate is mesophilic and grows optimally at ~20 degreesC. The total protein, fatty acid, phycocyanin, carotenoid, and chlorophyll a contents were 67.6, 6.1, 4.32, 0.29, and 0.76 grams per 100 grams of dry weight, respectively. The strain is rich in polyunsaturated fatty acids (PUFAs). An essential omega-3 fatty acid, docosahexaenoic acid (DHA), was detected, and gamma-linolenic acid (GLA) and DHA accounted for 28.3% of the total fatty acid content. These features of this newly isolated strain make it potentially useful in commercial mass culture in local areas or as a biofuel feedstock. It is also an alternative resource for studying the metabolic PUFA pathways and mechanisms of cold stress tolerance in cyanobacteria. 相似文献

Cytosine methylation is the major epigenetic modification of metazoan DNA. Although there is strong evidence that active DNA demethylation occurs in animal cells, the molecular details of this process are unknown. The recent discovery of the TET protein family (TET1–3) 5-methylcytosine hydroxylases has provided a new entry point to reveal the identity of the long-sought DNA demethylase. Here, we review the recent progress in understanding the function of TET proteins and 5-hydroxymethylcytosine (5hmC) through various biochemical and genomic approaches, the current evidence for a role of 5hmC as an early intermediate in active DNA demethylation and the potential functions of TET proteins and 5hmC beyond active DNA demethylation. Allelopathy of diterpenoids extracted from plants of the genus Robdosia on three common species of soil ciliates, Colpoda inflata, Colpoda cucullus and Euplotes muscicola, was studied by acute toxicity test, sub-lethal effect test and morphological observation. Acute toxicity test showed that there was remarkable toxicity of the diterpenoids on the individuals of the three soil ciliate species, and there was close correlation between toxicity and concentration of the diterpenoids. 12 h-LC50 values of the diterpenoids on the individuals of C. inflata, C. cucullus and E. muscicola were 161.40 mg L?1, 94.80 mg L?1 and 83.70 mg L?1 respectively, and 24 h-LC50 values were 114.90 mg L?1, 92.30 mg L?1and 65.80 mg L?1 separately. Sub-lethal effect test of soil ciliates suggested that there existed significant inhibition of the diterpenoids on population growth of the three ciliates with dose-dependant relationships, population density and growth rate of the test group was obviously lower than that of the control group. Morphological observation indicated that diterpenoids affected the body shapes of the three ciliates and made them shorter and thicker, and the higher the concentration of diterpenoids, the greater the affection. The results are of great significance for understanding the functions of ciliates and their relationships to other organisms, and for the application of allelopathy in biological pest control in the soil ecosystems. 相似文献

The chemokine family forms two different types of homodimer despite members sharing nearly identical folds. To study the formation of quaternary structure in this family, rational mutagenesis was employed on a representative member of each subfamily (MIP-1beta and IL-8). The variants were studied by analytical ultracentrifugation and NMR, and it was determined that formation of a folded monomer from a natural chemokine dimer is reasonably facile, while conversion between dimer types is not. Monomeric variants of MIP-1beta and IL-8 were randomly mutated and a lambda phage-based selection system was employed in a novel way to screen for dimerization. A total of 6,000,000 random mutants were screened, but no dimers were formed, suggesting again that the chemokine fold is robust and amenable to sequence variation, while the chemokine dimer is much more difficult to attain. This work represents a biophysical analysis of an array of chemokine quaternary state variants. 相似文献
