A polyclonal antibody to elastin-derived hexapeptide repeat, H-(Val-Gly-Val-Ala-Pro-Gly)(3)-NH(2), was prepared in order to investigate the differences between elastin fibres in intimal hyperplasia and media in human atheroscleroic lesions. The hexapeptide repeat and alpha-elastin were recognized by this polyclonal antibody in enzyme-linked immunosorbent assay (ELISA), but other elastin-derived peptides such as tetrapeptide repeat, pentapeptide repeat and nonapeptide were not. In the series of hexapeptide repeats, H-(VGVAPG)(n)-NH(2) where n is 1-7, the polyclonal antibody reacted strongly with oligomers (n = 3-7) and weakly with dimer (n = 2), but not with monomer (n = 1). CD measurements suggested that the beta-turn structure is important for recognition by the polyclonal antibody. In an immunohistochemical study, elastin was stained more strongly in intimal hyperplasia than in media, suggesting that newly synthesized elastin in intimal hyperplasia is morphologically distinct from that in media.  相似文献

The antitumor effect of indomethacin on Colon 26 tumor was investigated in conventional (CDF1) and nude mice (BALB/c nu/nu), and the telomerase activity in the tumor tissues treated with indomethacin was monitored. Growth of Colon 26 tumor was significantly suppressed with indomethacin treatment compared to the controls both in conventional and nude mice. And telomerase activity in the tumor tissues noticeably declined in contrast to normal somatic tissues (testis, liver and colon), which were not affected by indomethacin treatment. We also showed that indomethacin can suppress tumor growth in association with a preferential decrease in telomerase activity in tumor tissues both in conventional and nude mice to the same extent. This study suggests a method for investigating the mechanism of tumor suppression by indomethacin, and suggests that indomethacin might be useful as a novel agent for human cancer therapy.  相似文献

The fine structure of the cilium was examined by freeze-fracture-etch studies. In the interior of the transitional region, three types of plate structures were clearly observed. While the terminal plate contained fine fibrillar linkers suspending the central core plates from its peripheral doublet microtubules, two other types of plates had no suspending linkers. At the upper level of transitional region, one of the central microtubules elongated deeper than the other in the space surrounded by ring structure. Axosome-like structure was not observed in our replicas. Central vesicle of the basal body was also suspended by fine fibrillar linkers from peripheral triplets. Though membrane particles of ciliary necklace were recognized on protoplasmic and external fracture faces, and the external surface, particle arrays were not observed on protoplasmic surface. Instead, Y-shaped, cross bridges, one end of which attached to the doublet microtubules, merged in the circular ridge structure at opposite ends. This circular ridge structure at the necklace region may play a role as an anchoring site of both membrane particles of the necklace and cross bridges from peripheral doublet microtubules.  相似文献

The formation and rejoining rates of X-ray-induced DNA single-strand breaks (SSBs) were examined in radiosensitive and non-radiosensitive fibroblast lines from patients with tuberous sclerosis (TS), and fibroblasts from normal individuals, using the sensitive and quantitative alkaline elution method. No difference was found between these cell lines in the frequency of DNA SSBs directly produced by X-irradiation at any dose up to 750 rad. Kinetic analysis of the rate of rejoining of DNA SSBs after X-irradiation at 500 rad indicated that the rate of rejoining involved at least two components, an initial fast component and a slower component. TS fibroblast lines, either radiosensitive or nonsensitive, were proficient as to DNA SSB repair, but they showed an increased rate of rejoining in the initial fast repair process, when compared to normal fibroblast lines. Although the molecular basis for the accelerated rejoining of DNA SSBs remains unknown, it is possible that the abnormality may be related to a basic defect in TS.  相似文献

Summary Perinatal changes in the appearance of intramembranous particles (IMPs) of microvilli of enterocytes were analyzed quantitatively. In both the jejunum and the ileum, the IMP density on the P-face showed no significant changes from day 17 of gestation to day 5 of postnatal life. It increased between day 5 and day 12, reached a maximum at day 21, and thereafter decreased slightly. The IMP density on the E-face remained almost constant during the perinatal period in both intestinal parts. Measurements of particle diameters proved that neither the P-face nor the E-face membrane showed significant differences in either mean value or size distribution among different age groups.This study has revealed that the perinatal change in the IMP density on the P-face of microvilli correlates well with changes in the activity of certain enzymes found in the membranes of microvilli, e.g. disaccharidase and aminopeptidase.This study was supported by a Grant-in-Aid for Scientific Research to T. Synthesis of prostaglandins, TXA2 and PGI2, during one lung anesthesia   总被引:1,自引:0,他引:1  
This study is to determine histamine, serotonin, TXA2 and PGI2 to be the cause of Hypoxic Pulmonary Vasoconstriction (HPV) at the same time of one lung ventilation and thoracotomy. Five patients who were to undergo upper-lobe resection of the right lung, were included in this study. All patients underwent same premedication and anesthetic method. Endotracheal intubation was done with a Univent tracheal tube. Gas analysis and determinations of the substances were done at six times in total. Respiratory Index (RI) began to increase immediately after the start of one lung ventilation. Post-thoracotomy RI further increased. After closing of the thorax, RI returned to the control values. Serotonin and histamine showed no change in any case throughout the experiment. TXB2 began to increase along with the start of one lung ventilation. The 15-min value was 167.2 +/- 85.8 pg/ml and 30-min value was 345.6 +/- 261.2 pg/ml, showing significant increase. The values of 6-keto PGF1 alpha were 22.6 +/- 2.9 pg/ml (15-min value), 89.6 +/- 52.3 pg/ml (30-min value), 290.8 +/- 120.1 pg/ml (post opening value) and 84.4 +/- 21.3 pg/ml (post-closing value). In our study, we concluded that neither serotonin nor histamine was the direct factor of HPV. TXA2 was the direct chemical mediator of HPV and PGI2 showed a negative feedback to the pulmonary vasoconstriction.  相似文献

