The molecular mass of a biomolecule is characterized by the monoisitopic mass Mmono and the average isotopic mass Mav. We found that tryptic peptide masses mapped on a plane made by two parameters derived from Mmono and Mav form a peculiar feature in the form of a ‘band gap’ stretching across the whole ‘peptide galaxy’, with a narrow line in the
centre. The purpose of this study was to investigate possible reasons for the emergence of such a feature, provided it is
not a random occurrence. The a priori probability of such a feature to emerge by chance was found to be less than 1:100. Peptides contributing to the central line
have elemental compositions following the rules S = 0; Z = C - (N + H)/2 = 0, which nine out of 20 amino acid residues satisfy.
The relative abundances of amino acids in the peptides contributing to the central line correlate with the consensus order
of emergence of these amino acids, with ancient amino acids being overrepresented in on-Line peptides. Since linear correlation
between Mav and Mmono reduces the complexity of polypeptide molecules, and the turnover rate of less complex molecules should be faster in non-equilibrium
abiotic synthesis, we hypothesize that the line could be a signature of abiotic production of primordial biopolymers. The
linear dependence between the average isotopic masses and monoisotopic masses may have influenced the selection of amino acid

A novel human fibroblast growth factor (hFGF), which shows 75% sequence homology with fibroblast growth factor-9, was isolated in random sequencing of a human heart cDNA library. The full-length sequence is 928 bp, the encoded protein is composed of 168 amino acid residues, and its pI value and molecular weight were estimated to be 8.13 and 19.1 kDa, respectively. RT-PCR using Marathon human heart cDNA shows that the coding region is approximately 507 bp. Southern hybridization showed a single band which indicates that this is a single copy gene. Northern hybridization done on a human multiple tissues blot showed that the gene is preferentially expressed in human heart, very weakly detectable in human brain and not detectable in 18 other different human tissues. 相似文献

Post-translational modification by small ubiquitin-like modifier 1 (SUMO-1) is a highly conserved process from yeast to humans and plays important regulatory roles in many cellular processes. Sumoylation occurs at certain internal lysine residues of target proteins via an isopeptide bond linkage. Unlike ubiquitin whose carboxyl-terminal sequence is RGG, the tripeptide at the carboxyl terminus of SUMO is TGG. The presence of the arginine residue at the carboxyl terminus of ubiquitin allows tryptic digestion of ubiquitin conjugates to yield a signature peptide containing a diglycine remnant attached to the target lysine residue and rapid identification of the ubiquitination site by mass spectrometry. The absence of lysine or arginine residues in the carboxyl terminus of mammalian SUMO makes it difficult to apply this approach to mapping sumoylation sites. We performed Arg scanning mutagenesis by systematically substituting amino acid residues surrounding the diglycine motif and found that a SUMO variant terminated with RGG can be conjugated efficiently to its target protein under normal sumoylation conditions. We developed a Programmed Data Acquisition (PDA) mass spectrometric approach to map target sumoylation sites using this SUMO variant. A web-based computational program designed for efficient identification of the modified peptides is described. 相似文献

A liquid chromatography-full scan high resolution accurate mass spectrometry (LC-HRMS) method for quantifying prednisone and prednisolone in human plasma using a quadrupole time-of-flight mass spectrometer (Q-TOF) was developed. Plasma samples were extracted using a liquid-liquid extraction procedure. Full scan data were acquired in the TOF only mode and extracted ion chromatograms were generated post-acquisition with the exact masses of the analytes. The calibration range was 5-2500 ng/mL, with a Lower Limit of Quantitation (LLOQ) of 5 ng/mL. The assay accuracy was between 98.4% and 106.3%. The between-run (inter-day) and within-run (intra-day) precision were within 1.7% and 2.9%, respectively. The matrix effect was between 0.98 and 1.10 for the six different lots of human plasma evaluated. Pooled incurred samples were analyzed by the method and the results matched those obtained from an LC-MS/MS method. In addition, qualitative information on phospholipids, and other endogenous components were also extracted from the full-scan data acquired. 相似文献

A wide variety of vanadium-containing complexes have been tested, both in vivo and in vitro, as possible therapeutic agents for the oral treatment of type 2 diabetes mellitus. None so far has surpassed bis(maltolato)oxovanadium(IV) (BMOV) for glucose- and lipid-lowering in an orally available formulation. Ligand choice is clearly an important factor in pharmacological efficacy of vanadium compounds as insulin enhancing agents. In this study, we kept the ligand and dose the same, varying instead the metal ion bound to the maltolato ligand in a series of binary complexes of neutral charge. A requirement for vanadyl ion as the metal ion of choice was apparent; no other metal ion tested served as a suitable substitute. Amongst [MoO(2)](2+), Co(II), Cu(II), Cr(III), and Zn(II), only [MoO(2)](2+) and Co(II) showed any hypoglycemic activity at the ED(50) dose for bis(maltolato)oxovanadium(IV), 0.6 mmolkg(-1) by oral gavage in streptozotocin (STZ)-diabetic rats within 72 h of administration of compound. 相似文献

Field and laboratory experiments compared the feeding behaviours of two marine scavengers, the gastropod, Nassarius festivus and the hermit crab, Diogenes edwardsii, sympatric on the lower intertidal shore at Starfish Bay, Hong Kong. Field trials demonstrated that they both preferred bivalve and fish to other carrion. N. festivus arrived at bait in greater numbers and more quickly than D. edwardsii. Numbers of the former species attracted to fish bait (1 cm(3)) peaked at a mean of 41 after 30 min, whereas the latter peaked at a mean of only 2.3 after 55 min. Laboratory investigations revealed that N. festivus80 cm, feed quickly (x=13.20 min) and, after 14 days starvation, had a high consumption rate (0.124 mg wet weight or 0.034 mg dry weight of food individual(-1) min(-1), or 1.4% and 1.3% of its wet and dry body weights min(-1), respectively). In contrast, D. edwardsii119.75 min and a lower consumption rate (0.003 mg wet weight and 0.001 mg dry weight of food individual(-1) min(-1), or 0.1% of its wet and dry body weights min(-1), respectively). Manipulative experiments provided evidence for interspecific competition for carrion between the two species. A morphological advantage, i.e. an extendible proboscis allowing feeding at a distance, and chemoreceptors that permit long distance food detection, combined with numerical superiority on the shore, are mechanisms by which N. festivus outcompetes D. edwardsii and dominates feeding clusters. Interactive behaviour studies revealed the occurrence of interspecific interactions between the two species and intraspecific interactions among D. edwardsii but not N. festivus. The number of interspecific interactions fell when the numbers of N. festivus increased and those of D. edwardsii decreased, and vice versa. The aggressive D. edwardsii did not significantly affect the feeding behaviour of N. festivus. When the number of D. edwardsii increased and the number of N. festivus decreased, the number of intraspecific interactions in the former increased. The feeding time of N. festivus, however, decreased with increasing numbers of conspecifics and a decreasing number of D. edwardsii.
