This work aims to contribute to the number of urban metabolism case studies using a standardized methodology. An economy‐wide material flow analysis (EW‐MFA) was conducted on the Metropolitan Municipality of Cape Town (South Africa) for the year 2013, using the Eurostat framework. The study provides insights into the city's metabolism through various indicators including direct material input (DMI), domestic material consumption (DMC), and direct material output (DMO), among others. In order to report on the uncertainty of the data, a set of data quality indicators originating from the life cycle assessment literature was used. The results show that domestic extraction involves significant quantities of non‐metallic minerals, and that imports consist primarily of biomass and fossil fuels. The role of the city as a regional hub is also made clear from this study and illustrated by large quantities of food and other materials flowing through the city on their way to or from international markets. The results are compared with indicators from other cities and with previous metabolism work done on Cape Town. To fully grasp the impacts of the city's metabolism, more work needs to be done. It will be necessary to understand the upstream impact of local consumption, and consumption patterns should be differentiated on a more nuanced level (taking into account large differences between household income levels as well as separating the metabolism of industry and commerce from residential consumption). 相似文献

Insects are major conduits of resources moving from aquatic to terrestrial systems. While the ecological impacts of insect subsidies are well documented, the underlying mechanisms by which these resources change recipient ecosystems remain poorly understood. Most subsidy inputs enter terrestrial systems as detritus; thus, soil microbes will likely influence the processing of insect subsidies, with implications for plant community composition and net primary productivity (NPP). In a subarctic ecosystem near Lake Mývatn, Iceland where midge (Diptera: Chironomidae) deposition to land is high, we investigated how insect subsidies affected litter processing and microbial communities. We also evaluated how those belowground effects related to changes in inorganic nitrogen, plant composition and NPP. We simulated subsidies by adding midge carcasses to 1-m2 heathland plots, where we measured effects on decomposition rates and the plant community. We then studied how fertilization treatments (control, KNO3 and midge-carcass addition) affected graminoid biomass and inorganic nitrogen in greenhouse experiments. Lastly, we conducted a soil-incubation study with a phospholipid fatty acid analysis (PLFA) to examine how midge addition to heathland soils affected microbial respiration, biomass and composition. We found that midge addition to heathland soils increased litter decomposition and graminoid plant cover by 2.6× and 2×, respectively. Greenhouse experiments revealed similar patterns, with midge carcasses increasing graminoid biomass by at least 2× and NH4+ concentrations by 7×. Our soil-incubation study found that midge carcasses elevated microbial respiration by 64%, microbial biomass by 43% and shifted microbial functional composition. Our findings indicate that insect subsidies can stimulate soil microbial communities and litter decomposition in subarctic heathlands, leading to increased NPP and changes in plant community composition. 相似文献

Warp2D is a novel time alignment approach, which uses the overlapping peak volume of the reference and sample peak lists to correct misleading peak shifts. Here, we present an easy-to-use web interface for high-throughput Warp2D batch processing time alignment service using the Dutch Life Science Grid, reducing processing time from days to hours. This service provides the warping function, the sample chromatogram peak list with adjusted retention times and normalized quality scores based on the sum of overlapping peak volume of all peaks. Heat maps before and after time alignment are created from the arithmetic mean of the sum of overlapping peak area rearranged with hierarchical clustering, allowing the quality control of the time alignment procedure. The pathogenesis of the human muscular dystrophies is unknown, and several competing hypotheses have been proposed. The vascular hypothesis states that muscle fibre necrosis occurs in dystrophy as a result of transient muscle ischemia. Although abnormalities of the vascular system may be demonstrated in dystrophy, their role in pathogenesis remains obscure. The responses to serotonin (5-HT) and noradrenaline (NA) were examined in isolated ischiatic artery preparations from normal and genetically dystrophic chickens. The tension generated in response to 5-HT was greater in arteries from normal chickens than in arteries from dystrophic chickens, whereas responses to NA were similar. Analysis of the concentration-response relationships demonstrated that the dystrophic ischiatic artery was less sensitive to 5-HT than was the normal artery, although the sensitivity to NA was similar in both vessels. The results of this study are not consistent with the view that muscle fibre necrosis in avian dystrophy is a consequence of muscle anoxia. These data do demonstrate pharmacological differences between dystrophic avian arteries and arteries from normal chickens, but their presence may represent merely the expression of dystrophy in vascular smooth muscle. 相似文献

