Clostridium perfringens iota toxin is a binary toxin composed of the enzymatically active component Ia and receptor binding component Ib. Ia is an ADP-ribosyltransferase, which modifies Arg177 of actin. The previously determined crystal structure of the actin-Ia complex suggested involvement of Asp179 of actin in the ADP-ribosylation reaction. To gain more insights into the structural requirements of actin to serve as a substrate for toxin-catalyzed ADP-ribosylation, we engineered Saccharomyces cerevisiae strains, in which wild type actin was replaced by actin variants with substitutions in residues located on the Ia-actin interface. Expression of the actin mutant Arg177Lys resulted in complete resistance towards Ia. Actin mutation of Asp179 did not change Ia-induced ADP-ribosylation and growth inhibition of S. cerevisiae. By contrast, substitution of Glu270 of actin inhibited the toxic action of Ia and the ADP-ribosylation of actin. In vitro transcribed/translated human β-actin confirmed the crucial role of Glu270 in ADP-ribosylation of actin by Ia.  相似文献

Zusammenfassung Während des unter der Trägerschaft des Landesjagdverbandes Baden-Württemberg e. V. seit 1978 laufenden Projektes zur Wiedereinbürgerung des Birkhuhnes im Wurzacher Ried wurde behördlicherseits der Wegfang von Habichten unter bestimmten Auflagen genehmigt. Nach Beringung mußten die Vögel wieder freigelassen werden. Eine vergleichende Untersuchung der von den Projektbetreibern veröffentlichten Berichte und der Beringungsdokumente ergab erhebliche Ungereimtheiten. Die aus den Berichten der Projektbetreiber zu entnehmende Zahl von 168 gefangenen, angeblich beringten und angeblich wieder freigelassenen Habichten steht der Zahl von 98 in den Beringungslisten dokumentierten Vögel gegenüber. Bereits ein Vergleich zwischen der Wiederfundrate der dokumentierten Vögel (1 %) mit der Fänglingfundrate der Vogelwarte Radolfzell (13 %) ergab einen hochsignifikanten Unterschied. Auch der Vergleich mit der Fundrate (15 %) einer anderen, parallel laufenden Verfrachtungsstudie zeigte, daß die Wiederfundrate der Wurzacher Habichte hochsignifikant geringer ist. Die Darstellung der Projektbetreiber, wonach 1978–1981 insgesamt 90 Habichte durch die Landesanstalt für Umweltschutz verfrachtet worden sein sollen, wurde durch die Landesanstalt nicht bestätigt. Die Landesanstalt bestätigte nur fünf verfrachtete Habichte aus dem Wurzacher Projekt. Obwohl der Habichtfang im März nicht erlaubt war, haben die Projektbetreiber insgesamt vier Habichte im März gefangen und zumindest einen (nach eigenen Angaben jedoch alle!) auch noch verfrachtet. Vermutlich im Auftrag von Geflügelzüchtern wurden sieben Habichte, darunter vier Altvögel, innerhalb eines kleinen Gebietes in einer Entfernung von 13–18,5 km vom Wurzacher Ried gefangen. So besteht begründeter Verdacht, daß ein beträchtlicher Teil der im oder um das Wurzacher Ried gefangenen Habichte nicht wieder freigelassen, sondern getötet wurde. Eine Reihe der in den Projektberichten enthaltenen Angaben lassen zudem ganz erhebliche Zweifel an der Fachkompetenz der Projektbetreiber aufkommen.
Goshawk (Accipiter gentilis) trapping in the nature reserve Wurzacher Ried: critical questions on a Black Grouse (Tetrao tetrix) reintroduction project
Summary A Black Grouse reintroduction project was established in 1978 in the nature reserve Wurzacher Ried in southwestern Germany. Since the beginning of this project the staff had a licence to catch, ring and to release but not to kill goshawks. A comparison of the very low Euring-recovery-rate of 1 percent from this project with the Euring-recovery-rate of the Vogelwarte Radolfzell (13 %) showed a significant difference (p=0.00017). In contrast, we found no difference between the recovery-rate (15 %) of a study with almost similar design conducted by the University of Tübingen (Schmidt-Koenig 1982) only 100 km apart from the Wurzacher Ried and the Euring-recovery-rate of the Vogelwarte Radolfzell (p=0.624). The recovery-rate of the reintroduction project differed, however, significantly from the recovery-rate ofSchmidt-Koenig (p=0.006). Furthermore, we found serious inconsistencies between the project reports and the ringing documents concerning this project. Our results indicate, that most of the goshawks caught in the project Wurzacher Ried may not have been released but illegally killed by the project staff.

