We used 206.5-nm excited resonance Raman measurements to examine the angiotensin II (AII) secondary structure in H(2)O in the presence of dodecylphosphocholine (DPC) micelles, sodium dodecylsulfate (SDS) monomers and micelles, and in a 70% acetonitrile (ACN-d)-30% water solution. Our AII-SDS titration absorption studies indicate the formation of a 1:2 AII:SDS complex in which two negatively charged SDS molecules attach to the AII positively charged N terminus and to Arg(2). Our 206.5-nm excited Raman results indicate that the 1:2 AII:SDS complexation increases the AII beta-turn composition. We also used 228.9-nm Raman excitation to probe the local solvent accessibility of Tyr(4) (AII) in DPC and SDS micelles. Our Tyr (AII) solvent accessibility studies suggest that the Tyr residue is more exposed to the aqueous environment in SDS micelles than in DPC micelles.  相似文献

The effects of the egg density of Diaphania hyalinata (L.) on several biological parameters of Trichogramma exiguum Pinto & Platner and T. pretiosum Riley were investigated. For that, 24h-old egg masses were isolated in glass tubes (15 replicates; 1 egg mass = 1 replicate), and offered to parasitization by a newly-emerged female of T. pretiosum or T. exiguum 24h at the proportion of one female to 5, 10, 15, 20, 25, 30, 35 and 40 eggs of D. hyalinata. The following parameters were evaluated: number of parasitized eggs, number of individuals per egg, viability and sex ratio. Trichogramma exiguum parasitized more eggs than T. pretiosum when more than 25 eggs were available per female. The percentage of emergence was satisfactory to T. pretiosum in densities up to 15 eggs/female and up to 20 eggs/female for T. exiguum. The number of individuals per egg was not statistically different in both species except in the density of 25 eggs/female. It can be concluded that T. exiguum performed better than T. pretiosum at larger clutch sizes, as T. exiguum parasitization capacity increased as a result of the size of the host clutch size.  相似文献

In animals, microRNAs (miRNAs), typically, pair to sites of partial complementarity in the 3′-untranslated regions (3′UTRs) of target genes. Regulation by miRNAs often results in down-regulation of target mRNA and protein expression by mechanisms that are yet to be fully elucidated. Additionally, changes in environmental conditions have been shown to influence miRNA function in some cell culture systems. Here, we report the effect of nutrient deprivation on regulation of an endogenous miRNA target in developing worms. In Caenorhabditis elegans, the lin-4 miRNA recognizes multiple sites in the lin-14 3′UTR and directs mRNA degradation and translational repression, but it is unclear how these processes are coupled. In this study, we demonstrate that nutrient deprivation results in loss of lin-14 mRNA, but not protein, repression. In worms removed from feeding conditions, lin-14 mRNA reaccumulates despite the continued expression of lin-4 miRNA. The relative increase in lin-14 mRNA levels during nutrient deprivation is less pronounced in genetic mutants lacking lin-4 miRNA or the lin-14 3′UTR target sites. In conclusion, regulation of lin-14 at the mRNA and protein levels can be uncoupled by changes in culture conditions, indicating that miRNA function can be modulated by environment in multicellular organisms. The awareness that endogenous miRNA pathways can be sensitive to environment is an important consideration for elucidating the mechanism used by miRNAs to regulate target mRNA and protein expression.  相似文献

Among marine phytoplankton groups, diatoms span the widest range of cell size, with resulting effects upon their nitrogen uptake, photosynthesis and growth responses to light. We grew two strains of marine centric diatoms differing by ~4 orders of magnitude in cell biovolume in high (enriched artificial seawater with ~500 µmol L?1 µmol L?1 NO3 ?) and lower-nitrogen (enriched artificial seawater with <10 µmol L?1 NO3 ?) media, across a range of growth light levels. Nitrogen and total protein per cell decreased with increasing growth light in both species when grown under the lower-nitrogen media. Cells growing under lower-nitrogen media increased their cellular allocation to RUBISCO and their rate of electron transport away from PSII, for the smaller diatom under low growth light and for the larger diatom across the range of growth lights. The smaller coastal diatom Thalassiosira pseudonana is able to exploit high nitrogen in growth media by up-regulating growth rate, but the same high-nitrogen growth media inhibits growth of the larger diatom species.  相似文献

MicroRNAs (miRNAs) are approximately 22 nucleotide RNAs that negatively regulate the expression of protein-coding genes. In a present model of miRNA function in animals, miRNAs that form imperfect duplexes with their targets inhibit protein expression without affecting mRNA levels. Here, we report that in C. elegans, regulation by the let-7 miRNA results in degradation of its lin-41 target mRNA, despite the fact that its 3'UTR regulatory sequences can only partially base-pair with the miRNA. Furthermore, lin-14 and lin-28 are targets of the lin-4 miRNA, and we show that the mRNA levels for these protein-coding genes significantly decrease in response to lin-4 expression. This study reveals that mRNAs containing partial miRNA complementary sites can be targeted for degradation in vivo, raising the possibility that regulation at the level of mRNA stability may be more common than previously appreciated for the miRNA pathway.  相似文献
