A new cell line (NOE) of human ovarian endometrioid carcinoma was established and characterized. The cell line showed a short spindle-shaped morphology and continued to grow for more than 4 months without contact inhibition. The doubling time was approximately 15.5 h at the 10th passage. The chromosome number was aneuploid. The IC50 values of paclitaxel, cisplatin and carboplatin were 26.4 ng/mL, 2.4 microg/mL and 32.5 microg/mL, respectively. The NOE cells expressed estrogen receptor alpha. In nude mice, we confirmed tumor formation of NOE cells. These results indicated that NOE cells showed similar chemosensitivity and properties to those of the original tumor and might be useful in basic studies on the diagnosis, treatment and etiology of ovarian tumors. 相似文献

At an early stage of growth gametophytes support the sporophytes of ferns. Young sporophytes become independent of gametophytes
when the first leaves develop. Although large fern gametophytes produce multiple archegonia simultaneously, only one sporophyte
is typically established on one gametophyte. The number of sporophytes is believed to be controlled in two possible directions,
from gametophyte to sporophyte or from preceding sporophyte to another sporophyte. To investigate the effects of gametophytes
on their sporophytes, we studied the relationship between organic matter production by gametophytes and the growth of young
sporophytes of Thelypteris palustris. We cut gametophytes in half (CGs) to reduce the gametophytes’ production of matter. There was no significant difference
between the growth of sporophytes on intact gametophytes (IGs) and that on CGs. According to our estimates, based on the rate
of organic matter production, the large gametophyte was able to produce two or more sporophytes. The resources required for
CGs to make similar-sized sporophytes was twice that for IGs. In polyembryony each of the multiple sporophytes was similar
in size to the single sporophytes. Resource limitation does not seem to explain why fern gametophytes establish single sporophytes. 相似文献

Human and rabbit platelets fully aggregated byplatelet-activating factor (PAF) underwent slow disaggregation but wererapidly disaggregated by the PAF receptor antagonists WEB-2086,Y-24180, SM-12502, and CV-3988. Whereas the1-alkyl-2-[3H]acetyl-sn-glycero-3-phosphocholine([3H]acetyl-PAF)specifically bound to platelet receptors underwent slow and spontaneousdissociation, it dissociated promptly from its receptor when WEB-2086was added, in parallel with platelet disaggregation and disappearanceof P-selectin on the cell surface. Extracellular[3H]acetyl-PAF wasrapidly deacetylated by normal rabbit platelets; some of the[3H]acetyl-PAF wasbound to the cells and a very small amount of [3H]acetate wasdetected in the cells. In contrast, when1-[3H]alkyl-2-acetyl-sn-glycero-3-phosphocholinewas added to the platelets, the radioactivity was rapidly incorporatedinto the 1-alkyl-2-acyl-sn-glycero-3-phosphocholinefraction. These results indicate that1) continuous binding of PAF to itsreceptor is necessary for prolonged platelet aggregation, which may bemediated through an unknown signaling system for a long-term cellresponse rather than a transient signaling system, and2) most of the[3H]acetyl-PAF boundto platelets is metabolized extracellularly by ecto-type PAFacetylhydrolase, with the lyso-PAF generated being incorporated rapidlyinto the cells and converted to1-alkyl-2-acyl-sn-glycero-3-phosphocholine. 相似文献

Secondary pulmonary hypertension is a frequent condition after heart valve surgery. It may significantly complicate the perioperative management and increase patients' morbidity and mortality. The treatment has not been yet completely defined principally because of lack of the selectivity of drugs for the pulmonary vasculature. The usage of inhaled milrinone could be the possible therapeutic option. Inodilator milrinone is commonly used intravenously for patients with pulmonary hypertension and ventricular dysfunction in cardiac surgery. The decrease in systemic vascular resistance frequently necessitates concomitant use of norepinephrine. Pulmonary vasodilators might be more effective and also devoid of potentially dangerous systemic side effects if applied by inhalation, thus acting predominantly on pulmonary circulation. There are only few reports of inhaled milrinone usage in adult post cardiac surgical patients. We reported 2 patients with severe pulmonary hypertension after valve surgery. Because of desperate clinical situation, we decided to use the combination of inhaled and intravenous milrinone. Inhaled milrinone was delivered by means of pneumatic medication nebulizer dissolved with saline in final concentration of 0.5 mg/ml. The nebulizer was attached to the inspiratory limb of the ventilator circuit, just before the Y-piece. We obtained satisfactory reduction in mean pulmonary artery pressure in both patients, and they were successfully extubated and discharged. Although it is a very small sample of patients, we conclude that the combination of inhaled and intravenous milrinone could be an effective treatment of secondary pulmonary hypertension in high-risk cardiac valve surgery patient. The exact indications for inhaled milrinone usage, optimal concentrations for this route, and the beginning and duration of treatment are yet to be determined. 相似文献

alpha 2-Adrenergic receptors labelled by [3H]-clonidine (alpha 2-agonist) can be solubilized from the rat brain in a form sensitive to guanine nucleotides with a zwitterionic detergent, 3-[3-(cholamidopropyl)-dimethylammonio]-1-propane sulfonate (CHAPS). About 40% of the original [3H]CLO binding sites in the membranes were solubilized with 6 mM CHAPS. Separation of the soluble [3H]CLO-bound complex was performed by the vacuum filtration method using polyethylenimine-treated GF/B filters. Solubilized [3H]CLO binding sites retained the same pharmacological characteristics of membrane-bound alpha 2-adrenergic receptors. Scatchard plots of [3H]CLO binding to solubilized alpha 2-receptors were curvilinear, indicating the existence of the two distinct binding components. Solubilized receptors were eluted as a single peak from Bio-Gel A-1.5 m column with a Stokes radius of 6.6 nm. The isoelectric point was 5.6-5.8. Regulations of the receptor binding by guanine nucleotides, monovalent cations, and sulfhydryl-reactive agents were maintained intact in the soluble state, whereas those by divalent cations were lost. The apparent retention of receptors and guanine nucleotide binding regulatory component(s) in the soluble state may allow a investigation of the regulation mechanisms of the brain alpha 2-adrenergic receptor system at the molecular level. 相似文献

