The relationship of the pH-dependent and concentration-dependent dissociation of porcine heart mitochondrial malate dehydrogenase (L-malate: NAD+ oxidoreductase, EC 1.1.1.37) was investigated by means of gel filtration chromatography utilizing a standardized Sephacryl S-200 column. The results obtained indicate that the dimeric form of this enzyme dissociates to yield monomers at conditions of low protein concentration or at pH values below neutrality. In addition it is apparent that as the pH is lowered, the minimum concentration of protein required to maintain the enzyme in the dimeric form is increased.  相似文献

Enteropathogenic Escherichia coli (EPEC) uses a type 3 secretion system to transfer effector proteins into the host intestinal epithelial cell. Several effector molecules contribute to tight junction disruption including EspG1 and its homologue EspG2 via a mechanism thought to involve microtubule destruction. The aim of this study was to investigate the contribution of EspG‐mediated microtubule disruption to TJ perturbation. We demonstrate that wild type EPEC infection disassembles microtubules and induces the progressive movement of occludin away from the membrane and into the cytosol. Deletion of espG1/G2 attenuates both of these phenotypes. In addition, EPEC infection impedes barrier recovery from calcium switch, suggesting that inhibition of TJ restoration, not merely disruption, prolongs barrier loss. TJs recover more rapidly following infection with ΔespG1/G2 than with wild type EPEC, demonstrating that EspG1/G2 perpetuate barrier loss. Although EspG regulates ADP‐ribosylation factor (ARF) and p21‐activated kinase (PAK), these activities are not necessary for microtubule destruction or perturbation of TJ structure and function. These data strongly support a role for EspG1/G2 and its associated effects on microtubules in delaying the recovery of damaged tight junctions caused by EPEC infection.  相似文献

Experiments are presented to show that the phosphorylation of the light-harvesting chlorophyll ab-protein complex (LHC) induces structural reorganisation within the thylakoid membrane in response to the introduction of additional negative surface charges. The effect of cations of different valency on chlorophyll fluorescence measurements indicates that LHC-phosphorylation-induced reorganisation involves a change in the electrostatic screening capability of the added cation. At intermediate levels of cations (e.g., 1 or 2 mM Mg2+), which substantially stack non-phosphorylated membranes, it was found that membrane phosphorylation caused considerable unstacking as monitored by light scattering and electron microscopy. Concomitant with this was a large decrease in chlorophyll fluorescence indicative of randomisation of chlorophyll protein complexes which would result in an increase in energy transfer between the photosystems as well as an absorption cross-section change. At higher concentrations (e.g., above 5 mM Mg2+) a persistent ATP-induced decrease in chlorophyll fluorescence has been attributed to the displacement of charged phosphorylated LHC from the appressed granal to the non-appressed stromal lamellae, thus decreasing the absorption cross-section of Photosystem II. Under these circumstances only a small degree of unstacking was detected by light scattering and measurements of the percentage of thylakoid length which is stacked to form grana. However, when considered on a surface area basis, the structural changes observed can qualitatively account for the magnitude of the chlorophyll fluorescence quenching due to the lateral diffusion of LHC.  相似文献

Biological indicators were prepared using either Bacillus stearothermophilus or Bacillus subtilis incorporated into lyophilized calcium alginate beads. These were subjected to a variety of moist heat and dry heat treatments as appropriate and then dissolved in buffer to allow enumeration of survivors. The utility of these spore carriers as biological indicators for the quantitative assessment of heat sterilization processes is discussed.  相似文献

Rapid freeze-thaw injury to erythrocytes and erythrocyte ghosts has been shown to be strongly cation dependent. For the Group I ions this dependence is nonmonotonic in nature with injury increasing in the order Li+ less than Na+ less than Cs+ less than K+. Injury can be reduced by the inclusion in the freezing media of saccharide cryoprotectants or by the substitution with less injurious cations, e.g., Mg2+ or (CH3)4N+. In contrast to the situation observed with cations injury with anions follows Hofmeister lyotropic power series with injury increasing with decreasing hydrated ionic radius. Careful choice of electrolyte species allows injury to be reduced to levels comparable to that afforded by saccharide cryoprotectants. A possible mechanism for the nonmonotonic trends in injury observed with cations is considered.  相似文献

Abstract: This study examines reproductive efficiency in the common marmoset monkey, Callithrix jacchus, using sequential ultrasound examinations to establish ovulation number, implantation rate, and incidence of prenatal loss. Ultrasound was carried out with a 10 or 7.5 mHz probe in nonsedated animals, daily during the late follicular phase, approximately twice a week until day 20 after ovulation, and at days 35, 56, and 85 of pregnancy to enable visualization of gestation sacs, heartbeats, and fetal heads, respectively. Ovulatory follicles could be seen 3–4 days before ovulation and by day –2, 98% of ovulating follicles were >2mm diameter, although almost 10% of follicles of this size disappeared without ovulating. Total number of ovulating follicles for 15 females was 45 (mean ovulation rate ? 3.0, range 2–4). In the 14 animals that conceived, 41 corpora lutea were identified (mean ovulation rate = 2.9) within 10 days of ovulation. All pregnancies went to term (no abortion occurred) resulting in the birth of 37 neonates (9 triplets, 5 twins) and an average litter size of 2.64. All four losses were confined to the embryonic period (< day 85), two occurring before day 35, one between days 35 and 56, and one between days 56 and 85. In demonstrating that 90% of ovulatory follicles gave rise to live offspring, the results of this study indicate an extremely high reproductive efficiency in the marmoset monkey (when maintained under favorable captive conditions) and a rate of prenatal loss much lower than that reported for other primate species.  相似文献
