The disaccharide L-glycero-D-manno-heptosyl-D-glucose was isolated from the lipopolysaccharide (LPS) of Escherichia coli K-12 strain W3100 after partial hydrolysis with acid, and the structure was determined by methylation analysis, n.m.r. spectroscopy, and comparison with a synthetic standard. In addition, the oligosaccharides L,D-Hep-D-Glc-D-Glc and L,D-Hep-D-Glc-D-Glc-D-Glc were isolated, and their structures were established by g.l.c.-m.s. and methylation analysis. The results indicated that L-glycero-D-manno-heptose, a characteristic constituent of the inner core region, may also occur in the outer core region which, in E. coli, is generally composed of hexoses. A revised structure of the carbohydrate backbone of the hexose/heptose region of the LPS is given.  相似文献

The lactic acid bacteriumLactobacillus casei ssprhamnosus was cultivated in a system with complete cell recycling in order to obtain information on how this cultivation technique affected the microorganisms. Cultivations at two different glucose concentrations (25 g/L and 50 g/L) were performed. Hollow fiber filters were used for separating the cells from the spent broth. The cell recycling was carried out for 128–135 h. Samples were taken three to four times daily for analysis ofd- andl-lactate, glucose concentration, and cell mass. Protein patterns were studied with two-dimensional electrophoresis. A change in the protein pattern was observed. Dry weight of cell mass of 86 g/L was obtained when cultivated on 50 g glucose/L, which was approximately 15 times more than in the batch culture. The percentage ofd-lactate of the total lactate increased with time; when cultivated on 25 g glucose/L, it increased from 3% to 13%. No racemase was detected by the methods used. The data collected from these two recycling experiments show that this is an efficient way to obtain high cell densities, but that the method can affect the product formation pattern of the microorganism.  相似文献

Summary Pseudomonas cepacia ATCC 29351 was cultivated on salicylate as sole carbon source in a system with complete cell recycling by means of a hollow fiber microfilter. Comparisons with batch cultures and continuous cultures are made with respect to cell densities, yield coefficients, specific enzyme activities and volumetric productivities. In batch cultures the toxicity of the salicylate limits the cell densities to below 1 g/l. In a recycling cultivation, a cell concentration of 15.1 g/l was obtained and the volumetric productivity of the cell mass was increased by a factor of 3. Summary Thiobacillus ferrooxidans andAcidianus brierleyi were capable of oxidizing pure pyrite as well as oxidizing sulfur in coal. First order reactions were assumed in the kinetic analysis performed. For oxidation of pure pyrite the rate constant was higher forA. brierleyi than forT. ferrooxidans. An experimental system has been developed that can indirectly measure temperature in a high-intensity microwave field over a broad range of conditions. A RF amplifier connected closed-loop around a high Q cavity oscillates at one of the natural modes of the oven. A bandpass filter selects the mode of interest. As the frozen sample is thawed, an increase in dielectric constant occurs, decreasing the resonance frequency of the cavity. Calibration of the system is performed by measuring the frequency shift for samples whose temperatures are known, Rotation of samples during thawing often causes oscillations of the resonance frequency. These oscillations are generated by asymmetric sample properties and geometry, and hot spots developed during the thaw. Development of a method that would predict hot spot location from these resonance frequency oscillations and permit modulation of the magnetron or sample rotation to minimize thermal runaway is suggested.  相似文献

We studied the molecular forms of somatostatin-like immunoreactivity (SLI), newly released from isolated perfused preparations of the porcine antrum, stomach, pancreas and upper small intestine: Perfusion effluents were concentrated by Sep-Pak C-18 adsorption, eluted with ethanol, dessicated, and subjected to gel filtration with subsequent radioimmunoassays for somatostatin-14 and N-terminal somatostatin-28 immunoreactivity. All the SLI newly released from the stomach and antrum eluted at the position of somatostatin-14, and such was also the case for more than 95% of the SLI newly released from the pancreas, while 68 -/+ 7% and 75 -/+ 8% of the SLI newly released from the isolated perfused jejunum and ileum, respectively, corresponded to somatostatin-28. By reverse phase HPLC the identity of these peptides with synhetic somatostatin-14 and -28 was established.  相似文献

AThermus strain, producing an extracellular protease, was isolated in a hot spring in Iceland. The main growth characteristics of this isolate were studied with different cultivation vessels and different cultivation techniques. A clear and striking dependence of the growth behavior on the cultivation technique was apparent. Higher maximum yield of biomass, higher productivity of biomass, and higher maximum growth rate were found in continuous cultivations compared with batch cultivations. The substrate utilization and the yield of biomass of this strain were much higher than reported for several otherThermus strains. Reproducibility of kinetic data seemed not to depend on the type of cultivation vessel, on the basis of the types of vessels tested, and instability of the population was not observed during cultivations. Production of extracellular protease in our cultivations was apparently growth associated in batch culture, and the specific rate of production in continuous culture was dependent of the dilution rate, implying that certain kinds of regulatory mechanism(s) might be involved.  相似文献