Knowledge of the spatial scales of diversity is necessary to evaluate the mechanisms driving biodiversity and biogeography in the vast but poorly understood deep sea. The community structure of kinetoplastids, an important group of microbial eukaryotes belonging to the Euglenozoa, from all abyssal plains of the South Atlantic and two areas of the eastern Mediterranean was studied using partial small subunit ribosomal DNA gene clone libraries. A total of 1364 clones from 10 different regions were retrieved. The analysis revealed statistically not distinguishable communities from both the South-East Atlantic (Angola and Guinea Basin) and the South-West Atlantic (Angola and Brazil Basin) at spatial scales of 1000–3000 km, whereas all other communities were significantly differentiated from one another. It seems likely that multiple processes operate at the same time to shape communities of deep-sea kinetoplastids. Nevertheless, constant and homogenous environmental conditions over large spatial scales at abyssal depths, together with high dispersal capabilities of microbial eukaryotes, maintain best the results of statistically indistinguishable communities at larger spatial scales.  相似文献

A functional linkage of the structurally unrelated receptors HER2 and CXCR4 has been suggested for breast cancer but has not been evaluated for esophageal carcinoma. The inhibition of HER2 leads to a reduction of primary tumor growth and metastases in an orthotopic model of esophageal carcinoma. The chemokine receptor CXCR4 has been implicated in metastatic dissemination of various tumors and correlates with poor survival in esophageal carcinoma. The aim of this study was to investigate a correlation between the expression levels of HER2 and CXCR4 and to evaluate the involvemnent of CXCR4-expression in HER2-positive esophageal carcinoma. The effects of HER2-inhibition with trastuzumab and of CXCR4-inhibition with AMD3100 on primary tumor growth, metastatic homing, and receptor expression were evaluated in vitro and in an orthotopic model of metastatic esophageal carcinoma using MRI for imaging. The clinical relevance of HER2- and CXCR4-expression was examined in esophageal carcinoma patients. A significant correlation of HER2- and CXCR4-expression in primary tumor and metastases exists in the orthotopic model. Trastuzumab and AMD3100 treatment led to a significant reduction of primary tumor growth, metastases and micrometastases. HER2-expression was significantly elevated under AMD3100 treatment in the primary tumor and particularly in the metastases. The positive correlation between HER2- and CXCR4-expression was validated in esophageal cancer patients. The correlation of CXCR4- and HER2-expression and the elevation of HER2-expression and reduction of metastases through CXCR4-inhibition suggest a possible functional linkage and a role in tumor dissemination in HER2-positive esophageal carcinoma.  相似文献

All TGF-beta family members have a prodomain that is important for secretion. Lack of secretion of a TGF-beta family member GDF5 is known to underlie some skeletal abnormalities, such as brachydactyly type C that is characterized by a huge and unexplained phenotypic variability. To search for potential phenotypic modifiers regulating secretion of GDF5, we compared cells overexpressing wild type (Wt) GDF5 and GDF5 with a novel mutation in the prodomain identified in a large Pakistani family with Brachydactyly type C and mild Grebe type chondrodyslplasia (c527T>C; p.Leu176Pro). Initial in vitro expression studies revealed that the p.Leu176Pro mutant (Mut) GDF5 was not secreted outside the cells. We subsequently showed that GDF5 was capable of forming a complex with latent transforming growth factor binding proteins, LTBP1 and LTBP2. Furthermore, secretion of LTBP1 and LTBP2 was severely impaired in cells expressing the Mut-GDF5 compared to Wt-GDF5. Finally, we demonstrated that secretion of Wt-GDF5 was inhibited by the Mut-GDF5, but only when LTBP (LTBP1 or LTBP2) was co-expressed. Based on these findings, we suggest a novel model, where the dosage of secretory co-factors or stabilizing proteins like LTBP1 and LTBP2 in the microenvironment may affect the extent of GDF5 secretion and thereby function as modifiers in phenotypes caused by GDF5 mutations.  相似文献

