孕小鼠注射海洛因对仔鼠体重、行为及肺中相关活性物质表达的影响 总被引:1,自引:0,他引:1
为了探讨海洛因对仔鼠行为、体重及肺发育中KGF、c-Fos蛋白和Bax蛋白表达的影响,对4组共48例受孕小鼠(Mus musculus)从第8 d开始,每天早晚分别注射0.2 ml浓度为1.0 g/L、1.5 g/L和2.0 g/L的海洛因溶液和等量的生理盐水直到小鼠分娩,观察测量仔鼠行为及体重变化,采用免疫组织化学染色和体视学半定量方法检测15 d胚胎、出生后1 d.7 d、15 d仔鼠的肺中KGF、c-Fos蛋白和Bax蛋白表达情况.结果表明,海洛因影响仔鼠的行为活动,实验组各发育期仔鼠的体重明显低于对照组,海洛因组各发育期仔鼠肺中KGF、c-Fos蛋白和Bax蛋白的表达强度与对照组相比显著增强(P<0.01或P<0.05),且随海洛因浓度的增大表达越强,但随着发育的进行,海洛因组仔鼠肺中KGF、c-Fos蛋白和Bax蛋白的阳性表达减弱.海洛因影响仔鼠的行为与体重,海洛因对各发育期仔鼠肺的损伤可能与肺组织中KGF、c-Fos蛋白和Bax蛋白表达的增强有关. 相似文献

Interferon gamma (IFNgamma) is regarded as a potent antitumor agent, but therapy with IFNgamma is hampered by its short half-life and significant side effects. We developed a replication defective adenovirus carrying the human IFNgamma gene and evaluated the effects of adenovirus-mediated IFNgamma (Ad-IFNgamma) gene transfer on human prostate cancer cell lines in vitro and on xenografts in vivo. Our results showed infection of prostate cancer cells with Ad-IFNgamma led to production of an active cytokine and resulted in an antiproliferative effect on the prostate cancer cells. Intratumoral injection of Ad-IFNgamma significantly inhibited the growth of DU-145 cell xenografts in vivo, while no significant toxicity effect was observed. RT-PCR analysis indicated transgene expression mainly enriched in tumors in vivo, and slightly distributed in livers. These findings suggest adenovirus-mediated IFNgamma gene transfer is a promising approach in the treatment of advanced prostate cancer. 相似文献

目的:建立红细胞生长刺激蛋白(ESP)的二硫键连接方式的测定方法。方法:先将红细胞生长刺激蛋白的糖链用糖苷酶切除,再分别对ESP和还原烷基化后ESP用胰蛋白酶进行酶切,然后用MALDI-TOF测得该蛋白质的肽质量指纹图谱,通过比较还原烷基化前后各肽质量指纹图谱,找到差异肽段的分子离子峰[M+H]+,通过比对该蛋白理论酶切肽的[M+H]+,确定二硫键的连接方式。结果:ESP有4个半胱氨酸,通过比较还原烷基化前后的肽质量指纹图谱定位二硫键的位置为Cys7-Cys161和Cys29-Cys33,与理论上的二硫键连接相符。结论:建立了酶切结合质谱法测定蛋白质二硫键定位的方法,为今后生物技术产品的二硫键连接方式的质量控制提供了有效的方法。 相似文献

GRB2相关结合蛋白(GRB2-associated binding protein 2,GAB2)是Gabs家族重要成员,在细胞增殖、分化及迁移等过程中发挥重要作用。已有研究证明,GAB2与肿瘤侵袭转移相关,但目前GAB2在乳腺癌细胞侵袭中的研究少见报道。本研究证明,GAB2可提高乳腺癌MCF-7细胞迁移能力。Western印迹结果显示,GAB2蛋白在高转移细胞株MDA-MB-231中高表达|相反,在低转移细胞株MCF-7中低表达。瞬时转染结合Western印迹、酶联免疫吸附实验(ELISA)揭示,过表达GAB2明显增加MMP-2及MMP-9蛋白表达。Transwell检测显示,过表达GAB2明显增强MCF-7细胞的迁移能力。与未转染细胞比较,采用10 ng/mL表皮细胞生长因子(EGF)刺激转染细胞5 min,即可明显增强磷酸化的Akt、ARK5表达。上述结果提示,GAB2可通过参与Akt-ARK5信号通路,增加MMP-2、MMP-9蛋白表达,增强肿瘤细胞的迁移能力,这可能是GAB2促进肿瘤侵袭的部分机制。 相似文献

Vegetative propagule pressure may affect the establishment and structure of aquatic plant communities that are commonly dominated by plants capable of clonal growth. We experimentally constructed aquatic communities consisting of four submerged macrophytes (Hydrilla verticillata, Ceratophyllum demersum, Elodea nuttallii and Myriophyllum spicatum) with three levels of vegetative propagule pressure (4, 8 and 16 shoot fragments for communities in each pot) and two levels of water depth (30 cm and 70 cm). Increasing vegetative propagule pressure and decreasing water level significantly increased the growth of the submerged macrophyte communities, suggesting that propagule pressure and water depth should be considered when utilizing vegetative propagules to re-establish submerged macrophyte communities in degraded aquatic ecosystems. However, increasing vegetative propagule pressure and decreasing water level significantly decreased evenness of the submerged macrophyte communities because they markedly increased the dominance of H. verticillata and E. nuttallii, but had little impact on that of C. demersum and M. spicatum. Thus, effects of vegetative propagule pressure and water depth are species-specific and increasing vegetative propagule pressure under lower water level can facilitate the establishment success of submerged macrophyte communities. 相似文献