The main pathways for the repair of DNA double strand breaks (DSBs) are non-homologous end-joining (NHEJ) and homologous recombination directed repair (HDR). These operate mutually exclusive and are activated by 53BP1 and BRCA1, respectively. As HDR can only succeed in the presence of an intact copy of replicated DNA, cells employ several mechanisms to inactivate HDR in the G1 phase of cell cycle. As cells enter S-phase, these inhibitory mechanisms are released and HDR becomes active. However, during DNA replication, NHEJ and HDR pathways are both functional and non-replicated and replicated DNA regions co-exist, with the risk of aberrant HDR activity at DSBs in non-replicated DNA. It has become clear that DNA repair pathway choice depends on inhibition of DNA end-resection by 53BP1 and its downstream factors RIF1 and MAD2L2. However, it is unknown how MAD2L2 accumulates at DSBs to participate in DNA repair pathway control and how the NHEJ and HDR repair pathways are appropriately activated at DSBs with respect to the replication status of the DNA, such that NHEJ acts at DSBs in pre-replicative DNA and HDR acts on DSBs in post-replicative DNA. Here we show that MAD2L2 is recruited to DSBs in H4K20 dimethylated chromatin by forming a protein complex with 53BP1 and RIF1 and that MAD2L2, similar to 53BP1 and RIF1, suppresses DSB accumulation of BRCA1. Furthermore, we show that the replication status of the DNA locally ensures the engagement of the correct DNA repair pathway, through epigenetics. In non-replicated DNA, saturating levels of the 53BP1 binding site, di-methylated lysine 20 of histone 4 (H4K20me2), lead to robust 53BP1-RIF1-MAD2L2 recruitment at DSBs, with consequent exclusion of BRCA1. Conversely, replication-associated 2-fold dilution of H4K20me2 promotes the release of the 53BP1-RIF1-MAD2L2 complex and favours the access of BRCA1. Thus, the differential H4K20 methylation status between pre-replicative and post-replicative DNA represents an intrinsic mechanism that locally ensures appropriate recruitment of the 53BP1-RIF1-MAD2L2 complex at DNA DSBs, to engage the correct DNA repair pathway. 相似文献

Background
Quantifying the amount of standing genetic variation in fitness represents an empirical challenge. Unfortunately, the shortage of detailed studies of the genetic architecture of fitness has hampered progress in several domains of evolutionary biology. One such area is the study of sexual selection. In particular, the evolution of adaptive female choice by indirect genetic benefits relies on the presence of genetic variation for fitness. Female choice by genetic benefits fall broadly into good genes (additive) models and compatibility (non-additive) models where the strength of selection is dictated by the genetic architecture of fitness. To characterize the genetic architecture of fitness, we employed a quantitative genetic design (the diallel cross) in a population of the seed beetle Callosobruchus maculatus, which is known to exhibit post-copulatory female choice. From reciprocal crosses of inbred lines, we assayed egg production, egg-to-adult survival, and lifetime offspring production of the outbred F1 daughters (F1 productivity). 相似文献78.

Infectious bursal disease virus (IBDV) is the major viral pathogen in the poultry industry. Live attenuated serotype 1 vaccine strains are commonly used to protect susceptible chickens during their first 6 weeks of life. Wild-type serotype 1 IBDV strains are highly pathogenic only in chickens, whereas serotype 2 strains are apathogenic in chickens and other birds. Here we describe the replacement of the genomic double-stranded RNA (dsRNA) encoding the N- or C-terminal part of VP3 of serotype 1 very virulent IBDV (vvIBDV) (isolate D6948) with the corresponding part of serotype 2 (isolate TY89) genomic dsRNA. The modified virus containing the C-terminal part of serotype 2 VP3 significantly reduced the virulence in specific-pathogen-free chickens, without affecting the distinct bursa tropism of serotype 1 IBDV strains. Furthermore, by using serotype-specific antibodies we were able to distinguish bursas infected with wild-type vvIBDV from bursas infected with the modified vvIBDV. We are currently evaluating the potential of this recombinant strain as an attenuated live vaccine that induces a unique serological response (i.e., an IBDV marker vaccine). 相似文献