Cyclin D2 is the only D-type cyclin expressed in mature mouse B-lymphocytes, and its expression is associated with retinoblastoma protein (pRB) and pRB-related protein phosphorylation and induction of E2F activity, as B-cells enter the cell cycle following stimulation via surface IgM and/or CD40. Cyclin D-dependent kinase activity is required for cell proliferation, yet cyclin D2(-/-) mice have normal levels of mature B-lymphocytes. Here we show that B-lymphocytes from cyclin D2(-/-) mice can proliferate in response to anti-IgM and anti-CD40, but the time taken to enter S-phase is longer than for the corresponding cyclin D2(+/+) cells. This is due to the compensatory induction of cyclin D3, but not cyclin D1, which causes pRb phosphorylation on CDK4-specific sites. This is the first demonstration that loss of a D-type cyclin causes specific expression and functional compensation by another member of the family in vivo and provides a rationale for the presence of mature B-lymphocytes in cyclin D2(-/-) mice.  相似文献

Fifteen species of Heterostropha are described, 12 of them for the first time. All are newly interpreted with regard to their taxonomic relation to fossil and living gastropods. The Streptacidoidea with long Paleozoic history are represented in the Late Triassic St. Cassian Formation by several genera that can be differentiated into four families. The Ebalidae are represented byEbala, with smooth protoconch, Cassianebalidae byCassianebala andLoxebala with axially ornamented protoconch. The Donaldinidae of St. Cassian are represented by one species ofDonaldina and two ofNeodonaldina that stand in the continuation of Paleozoic species ofDonaldina. Architectonicoidea with shells coiled in a plane and Valvatoidea appear in the St. Cassian fauna without known Paleozoic relation. In the former superfamily the Architectonicidae can be recognized in the genusRinaldoconchus with two species. Cassianaxidae withCassianaxis, Amphitomariidae withAmphitomaria, Stuoraxidae withStuoraxis andAmpezzogyra have a sinistral protoconch and planispirally coiled dextral teleconchs. They all resemble different modern species that have similarly small shells. Modern Hyalogyrinidae have withAlexogyra a new representative from the Triassic. The Valvatoidea are represented with the generaCarboninia andBandellina of the Cornirostridae in the Triassic representatives. The relation of described species in the system of the Heterostropha is discussed.  相似文献

Dynamics and possible function of the lutein epoxide (Lx) cycle, that is, the reversible conversion of Lx to lutein (L) in the light-harvesting antennae, were investigated in leaves of tropical tree species. Photosynthetic pigments were quantified in nine Inga species and species from three other genera. In Inga , Lx levels were high in shade leaves (mostly above 20 mmol mol−1 chlorophyll) and low in sun leaves. In Virola surinamensis , both sun and shade leaves exhibited very high Lx contents (about 60 mmol mol−1 chlorophyll). In Inga marginata grown under high irradiance, Lx slowly accumulated within several days upon transfer to deep shade. When shade leaves of I. marginata were briefly exposed to the sunlight, both violaxanthin and Lx were quickly de-epoxidized. Subsequently, overnight recovery occurred only for violaxanthin, not for Lx. In such leaves, containing reduced levels of Lx and increased levels of L, chlorophyll fluorescence induction showed significantly slower reduction of the photosystem II electron acceptor, Q A, and faster formation as well as a higher level of non-photochemical quenching. The results indicate that slow Lx accumulation in Inga leaves may improve light harvesting under limiting light, while quick de-epoxidation of Lx to L in response to excess light may enhance photoprotection.  相似文献
