顶空固相微萃取/气相色谱/质谱法分析河南产紫丁香挥发性成分

采用顶空固相微萃取和气质联用技术（HS—SPME—GC—MS）,首次分析了河南产紫丁香花营和花的挥发性成分。从紫丁香花蕾和花中共鉴定了65种组分,其中17个成分是二者共有的;酯类化合物只存在于花蕾中,而酸类和醇类化合物在花中的含量较高。这表明酯类化合物在紫丁香花开放的过程中可能逐步被植物体内的水解酶水解成酸类和醇类化合物。     

银杏内生真菌的分离及其对苹果腐烂病病原菌的拮抗作用

采用组织块分离法、单菌丝挑取法,从采自3个不同地点的银杏(Ginkgo biloba L.)叶和茎部中分离出16株内生真菌,对其进行了抗菌活性的初步研究.结果表明:有9株能够抑制苹果腐烂病病原菌的生长,其中4株菌对病原菌有显著的抑制作用,并大于同样条件下拮抗培养的瑞氏木霉的抑菌效果.与苹果腐烂病病原菌进行两点对峙培养,经镜检发现,与拮抗内生菌一起培养的病原菌的菌丝生长都出现不同程度的畸形和断裂.可见,银杏中的内生真菌对苹果腐烂病有明显的抑制作用.     

啤酒酵母生产的重组水蛭素的纯化及脱色

对啤酒酵母生产的重组水蛭素变异体1(rHV1)进行多步骤的纯化。首先将分泌到培养上清液中的水蛭素进行硫酸铵沉淀和SephadexG-50凝胶过滤,再用Q-SepharoseHP阴离子交换层析分离,最后用HPLCSP-5PW阳离子交换柱脱色及HPLCC8柱反相层析。真空干燥后得到的水蛭素蛋白经SPS-PAGE、N端氨基酸序列分析、抗凝血酶活力分析鉴定,证明已获得高纯度的重组水蛭素HV1制剂,为利用基因工程方法生产重组水蛭素的规模化生产及临床应用提供了依据     

Beyond ecosystem modeling: A roadmap to community cyberinfrastructure for ecological data‐model integration

In an era of rapid global change, our ability to understand and predict Earth's natural systems is lagging behind our ability to monitor and measure changes in the biosphere. Bottlenecks to informing models with observations have reduced our capacity to fully exploit the growing volume and variety of available data. Here, we take a critical look at the information infrastructure that connects ecosystem modeling and measurement efforts, and propose a roadmap to community cyberinfrastructure development that can reduce the divisions between empirical research and modeling and accelerate the pace of discovery. A new era of data‐model integration requires investment in accessible, scalable, and transparent tools that integrate the expertise of the whole community, including both modelers and empiricists. This roadmap focuses on five key opportunities for community tools: the underlying foundations of community cyberinfrastructure; data ingest; calibration of models to data; model‐data benchmarking; and data assimilation and ecological forecasting. This community‐driven approach is a key to meeting the pressing needs of science and society in the 21st century.     

树干毕赤酵母酒精发酵碳元素代谢流向分析

实验对树干毕赤酵母（Pichia stipitis）进行了4个阶段共400 h连续恒化培养,在不同阶段以30.0 g/L葡萄糖作为基本碳源,添加30.0或15.0 g/L的木糖,通过控制温度（35±1）℃,进气量100～150mL/min,搅拌转速250～300 r/min。4个阶段共建立4个连续培养的"稳态"。对碳元素进行物料衡算发现,四个阶段碳元素回收率分别为118.0 %、105.6 %、113.5 %和94.7 %。对4个近似"稳态"的碳元素的代谢流向进行分析发现：将近50.0 %左右碳元素流向产物酒精,其次是CO2和酵母细胞;木糖醇浓度与流入底物中木糖浓度有直接关系,在相同发酵条件下流入的木糖浓度越大代谢生成木糖醇浓度也越高;实验所采用的通气条件更适合底物为30.0 g/L葡萄糖和30.0 g/L木糖混合液的连续发酵。     

Human CD34+ HLA-DR- bone marrow cells contain progenitor cells capable of self-renewal, multilineage differentiation, and long-term in vitro hematopoiesis.

Human bone marrow cells expressing CD34 but not HLA-DR were isolated by immunofluorescence flow cytometric cell sorting. These cells contained a hematopoietic cell (CFU-B1) capable of producing, in an in vitro semisolid culture system, blast-cell-containing colonies, which possessed the capacity for self-renewal and commitment to multipotential differentiation. In addition, CD34+ HLA-DR- marrow cells contained primitive megakaryocyte progenitor cells, the burst-forming unit-megakaryocyte (BFU-MK). A subset of CD34+ HLA-DR- marrow cells lacking the expression of CD15 and CD71 was obtained by flow cytometric cell sorting and was capable of sustaining in vitro hematopoiesis in suspension culture for up to 8 weeks in the absence of a preestablished adherent marrow cell layer. The combination of IL-3 + IL-1 alpha and IL-3 + IL-6 sustained proliferation of these cells for 8 weeks, induced maximal cellular expansion, and increased the numbers of assayable progenitor cells. These studies demonstrate that human CD34+ HLA-DR- marrow cells and their subsets contain primitive multipotential hematopoietic cells capable of self-renewal and of differentiation into multiple hematopoietic lineages.     

聚丙烯酰胺凝胶电泳中LPS样品制备方法的改进

为了对革兰阴性细菌的LPS进行快速分析,观察LPS的产量、分子量分布,进行菌种筛选和抗原传代稳定性研究,实验中对热酚法、酶水解法、冷酚法三种制备LPS的方法进行了比较。结果证明,改进的水饱和冷酚LPS提取方法,需要时间短、经济、方便,只须几个细菌菌落就可以提取LPS进行SDS-PAGE电泳分析,说明该方法有很好的实用性。     

延缓番茄果实成熟的分子机理

对延缓番茄果实成熟的热处理、反义ＲＮＡ技术及气调技术（高ＣＯ２／低Ｏ２,１００％Ｎ２）等方法进行了阐述,并从分子水平上（包括与乙烯生成有关及无关的蛋白质的合成、基因表达等方面）探讨了这些方法延缓果实成熟的机理。