Photosynthetic activity and dark respiration were measured in some species of lichens,Umbilicaria, Cladonia, Stereocaulon, Usnea, etc. sampled in the Antarctic and subalpine zones of central and northern Japan. On the basis of the responses of their
activities to thallus temperature, the response patterns were divived into three groups, and further, each group was subdvided
into some adaptation types for net photosynthesis and repiration. Lichens collected in the Antarctic were adapted to cool
condition and some species collected in subalpine zones were adapted to warm condition. For exmaple, the optimal temperatures
for net photosynthetic rates in Antarctic lichens were lower than 5 C and those in lichens which lived on rock surface at

The response patterns of net photosynthesis to moisture level of mosses in xeric habitats were compared with those in mesic
habitats, in order to determine whether the former species are better adapted to the xeric condition with regard to carbon
gain.
Moss species examined wereRhacomitrium lanuginosum andR. barbuloides in xeric open habitats andDicranum japonicum, Hypnum plicatulum, Ptilium crista-castrensis, Pleurozium schreberi andHylocomium splendens in mesic habitats on the coniferous forest floor in the upper subalpine zone of Mt. Fuji. Three additional xerophytic species
collected at other localities,Ptychomitrium polyphylloides, Grimmia pilifera andHedwigia ciliata, were also examined.
Five species in the xeric habitats showed an optimum range of moisture level for net photosynthesis, 2 to 3g·g−1. On the other hand, species in the forest showed a wider optimum range, 3 to 8g·g−1. Net photosynthetic rate at the moisture level of 0.5g·g−1 was positive in xerophytic mosses, but negative in most forest mosses.
Moisture levels where external capillary water disappeared and drop of water potential began was determined by blotting water-saturated
shoots with membrane filters. These moisture levels were low in the xerophytic mosses and high in the forest mosses, although
there were some exceptions.
It was concluded that mosses in xeric habitats are better adapted for the efficient use of water for photosynthesis than those

Frameshift mutation hisD497 occurs in the operator-proximal portion of the Salmonella typhimurium gene coding for the dimeric protein, L-histidinol dehydrogenase (HDH). Rare revertants of hisD497 are deletions fusing the hisD gene to the adjacent preceding structural gene, hisG (adenosine 5'-triphosphate-PR transferase). HDH purified from one revertant, hisGD4908, contains subunits of approximately normal molecular weight but with no clearly demonstrable unique amino-terminal sequence. We propose that a combined inactive G-D polypeptide is synthesized and then cleaved at a number of closely juxtaposed sites by endoproteolytic activity. Bestatin, one of the biological response modifiers (BRMs), is an inhibitor of aminopeptidase B (AP-B), leucine aminopeptidase (LAP) and aminopeptidase M (AP-M). In this report, we investigated the direct effect of bestatin on the growth of cancer cells in vitro using established four choriocarcinoma cell lines. In vitro chemosensitivity was evaluated by the succinate dehydrogenase inhibition (SDI) test. Bestatin showed the growth-inhibitory effect on all the choriocarcinoma cell lines dose-dependently, especially on NaUCC-4 cells. Both an isomer of bestatin with no inhibitory activity against aminopeptidases, (2R, 3S)-AHPA-(R)-Leu, and another isomer with stronger inhibitory activity against AP-B than bestatin, (2S, 3S)-AHPA-(R)-Leu, did not show growth inhibition on NaUCC-4 cells. So it is suggested that one of the possible mechanisms responsible for the direct action of bestatin on the choriocarcinoma cells may be related to the inhibition of activity of LAP or AP-M rather than that of AP-B. Furthermore, cytotoxicity of actinomycin D on the choriocarcinoma cells was significantly enhanced by combination with bestatin. These results suggest that bestatin has not only an indirect host-mediated anti-tumor activity, but also a direct growth-inhibitory effect on some kinds of cancer cell lines. 相似文献

Net photosynthesis and dark respiration (CO2 flux) of Antarctic mosses were measured at Langhovde, East Antarctica, from 9 to 17 January 1988. Moss blocks were taken
from communities in the Yukidori Valley (69°14′30″S, 39°46′00″E) at Langhovde. Each block was composed ofCeratodon purpureus andBryum pseudotriquetrum, orB. pseudotriquetrum. The upper part of the block was used to measure net photosynthesis and dark respiration. The net photosynthesis of each
sample was measured in the field for one or three days with two infrared CO2 gas analyzers and an assimilation chamber. The relationships of net photosynthetic rate and dark respiration rate, to the
water content of the sample, the intensity of solar radiation and the moss temperature were estimated from the field data.
The maximum rate of net photosynthesis was about 4 μmol CO2 m−2s−1 at saturating radiation intensity and at optimum temperature, about 10°C. Environmental features of moss habitats in the
Yukidori Valley are discussed in relation to these results. 相似文献
