In this study, alpha-bisabolol, a sesquiterpene alcohol present in natural essential oil, was found to have a strong time- and dose-dependent cytotoxic effect on human and rat glioma cells. After 24 h of treatment with 2.5-3.5 microM alpha-bisabolol, the viability of these cells was reduced by 50% with respect to untreated cells. Furthermore, the viability of normal rat glial cells was not affected by treatment with alpha-bisabolol at the same concentrations as above. Glioma cells treated with high concentration of alpha-bisabolol (10 microM) resulted in a 100% cell death. Judging from hypo-G1 accumulation, poly(ADP-ribose) polymerase cleavage, and DNA ladder formation, the cytotoxicity triggered by alpha-bisabolol resulted from apoptosis induction. Moreover, the dissipation of mitochondrial-inner transmembrane potential and the release of cytochrome c from mitochondria indicated that, in these glioma cells, apoptosis occurred through an intrinsic pathway. As pointed out by the experimental results, alpha-bisabolol may be considered a novel compound able to inhibit glioma cell growth and survival.  相似文献

Roles for c-Myc in self-renewal of hematopoietic stem cells   总被引:12,自引:0,他引:12  
Notch and HOXB4 have been reported to expand hematopoietic stem cells (HSCs) in vitro. However, their critical effector molecules remain undetermined. We found that the expression of c-myc, cyclin D2, cyclin D3, cyclin E, and E2F1 was induced or enhanced during Notch1- or HOXB4-induced self-renewal of murine HSCs. Since c-Myc can act as a primary regulator of G(1)/S transition, we examined whether c-Myc alone can induce self-renewal of HSCs. In culture with stem cell factor, FLT3 ligand, and IL-6, a 4-hydroxytamoxifen-inducible form of c-Myc (Myc/ERT) enabled murine Lin(-)Sca-1(+) HSCs to proliferate with the surface phenotype compatible with HSCs for more than 28 days. c-Myc activated by 4-hydroxytamoxifen augmented telomerase activities and increased the number of CFU-Mix about 2-fold in colony assays. Also, in reconstitution assays, HSCs expanded by c-Myc could reconstitute hematopoiesis for more than 6 months. As for the mechanism of c-myc induction by Notch1, we found that activated forms of Notch1 (NotchIC) and its downstream effector recombination signal-binding protein-J kappa (RBP-VP16) can activate the c-myc promoter through the element between -195 bp and -161 bp by inducing the DNA-binding complex. Together, these results suggest that c-Myc can support self-renewal of HSCs as a downstream mediator of Notch and HOXB4.  相似文献

We studied changes in the carbohydrate expression following apoptotic cell death induced by treatment with interferon (IFN)- and anti-Fas antibody using human colon adenocarcinoma HT-29 cells. An apoptotic cell death of HT-29 accompanied with typical DNA fragmentation was observed when the cells were cultured sequentially with IFN- and anti-Fas antibody. In flow cytometric analyses, the expression of Lex and Ley antigen was strongly and slightly enhanced, respectively, on the cell surface in accordance with the apoptosis. When the fucosyltransferase (Fuc-T) activities of the lysates from the treated cells were examined relative to those from untreated cells, a 2.5-fold increase of (1,3)-Fuc-T activities and a slight increase of (1,2)-Fuc-T activities were observed, but little or no increase of (1,4)-Fuc-T activity was detected. In Northern blot analyses using probes for Fuc-T III, IV, V, VI and VII genes, strong RNA messages for Fuc-T III, V and/or VI and a weak RNA message for Fuc-T IV were detected in the untreated HT-29 cells. On the other hand, in the treated cells, the messages for Fuc-T III, V and/or VI were found to almost disappear and the 2.3 kb message for Fuc-T IV was observed to elevate 2.8-fold. From a monthly survey over a ten-year period (1983–1992) of the Ishitegawa Reservoir, Japan, two multiple regression equations describing the mean chlorophylla (Chl-a) concentration at 0.5 m depth during July, August and September (C S ) and the maximum Chl-a concentration in the photic zone, including its subsurface maximum during this period (C M ), were obtained. The conductivity at 0.5 m depth in May and the mean air temperature during May or during May and June were used as independent variables. These equations were calculated using seven years of data (1983–1988 and 1992). From 1989 to 1991 two impermeable plastic-coated sheet fences were installed across the upper end of the reservoir along the top 5 m of water column. The equations were used to determine the effectiveness of this flow regulation on the summer Chl-a concentration. In 1989 and 1991, when the fences were in place from June to August, no effects were found on the observed C S and C M values. In 1990, when the fences had been in place since October 1989, the observed values were lower than the predicted values.  相似文献