This study examined the effects of exogenous somatostatin and insulin on the release of islet amyloid polypeptide (IAPP), or amylin, from the isolated perfused rat pancreas. Somatostatin inhibited the release of both amylin and insulin from the perfused pancreas to the same extent. The infusion of 10 nM somatostatin resulted in 40% inhibition of the secretion of both amylin and insulin induced by 11.1 mM glucose and 10 mM arginine, and this inhibition was significantly increased to 70% by the infusion of 100 nM somatostatin (p less than 0.05). The amylin/insulin molar ratios remained constant at 0.8% and were not changed by the infusion of somatostatin. On the other hand exogenous insulin at a concentration of 1.8 nM did not affect the release of amylin induced by 11.1 mM glucose and 10 mM arginine, whereas 180 nM insulin slightly, although not significantly, inhibited the release of amylin by 15%. These findings suggest that the release of amylin may be negatively regulated by somatostatin and that circulating insulin may have no direct effect on the release of amylin at least at a physiological concentration.  相似文献

1. A novel approach using a Gaussian white noise as stimulus is described which allowed quantitative analysis of neuronal responses in the cercal system of the cockroach, Periplaneta americana. Cerci were stimulated by air displacement which was modulated by a sinusoidal and a white noise signal. During the stimulation, intracellular recordings were made from a uniquely identifiable, nonspiking, local interneuron which locates within the terminal abdominal ganglion. The white noise stimulation was cross-correlated with the evoked response to compute first- and second-order kernels that could define the cell's response dynamics. 2. The interneuron, cell 101, has an exceptionally large transverse neurite that connects two asymmetrical dendritic arborizations located on both sides of the ganglion. 3. The first-order Wiener kernels in cell 101 were biphasic (differentiating). The waveforms of the kernels produced by the ipsilateral and contralateral stimulations were roughly mirror images of each other: the kernels produced by wind stimuli on the side ipsilateral to the cell body of the interneuron are initially depolarized and then hyperpolarized, whereas those on the other side are initially hyperpolarized. The polarity reversal occurred along the midline of the animal's body, and no well-defined kernel was produced by a stimulus directed head on or from the tail. 4. Mean square error (MSE) between the actual response and the model prediction suggests that the linear component in cell 101 comprises half of the cell's total response (MSEs for the linear models were about 50% at preferred directions), whereas the second-order, non-linear component is insignificant. The linear component of the wind-evoked response was bandpass with the preferred frequency of 70-90 Hz. 5. Accounting for a noise, we reasonably assumed that at high frequencies the graded response in cell 101 is linearly related to a modulation of the air displacement and sensitive to the rate of change of the signal (i.e., wind velocity) and the direction of its source. It is suggested that the dynamics of the first-order kernel simply reflect the dynamics of sensory receptors that respond linearly to wind stimulation.  相似文献

Summary The response dynamics of cercal afferents in the cockroach, Periplaneta americana, were determined by means of a cross-correlation technique using a Gaussian white noise modulation of wind as a stimulus. The white noise stimulus could evoke sustained firing activity in most of the afferents examined (Fig. 1). The spike discharges were unitized and then cross-correlated with the stimulus to compute 1st- and 2nd-order Weiner kernels. The Ist-order kernels from a total of 28 afferents were biphasic and closely matched the time differential of a pulse (Figs. 1, 3 and 4). The amplitude and waveform of the kernels depended on the stimulus angle in such a way that the kernels were the mirror image of those on the polar opposite side (Figs. 2 and 3). The 2nd-order kernels were also differential. They had 2 diagonal peaks and 2 off-diagonal valleys in a 2-dimensional plot with 2 time axes (Figs. 1, 5 and 6). This 4-eye configuration was basically invariant irrespective of the stimulus angle, although the kernels varied in amplitude when the stimulus angle was changed. The time between the peak and a following trough of the 1st-order kernel was constant and had a mean of 4.6±0.1 ms, whereas the time between 2 diagonal peaks of the 2nd-order kernels was 4.7±0.1 ms (Figs. 4 and 6), suggesting that wind receptors (filiform sensilla) on cerci act as a band-pass filter with a peak frequency of about 106 Hz. The peak time, however, varies from 2.3 to 6.9 ms in both kernels, which may reflect the spatial distribution of the corresponding hairs on the cercus. The summation of the 1st- (linear) and 2nd-order (nonlinear) models precisely predicted the timing of the spike firing (Fig. 8). Thus, these 2 lower-order kernels can totally characterize the response dynamics of the wind receptors. The nonlinear response explains the directional sensitivity of the sensory neurons, while the differentiating 1st-order kernel explains the velocity sensitivity of the neurons. The nonlinearity is a signal compression in which one of the diagonal peaks of the 2nd-order kernel always offsets the downward phase of the 1st-order kernel (Fig. 7) and obviously represents a half-wave rectification property of the wind receptors that are excited by hair movement in only one direction and inhibited by hair movement in the polar opposite direction.  相似文献

We have constructed a chromosome-specific cosmid library from electrophoretically separated chromosomes of the basidiomyceteCoprinus cinereus and performed contig mapping and analysis of chromosome length polymorphisms (CLPs) for the smallest chromosome of the 5302 strain. A contig map of about 300 kb indicated that the novel size chromosomes in the F1 progeny were apparently recombinants containing physical markers derived from both ends and central regions in this map. This may be the first case in which the formation of CLPs in the F1 generation has been explained using the contig map. The results obtained were consistent with the hypothesis that novel CLPs were produced by meiotic recombination between the parental homologous chromosomes of unequal sizes.  相似文献