Hypericum perforatum is a medicinal plant species containing many polyphenolic compounds, namely flavonoids and phenolic acids. Since polyphenolic compounds have high antioxidant potential, we have investigated the effects of H. perforatum extract on the development of multiple organ dysfunction syndrome caused by zymosan (500 mg/kg, administered i.p. as a suspension in saline) in mice. Organ failure and systemic inflammation in rats was assessed 18 h after administration of zymosan and/or H. perforatum extract and monitored for 12 days (for loss of body weight and mortality). Treatment of mice with H. perforatum extract (30 mg/kg i.p., 1 and 6h after zymosan) attenuated the peritoneal exudation and the migration of polymorphonuclear cells caused by zymosan, pulmonary, intestinal and pancreatic injury, and renal dysfunction as well as the increase in myeloperoxidase in the lung and intestine. Immunohistochemical analysis for inducible nitric oxide synthase (iNOS), nitrotyrosine, and poly(ADP-ribose) (PAR) revealed positive staining in lung and intestine tissues obtained from zymosan-injected mice. The degree of staining for nitrotyrosine, iNOS, and PAR was markedly reduced in tissue sections obtained from zymosan-treated mice, which received H. perforatum extract. In conclusion, this study provides evidence, for the first time, that H. perforatum extract attenuates the degree of zymosan-induced multiple organ dysfunction syndrome in mice.  相似文献

Dental caries affects people of all ages and is a worldwide health concern. Streptococcus mutans is a major cariogenic bacterium because of its ability to form biofilm and induce an acidic environment. In this study, the antibacterial activities of magnolol and honokiol, the main constituents of the bark of magnolia plants, toward planktonic cell and biofilm of S. mutans were examined and compared with those of chlorhexidine. The minimal inhibitory concentrations of magnolol, honokiol and chlorhexidine for S. mutans were 10, 10 and 0.25 µg/mL, respectively. In addition, each agent showed bactericidal activity against S. mutans planktonic cells and inhibited biofilm formation in a dose‐ and time‐dependent manner. Magnolol (50 µg/mL) had greater bactericidal activity against S. mutans biofilm than honokiol (50 µg/mL) and chlorhexidine (500 µg/mL) at 5 min after exposure, while all showed scant activity against biofilm at 30 s. Furthermore; chlorhexidine (0.5–500 µg/mL) exhibited high cellular toxicity for the gingival epithelial cell line Ca9‐22 at 1 hr, whereas magnolol (50 µg/mL) and honokiol (50 µg/mL) did not. Thus; it was found that magnolol has antimicrobial activities against planktonic and biofilm cells of S. mutans. Magnolol may be a candidate for prevention and management of dental caries.  相似文献

Lilium auratum var. platyphyllum, which is endemic to the Izu Islands, has white petals and emits a strong floral scent, typical of moth‐pollinated plants. Moths might be important pollinators within the Izu Islands because swallowtail butterflies are absent or rare there. Therefore, we investigated insular var. platyphyllum and widespread L. auratum var. auratum on the mainland of Japan to clarify the relationship between floral characteristics and pollinators. Measurement of floral scent intensity using an odor sensor indicated that the intensity increased in the evening and at night in var. platyphyllum, whereas intensity increased at night in var. auratum. Total sugar weight in nectar, which was calculated by nectar volume and sugar concentration, showed that the flowers of var. platyphyllum secreted nectar abundantly both in the evening and at night, whereas those of var. auratum secreted an almost constant amount of nectar throughout the day. Flower visitor assemblages and their frequencies studied using digital cameras suggested that crepuscular and nocturnal hawkmoths are important pollinators for var. platyphyllum. In contrast, both diurnal swallowtail butterflies and nocturnal large hawkmoths are important and effective pollinators for var. auratum. These conclusions were also supported by the exclusion experiments of either diurnal or nocturnal flower visitors using mesh bags. The rates of seed sets of var. platyphyllum were significantly higher from nocturnal pollination than from diurnal pollination. Thus, this study revealed that floral traits of var. platyphyllum show more adaptation for crepuscular and nocturnal hawkmoths, which are relatively abundant in the Izu Islands, than those of var. auratum.  相似文献

Summary Escherichia coli mutants deficient in hydrogenase activity (Hyd-) were derived from E. coli C600 by mutagenesis with nitrosoguanidine. Hydrogenase activities of mutant strains; HK-2, HK-7, HK-8, HK-16, HK-23, and HK-26 were below 1/100 that of the parental strain E. coli C600. Conjugational transfer of plasmid F-143 to the mutants was carried out and hydrogenase activities of the transformants were assayed. Recovery of hydrogenase activities in mutant strains; HK-2, HK-7, HK-8, HK-16, and HK-23 was observed, but not for HK-26. Two kinds of hydrogenase genes of Citrobacter freundii were cloned on pBR 322 and hybrid plasmids pCBH2 and pCFH1 were obtained. Hydrogenase activities of mutant strains HK-2, HK-8 and HK-16 were complemented with pCBH2 and strain HK-7 with pCFH1 respectively. The other mutant strains, HK-23, HK-26, however, were not complemented with these plasmids.  相似文献
